Intravenous administration of 125I-hCG to 7-8 day pseudopregnant rats resulted in maximum uptake of radioactivity to corpora lutea 2 hours after treatment. At this time tissue/plasma radioactivity ratios on an equal weight basis were: corpora lutea, 70.2 +/- 12.8; ovarian interstitium, 4.6 +/- 0.2; kidney, 2.2 +/- 0.1. No appreciable uptake was seen by adrenals or liver. Radioactivity in corpora lutea was associated primarily with membranes which sedimented at 2000g and when released by heat it was more than 63% bound to luteal LH receptor preparation in vitro. Radioactivity in renal tissue was associated primarily with the 100,000g supernatant fraction and was bound less than 1% to luteal LH receptors in vitro. PGF2alpha significantly reduced uptake (p less than .001) of 125I-hCG by corpora lutea within 30 minutes (-63%) as well as at 1 (-64%), 2 (-75%), 4 (-68%) and 24 hours (-85%). No clear effect of PGF2alpha on uptake of 125I-hCG by ovarian interstitial tissue was seen. Plasma progesterone was significantly decreased (p less than .001) within 30 minutes (-47%; p less than .01) after PGF2alpha treatment and also at 1 (-65%), 2 (-82%), 4 (-68%) and 24 hours (-92%). Two hours after PGF2alpha treatment the content of progesterone in corpora lutea was depressed (-46%; p less than .001). It is suggested that the rapid inhibition of luteal progesterone production induced by PGF2alpha in vivo occurs through a block in gonadotropin uptake by corpora lutea. 相似文献

Among the complications in a series of 1,400 consecutive Charnley low friction arthroplasty procedures, there were 8 dislocations, and 3 highly unstable hips. Three dislocations followed severe trauma, in 2 of the 3 there was only fibrous union of the greater trochanter. In all but 2 of the hips, more than one previous operation had been performed. More than one technical fault was evident in most dislocations. Six of the 8 dislocations required reoperation. Meticulous attention to the Charnley procedure, including soft tissue elements of the hip joint is necessary to avoid instability and dislocation following total hip arthroplasty. 相似文献

The study concerns the effect of angiotensin II when infused into the systemic and portal blood flow upon the general and renal haemodynamics in normal dogs and in hypertensive animals, as well as the effect of the operation of porto-caval transposition upon the course of renovascular hypertension. When peptide is infused, at a rate that causes a moderate pressor effect, into the systemic blood flow of normal animals, an antidiuretic and antinatriuretic effect is obtained, in hypertensive animals--an increase of diuresis, natriuresis and a less distinct pressor effect are obtained. When angiotensin II is infused into the portal flow, a less distinct pressor and renal effect is seen in animals with renovascular hypertension. The operation of porto-caval transposition of the vessels results in a hypotensive effect in the animals with renovascular hypertension. 相似文献

Ten patients with severe liver disease and the hepatorenal syndrome underwent exchange transfusions with 9 to 16 units of fresh heparinized blood to improve renal function by either removing a vasoconstrictive substance or adding a vasodilatory substance. One patient recovered from renal failure within ten days without showing natriuresis. The renal function of one patient improved somewhat, but he died 35 days after the transfusion. The other eight aptients died from 1 to 35 days after exchange transfusion, without any appreciable improvement in renal function. In six patients, renin substrate levels increased after the transfusion, but renal function remained unchanged. The results of this study failed to support a humoral concept of pathogenesis of the hepatorenal syndrome. 相似文献

Some reaction parameters were tested in the hydrolysis of casein by papain, in order to prepare hydrolysates with high oligopeptide contents, for either dietetic or pharmaceutical purposes. Five casein hydrolysates were prepared and then fractionated by size‐exclusion HPLC. The rapid correct fraction area method was used for quantifying peptides and free amino acids. Among the five reaction conditions tested, three produced similar peptide profiles. However, the use of a temperature of 37°C and an E:S ratio of 2% is probably the most economical condition for use in large‐scale manufacture. With the aim of masking the bitterness of these preparations, a new method, based on the encapsulation in lipospheres, was used. Also, second derivative spectrophotometry was used for the first time to measure the extent of encapsulation of protein hydrolysates, which changed from 50% to 83%. Moreover, the efficiency of this system was evaluated by analysing other parameters, which showed a reduction of hydrophobicity and bitterness of all samples, as well as good chemical stability during 60 days of storage under refrigeration. The Drosophila melanogaster genes, transient receptor potential (trp) and transient receptor potential-like (trpl) encode putative plasma membrane cation channels TRP and TRPL, respectively. We have stably co-expressed Drosophila TRPL with a Drosophila muscarinic acetylcholine receptor (DM1) in a Drosophila cell line (S2 cells). Basal Ca2+ levels measured using Fura-2/AM in unstimulated S2-DM1-TRPL cells were low and indistinguishable from untransfected cells, indicating that the TRPL channels were not constitutively active in this expression system. Activation of DM1 receptor in S2-DM1-TRPL cells by 100 microM carbamylcholine induced Ca2+ release from an intracellular Ca2+ pool followed by a Gd(3+)-insensitive Ca2+ influx. Pretreatment of S2-DM1-TRPL cells with 10 microM atropine abolished Gd(3+)-insensitive Ca2+ influx triggered by carbamylcholine, but the response was not blocked by prior incubation with pertussis toxin. TRPL channels could also be reliably activated by bath application of 1 microM thapsigargin for 10 min or 100 nM thapsigargin for 60 min in Ca(2+)-free solution. In some cells, TRPL channels activated by thapsigargin could further be activated by carbamylcholine. The findings suggest that, when stably expressed in the S2 cell line, TRPL may be regulated by two distinct mechanisms: (i) store depletion; and (ii) stimulation of DM1 receptor via pertussis-toxin insensitive G-protein (or the subsequent activation of PLC), but without further requirement for Ca2+ release. 相似文献

In the present research it is reported the synthesis and characterization of CdS nanoparticles (NPs) prepared using carboxylic-functionalized poly (vinyl alcohol) (PVA) as the ligand via aqueous route at room temperature and ambient pressure. Different molar concentrations of carboxylic-PVA and PVA were investigated aiming at producing stable colloidal systems. Carboxylic-PVA was conjugated with BSA (bovine serum albumin) and used as capping ligand in the preparation of CdS nanocrystals. UV-visible spectroscopy, photoluminescence spectroscopy, and transmission electron microscopy were used to characterize the kinetics and the relative stability of polymer-capped CdS nanocrystals. The results have clearly indicated that the carboxylic-functionalized PVA was much more effective on nucleating and stabilizing colloidal CdS nanoparticles in aqueous suspensions compared to PVA. In addition, the CdS nanocrystals were obtained in the so-called “quantum-size confinement regime”, with the calculated average size below 4.0 nm and fluorescent activity. Thus, a novel simple route was successfully developed for synthesizing nanohybrids based on quantum dots and water-soluble chemically functionalized polymers with incorporated carboxylic moiety with the possibility of direct bioconjugation. 相似文献

During extraction of crude oil, water is generally present in the oil. This water‐in‐oil (w/o) mixture undergoes turbulent flow that promotes sheer forces, resulting in the appearance of emulsions. These emulsions can be highly stable due to the presence of compounds with polar characteristics such as asphaltenes, which act as natural emulsifiers and form resistant films at the oil–water interface. Nonionic surfactants based on polyoxides are widely used to prevent the formation or to break down w/o emulsions. To shed more light on the destabilization mechanism of w/o emulsions promoted by these surfactants, in this study the techniques of tensiometry and Fourier transform infrared spectroscopy with attenuated total reflectance (FTIR‐ATR) were applied to study the interface formed by poly(ethylene oxide)‐poly(propylene oxide) (PEO‐PPO) block copolymers and asphaltenic petroleum fractions. Initially, the critical micelle concentration of the copolymers in aqueous solution was determined. The results agreed with those found by tensiometry. The bottle test was used to evaluate the break‐down of the w/o emulsions in the presence of the PEO‐PPO block copolymers, and the results presented good agreement with those obtained by tensiometry and FTIR‐ATR. © 2012 Wiley Periodicals, Inc. Polyethersulfone (PES) hollow fiber membranes were fabricated via the dry‐wet phase inversion spinning technique, aiming to produce an asymmetric, micro porous ultrafiltration hollow‐fiber specifically for hemodialysis membrane. The objective of this study is to investigate the effect of spinning conditions on the morphological and permeation properties of the fabricated membrane. Among the parameters that were studied in this work are air gap distance, dope extrusion rate, bore fluid flow rate, and the take‐up speed. The contact angle was measured to determine the hydrophilicity of the fibers. Membrane with sufficient hydrophilicity properties is desired for hemodialysis application to avoid fouling and increase its biocompatibility. The influences of the hollow fiber's morphology (i.e., diameter and wall thickness) on the performance of the membranes were evaluated by pure water flux and BSA rejection. The experimental results showed that the dope extrusion rate to bore fluid flow rate ratio should be maintained at 1:1 ratio to produce a perfectly rounded asymmetric hollow fiber membrane. Moreover, the flux of the hollow fiber spun at higher air gap distance had better flux than the one spun at lower air gap distance. Furthermore, spinning asymmetric hollow fiber membranes at high air gap distance helps to produce a thin and porous skin layer, leading to a better flux but a relatively low percentage of rejection for BSA separation.
