This experiment was conducted to investigate the effect of starter cultures on the physicochemical properties, texture, and consumer preferences of soft white cheese (SWC) made from camel (Camelus dromedarius) milk. The experiment was laid out in a completely randomized design with 5 treatments [starter cultures; i.e., 1 thermophilic (STI-12), 2 blended (RST-743 and XPL-2), and 2 mesophilic (R-707 and CHN-22) cultures]. Starter cultures STI-12 and RST-743 were inoculated at 37°C, whereas XPL-2, R-707, and CHN-22 were inoculated at 30°C. Camel milk inoculated using STI-12 and RST-743 cultures resulted in faster acidification than XPL-2, R-707, and CHN-22 cultures. Camel milk SWC made using STI-12 and CHN-22 cultures gave lower pH (4.54) and titratable acidity (0.59), respectively, whereas R-707 culture resulted in high cheese yield (13.44 g/100 g). In addition, high fat (20.91 g/100 g), protein (17.49 g/100 g), total solids (43.44 g/100 g), and ash (2.40 g/100 g) contents were recorded for SWC made from camel milk made using RST-743 culture. Instrumental analysis of cheese texture revealed differences in resistance to deformation in which camel milk SWC made using RST-743 culture gave higher firmness (3.20 N) and brittleness (3.12 N). However, no significant difference was observed among camel milk SWC adhesiveness made using different starter cultures. Consumer preference for appearance, aroma, taste, and overall acceptances of SWC were affected by inoculation of starter cultures. Considering curd firmness, cheese yield, compositional quality, and textures using STI-12, RST-743, and R-707, these cultures were found to be better for the manufacture of camel milk SWC. 相似文献

Rotational 3-breed crossbred cows of Montbéliarde, Viking Red, and Holstein (CB) were compared with Holstein (HO) cows for alternative measures of feed efficiency as well as income over feed cost (IOFC) and residual feed intake (RFI) during the first 150 d of first, second, and third lactations. Primiparous and multiparous CB (n = 63 and n = 43, respectively) and HO (n = 60 and n = 37, respectively) cows were fed the same total mixed ration twice daily with refusals weighed once daily. Feed was analyzed for dry matter content, net energy for lactation, and crude protein content. Body weight (BW) was recorded twice weekly. Daily production of milk, fat, and protein were estimated from monthly test days with best prediction. Measures of efficiency from 4 to 150 d in milk (DIM) were feed conversion efficiency (FCE), defined as fat plus protein production (kg) per kilogram of dry matter intake (DMI); ECM/DMI, defined as kilograms of energy-corrected milk (ECM) per kilogram of DMI; net energy for lactation efficiency (NELE), defined as ECM (kg) per megacalorie of net energy for lactation intake; crude protein efficiency (CPE), defined as true protein production (kg) per kilogram of crude protein intake; and DMI/BW, defined as DMI (kg) per kilogram of BW. The IOFC was defined as revenue from fat plus protein production minus feed cost. The RFI from 4 to 150 DIM for each lactation was the residual error remaining from regression of DMI on milk energy output (Mcal), metabolic BW, and energy required for change in BW (Mcal). Statistical analysis of measures of feed efficiency and RFI for primiparous cows included the fixed effects of year of calving and breed group. For multiparous cows, statistical analysis included breed as a fixed effect and cow as a repeated effect nested within breed group. Primiparous CB cows had higher means for FCE (+5.5%), ECM/DMI (+4.0%), NELE (+4.0%), and CPE (+5.2%) and a lower mean DMI/BW (–5.3%) than primiparous HO cows. Primiparous CB cows ($875) also had higher mean IOFC than primiparous HO cows ($825). In addition, mean RFI from 4 to 150 DIM was significantly lower (more desirable) for primiparous CB cows than HO cows. Likewise, multiparous CB cows had higher means for FCE (+8.2%), ECM/DMI (+5.9%), NELE (+5.8%), and CPE (+8.1%) and a lower mean for DMI/BW (–4.8%) than multiparous HO cows. When advising farmers on how to control Johne's disease in an infected herd, one of the main recommendations is to avoid feeding waste milk to calves and instead feed calf milk replacer (CMR). This advice is based on the assumption that CMR is free of viable Mycobacterium avium ssp. paratuberculosis (MAP) cells, an assumption that has not previously been challenged. We tested commercial CMR products (n = 83) obtained from dairy farms around the United States by the peptide-mediated magnetic separation (PMS)-phage assay, PMS followed by liquid culture (PMS-culture), and direct IS900 quantitative PCR (qPCR). Conventional microbiological analyses for total mesophilic bacterial counts, coliforms, Salmonella, coagulase-negative staphylococci, streptococci, nonhemolytic Corynebacterium spp., and Bacillus spp. were also performed to assess the overall microbiological quality of the CMR. Twenty-six (31.3%) of the 83 CMR samples showed evidence of the presence of MAP. Seventeen (20.5%) tested positive for viable MAP by the PMS-phage assay, with plaque counts ranging from 6 to 1,212 pfu/50 mL of reconstituted CMR (average 248.5 pfu/50 mL). Twelve (14.5%) CMR samples tested positive for viable MAP by PMS-culture; isolates from all 12 of these samples were subsequently confirmed by whole-genome sequencing to be different cattle strains of MAP. Seven (8.4%) CMR samples tested positive for MAP DNA by IS900 qPCR. Four CMR samples tested positive by both PMS-based tests and 5 CMR samples tested positive by IS900 qPCR plus one or other of the PMS-based tests, but only one CMR sample tested positive by all 3 MAP detection tests applied. All conventional microbiology results were within current standards for whole milk powders. A significant association existed between higher total bacterial counts and presence of viable MAP indicated by either of the PMS-based assays. This represents the first published report of the isolation of viable MAP from CMR. The objective of the present study was to evaluate the influence of two diets containing either Brussels sprouts or inulin/rape seed cake, compared with a standard diet (control) for slaughter pigs on flavour and odour attributes and sensory profile of cooked pork. Three weeks prior to slaughter 24 female pigs were allocated to three diets: (1) a standard grower-finishing diet (control) for slaughter pigs containing barley, wheat and soy-bean meal, (2) the control diet containing 11 energy percent Brussels sprouts and (3) a diet containing 25% inulin and 55% rape seed cake. The odour and flavour of the cooked meat from inulin/rape seed cake-fed pigs differed significantly from the other two diets, showing reduced meat odour, increased pig and acrid odour, increased pig flavour, reduced fresh flavour and total impression. Dehalococcoides strains reductively dechlorinate a wide variety of halogenated compounds including chlorinated benzenes, biphenyls, naphthalenes, dioxins, and ethenes. Recent genome sequencing of the two Dehalococcoides strains CBDB1 and 195 revealed the presence of 32 and 18 reductive dehalogenase homologous genes, respectively, and therefore suggested an even higher dechlorinating potential than previously anticipated. Here, we demonstrate reductive dehalogenation of chlorophenol congeners by Dehalococcoides strains CBDB1 and 195. Strain CBDB1 completely converted 2,3-dichlorophenol, all six trichlorophenols, all three tetrachlorophenols, and pentachlorophenol to lower chlorinated phenols. Observed dechlorination rates in batch cultures with cell numbers of 10(7) mL(-1) amounted up to 35 microM day(-1). Chlorophenols were preferentially dechlorinated in the ortho position, but also doubly flanked and singly flanked meta- or para-chlorine substituents were removed. We used a newly designed computer-assisted direct cell counting protocol and quantitative PCR to demonstrate that strain CBDB1 uses chlorophenols as electron acceptors for respiratory growth. The growth yield of strain CBDB1 with 2,3-dichlorophenol was 7.6 x 10(13) cells per mol of Cl- released, and the growth rate was 0.41 day(-1). For strain 195, fast ortho dechlorination of 2,3-dichlorophenol, 2,3,4-trichlorophenol, and 2,3,6-trichlorophenol was detected, with only the ortho chlorine removed. Because chlorinated phenolic compounds are widely distributed as natural components in anaerobic environments, our results reveal one mode in which the Dehalococcoides species could have survived through earth history. 相似文献

A new integrated methodology for forensic oil spill identification is presented. It consists of GC-MS analysis, chromatographic data processing, variable-outlier detection, multivariate data analysis, estimation of uncertainties, and statistical evaluation. The methodology was tested on four groups of diagnostic ratios composed of petroleum biomarkers and ratios within homologous PAH categories. Principal component analysis (PCA) was employed and enabled the simultaneous analysis of many diagnostic ratios. Weathering was taken into account by considering the sampling uncertainties estimated from replicate spill samples. Statistical evaluation ensured an objective matching of oil spill samples with suspected source oils as well as classification into positive match, probable match, and nonmatch. The data analysis is further refined if two or more source oils are classified as probable match by using weighted least squares fitting of the principal components, local PCA models, and additional information relevant to the spill case. The methodology correctly identified the source of two spill samples (i.e., crude oils from Oseberg East and Oseberg Field Centre) and distinguished them from closely related source oils. 相似文献

Differences in heat tolerance between preimplantation embryos from Brahman, Romosinuano, and Angus breeds 总被引:1,自引:0,他引:1
Exposure to 41 degrees C reduces development of embryos of heat-sensitive breeds (Holstein and Angus) more than for embryos of the heat-tolerant Brahman breed. Here it was tested whether embryonic resistance to heat shock occurs for a thermotolerant breed of different genetic origin than the Brahman. In particular, the thermal sensitivity of in vitro produced embryos of the Romosinuano, a Bos taurus, Criollo-derived breed, was compared to that for in vitro produced Brahman and Angus embryos. At d 4 after insemination, embryos > or = 8 cells were randomly assigned to control (38.5 degrees C) or heat shock (41 degrees C for 6 h) treatments. Heat shock reduced the proportion of embryos that developed to the blastocyst stage on d 8 after insemination. At 38.5 degrees C, there were no significant differences in development between breeds. Among embryos exposed to 41 degrees C, however, development was lower for Angus embryos than for Brahman and Romosinuano embryos. Furthermore, an Angus vs. (Brahman + Romosinuano) x temperature interaction occurred because heat shock reduced development more in Angus (30.3 +/- 4.6% at 38.5 degrees C vs. 4.9 +/- 4.6% at 41 degrees C) than in Brahman (25.1 +/- 4.6% vs. 13.6 +/- 4.6%) and Romosinuano (28.3 +/- 4.1% vs. 17.5 +/- 4.1%). Results demonstrate that embryos from Brahman and Romosinuano breeds are more resistant to elevated temperature than embryos from Angus. Thus, the process of adaptation of Brahman and Romosinuano breeds to hot environments resulted in both cases in selection of genes controlling thermotolerance at the cellular level. 相似文献

The primary aim of this study was to evaluate the phenotypic and genetic trends for stillbirth in Danish Holsteins. Trends of calving difficulty and calf size were also evaluated. The second aim was to compare predicted transmitting abilities (PTA) of sires for stillbirth using a linear and a threshold model. Direct and maternal genetic effects were modeled by fitting correlated additive genetic effects of the sire and the maternal grandsire (MGS). For both the calf and the dam, covariates of breed proportions of Holstein-Friesian (HF) and the heterozygosity between HF and the original Danish Black and White (ODBW) were included. Records from 1.8 million first-calving Danish Holstein cows calving from 1985 to 2002 were used. In this period, the overall frequency of stillbirth increased from 0.071 to 0.090. An unfavorable genetic trend of stillbirth was found for both the direct and maternal effect. The background for the genetic trends was an intense use of HF sires as sires of sons, which increased the proportion of HF genes to 94% in the Danish Holstein calves born in 2002. The effect of the imported HF genes was higher direct effects of calf size, calving difficulty, and stillbirth compared with the ODBW genes. The maternal effect of stillbirth was poorer for HF than for ODBW even though HF had a better maternal calving performance than ODBW. The threshold and the linear models showed almost similar predictions of transmitting abilities of sires. 相似文献

The effects of various thermal processing treatments (blanching, boiling and steaming) of red cabbage, Brassica oleracea L. ssp. capitata f. rubra cv. ‘Autoro’, were assessed for the levels of glucosinolates (GLS), total phenols (TP), total monomeric anthocyanins (TMA), l-ascorbic acid (L-AA) and soluble sugars, as well as for the antioxidant potential by the ferric reducing ability power (FRAP) and oxygen radical absorbance capacity (ORAC) assays. Individual native GLS were determined by ion-pair HPLC-MS/DAD. There were significant (p < 0.05) losses in blanched red cabbage: TP, 43%, TMA 59%, FRAP 42%, ORAC 51%, L-AA 48% and soluble sugars 45%. Boiling gave less extensive reductions: TP 16%, TMA 41%, FRAP 17%, ORAC 19%, L-AA 24% and soluble sugars 19%. Steaming caused no losses for TP, ORAC, FRAP or soluble sugars. However, significant reductions were found for TMA and L-AA, with 29% and 11%, respectively. In general losses were accounted for in the processing waters; however, TMA was not fully recovered, indicating degradation. Total GLS were severely affected by processing, with reductions of 64%, 38% and 19% in blanched, boiled and steamed red cabbage, respectively. Total aliphatic and indole GLS were similarly affected. Aroma compounds emitted from elderberry juices processed from seven cultivars were collected by the dynamic headspace technique and analysed by GC–FID and GC–MS. Forty aroma compounds were identified and quantified, including nine compounds which had not previously been detected in elderberry juice. Significant differences were found among cultivars in the concentration levels of 30 compounds. The sensory characteristics of the individual aroma compounds in elderberry juice were determined by a GC‐sniffing technique, and the compounds were grouped according to their odour. The characteristic elderberry odour is due to dihydroedulan and β‐damascenone, of which the former occurs in relatively high concentrations in the headspace of elderberry juice. The fruity group consisted of aliphatic alcohols and aldehydes and aromatic esters, of which 1‐pentanal, 2‐methyl‐1‐propanol, 2‐ and 3‐methyl‐1‐butanol, 1‐octanal, 1‐octanol and methyl and ethyl benzoate contributed with fruity notes. In the flowery group, 1‐nonanal, nerol oxide and (Z)‐ and (E)‐rose oxide contributed with characteristic elder flower odour, whereas other flowery notes were associated with hotrienol, linalool and α‐terpineol. Fresh and grassy odours were correlated with 1‐hexanal, (E)‐2‐hexen‐1‐al, (Z)‐3‐hexen‐1‐ol, (E)‐2‐hexen‐1‐ol and (E)‐2‐octen‐1‐al of the grassy group, whereas 1‐octen‐3‐ol and 1‐octen‐3‐one of the agrestic group contributed significantly with the characteristic aroma of mushrooms. © 2000 Society of Chemical Industry 相似文献
