The gene responsible for the second form of autosomal dominant polycystic kidney disease, PKD2, has recently been identified. We now describe the cloning, genomic localization, cDNA sequence, and expression analysis of its murine homologue, Pkd2. The cloned cDNA sequence is 5134 bp long and is predicted to encode a 966-amino-acid integral membrane protein with six membrane-spanning domains and intracellular NH2 and COOH termini. Pkd2 is highly conserved with 91% identity and 98% similarity to polycystin-2 at the amino acid level. Pkd2 mRNA is widely expressed in mouse tissues. Pkd2 maps to mouse Chromosome 5 and is excluded as a candidate gene for previously mapped mouse mutations resulting in a polycystic kidney phenotype.  相似文献

Treatments of Chinese hamster V79 cells during one cell cycle with a new type of topoisomerase II inhibitor, ICRF-193, which does not accumulate cleavable topoisomerase-DNA complexes induced both chromosome- and chromatid-type aberrations with high frequencies. Furthermore, ICRF-193 synergistically enhanced the yield of UVB-induced chromatid-type aberrations, chromatid exchanges in particular. Treated with ICRF-193 for the last 3 h before harvest, cells showed frequent incidence of chromatid-type aberrations and synergistic enhancement of UVB-induced chromatid-type aberrations, chromatid exchanges in particular. These results suggest that spontaneous and UVB-induced lesions might be ultimately transformed into chromatid-type aberrations by topoisomerase II-dependent checkpoint process(es) in the G2 phase of the cell cycle.  相似文献

The objective of this study was to determine the effects of fat supplementation from canola seed (CS) on ruminal fermentation and postruminal digestion of OM, carbohydrates, and energy of diets containing different levels of forage. Six ruminally and duodenally cannulated beef steers (354 kg +/- 18) were given ad libitum access to six isonitrogenous diets that were offered twice daily in a 6 x 6 Latin square design. Treatments were arranged as a 2 x 3 factorial with two forage levels (70 vs 30% of dietary DM as corn silage) and three forms of CS supplementation including no CS or CS added at 10% of dietary DM as whole CS treated with alkaline hydrogen peroxide or untreated crushed CS. Fat from CS provided 5% of dietary DM. The remaining dietary ingredients were corn, canola meal, molasses, and urea. No interactions (P > .05) between dietary forage level and CS supplementation were observed for ruminal characteristics or digestion of OM, carbohydrates, and energy in the rumen, postruminally, or in the total tract. Fat supplementation from CS did not affect (P > .05) DMI. With few exceptions, fat supplementation did not affect (P > .05) ruminal, postruminal, or total tract digestibilities of OM, structural and nonstructural carbohydrates, and GE. Ruminal disappearance of GE was decreased (P < .05) when diets were supplemented with fat from whole treated CS, and total tract digestibilities of OM and GE were decreased (P < .05) when diets were supplemented with fat from CS in either form. Ruminal pH, concentrations of NH3 N and total VFA, and molar proportions of acetate, propionate, and butyrate were not affected (P > .05) by fat supplementation. Results suggest that fat supplementation from CS (at 5% of dietary DM) as whole treated or untreated crushed had no negative effects on ruminal fermentation of OM, carbohydrates, or energy when steers were given ad libitum access to diets containing high or low forage.  相似文献

The treatment of frying oils with adsorbents could practically extend the frying life of oils. Combined synthetic adsorbent treatment of used frying oils was studied the first time. The combinations of four commonly used filter aids: Britesorb (Br), Hubersorb 600 (HB), Frypowder (Fr), and Magnesol (Ma) were evaluated for frying oil recovery. AOCS official methods were used to evaluate their adsorptiveness, including free fatty acids (FFA), conjugated diene value, total polar components, oxidative stability index (OSI), and absorbance at 420 nm. The selected combinations HB+Ma+Fr and HB+Ma+Br exhibited consistent high recovery abilities on various used oil samples. A 3, 3, and 2% HB, Ma, and Fr, respectively, for the first combination (F), and 2, 3, and 2% HB, Ma, and Br, respectively, for the second combination (B) were the most effective. The optimal treatment duration was 6–9 min and 3–6 min for combinations F and B, respectively, which reduced FFA by 82.6–87.6%, absorbance by 26.8–32.6%, and Foodoil Sensor readings by 5.6–8.6%. Addition of antioxidant, such as 50 ppm butylated hydroxytoluene and 50 ppm propyl gallate, increased the OSI value by 48.9–80.8%. Such adsorbent combinations may be used in practical operation to extend frying life of frying oils and improve the healthy aspects of used frying oils.  相似文献

Aerosol particles expelled during human coughs are a potential pathway for infectious disease transmission. However, the importance of airborne transmission is unclear for many diseases. To better understand the role of cough aerosol particles in the spread of disease and the efficacy of different types of protective measures, we constructed a cough aerosol simulator that produces a human-like cough in a controlled environment. The simulated cough has a 4.2 l volume and is based on coughs recorded from influenza patients. In one configuration, the simulator produces a cough aerosol containing particles from 0.1 to 100 μm in diameter with a volume median diameter (VMD) of 8.5 μm and a geometric standard deviation (GSD) of 2.9. In a second configuration, the cough aerosol has a size range of 0.1–30 μm, a VMD of 3.4 μm, and a GSD of 2.3. The total aerosol volume expelled during each cough is 68 μl. By generating a controlled and reproducible artificial cough, the simulator allows us to test different ventilation, disinfection, and personal protection scenarios. The system can be used with live pathogens, including influenza virus, which allows isolation precautions used in the healthcare field to be tested without risk of exposure for workers or patients. The information gained from tests with the simulator will help to better understand the transmission of infectious diseases, develop improved techniques for infection control, and improve safety for healthcare workers and patients.

Theories of OPEC such as price leadership, cartel, or game theoretic models suggest an incentive for OPEC members to expand their production capacity well above current levels in order to maximize revenues. Yet individual OPEC members consistently explore for and develop oil fields at a level well below their potential. The cause of low oil exploration and development efforts among OPEC members, and even some non-OPEC members, may have to do with risk aversion. This paper describes an alternative theory for OPEC behavior based on risk aversion using a two piece non Neumann–Morgenstern utility function similar to Fishburn and Kochenberger (1979, Decision Science 10, 503–518), and Friedman and Savage (1948, Journal of political Economy 56). Results of the first fabrication and measurement of travelling-wave photodetectors are reported. The devices have bandwidths as high as 172 GHz, the highest reported for a p-i-n photodetector, and bandwidth-efficiency products as large as 76 GHz, the largest reported for any photodetector without gain. Comparisons with vertically illuminated and waveguide photodetectors fabricated on the same wafer establish the superior performance of travelling-wave photodetectors. Twenty-four patients who failed uvulopalatopharyngoplasty (UPPP) for obstructive sleep apnea (OSA) had an adjustable oral (Herbst) appliance made to treat the persistent apnea. Six patients discontinued the device prior to sleep evaluation. Eighteen patients had polysomnographic evaluations at baseline, post-UPPP, and with the Herbst appliance in place. The apnea-hypopnea index baseline (AHI) and arterial oxygen saturation (SaO2) nadir were 42.3+/-6.1 and 83.6+/-1.8%, respectively. There was no significant change in either parameter with surgery. With the oral appliance, the AHI fell to 15.3+/-4.4 (p < or = 0.01) and the SaO2 nadir increased to 87.9+/-1.2% (p < or = 0.05). Ten of the patients had control of the OSA with the Herbst appliance with a fall in the AHI to < 10. There were, in addition, two partial responders as defined by an AHI of <20 and a >50% fall in AHI compared with baseline and post-UPPP values. All but one of the responders and partial responders had complete resolution of subjective symptoms of daytime sleepiness with the appliance. An adjustable oral appliance appears to be an effective mode of therapy to control OSA after an unsuccessful UPPP.  相似文献

Strict regulations concerning the content of volatile organic compounds (VOCs) and heavy metals (Cr6+) in aircraft coating systems have increased the economic burden of the United States Air Force (USAF) in the area of coating maintenance. To this end, it is critical to have methods to characterize new coating systems in such a manner that the data can be used to predict accurately and reliably the expected lifetime of the coatings in service. Electrochemical noise method (ENM) and electrochemical impedance spectroscopy (EIS) are two techniques used to monitor extent and rate of corrosion. The USAF is currently employing these methods in order to supplement data acquired from traditional salt-spray methods. ENM and EIS are used to evaluate each component of the coating system and its contribution to corrosion prevention. Preliminary evaluations of an aircraft coating system on aluminum substrate (Al 2024-T3) produces resistance noise values of 106 to 107 Ω/cm2. It is hoped that these results will form the basis of coatings that give increased USAF fleet service life and reduction in maintenance manpower and materials costs.  相似文献

Flexible polymers such as poly dimethyl siloxane (PDMS) can be patterned at the micro‐ and nanoscale by casting, for a variety of applications. This replication‐based fabrication process is relatively cheap and fast, yet injection molding offers an even faster and cheaper alternative to PDMS casting, provided thermoplastic polymers with similar mechanical properties can be used. In this paper, a thermoplastic polyurethane is evaluated for its patterning ability with an aim to forming the type of flexible structures used to measure and modulate the contractile forces of cells in tissue engineering experiments. The successful replication of grating structures is demonstrated with feature sizes as low as 100 nm and an analysis of certain processing conditions that facilitate and enhance the accuracy of this replication is presented. The results are benchmarked against an optical storage media grade polycarbonate.
