2-(2,4-Dimethylphenyl)indan-1,3-dione was shown to be a potent hypolipidemic agent in rodents, lowering significantly both serum cholesterol and triglyceride levels at 20 mg/kg/day. The agent in vivo inhibited the enzymatic activities of ATP-dependent citrate lyase, acetyl-CoA synthetase, cholesterol-7α-hydroxylase, acyl-CoA cholesterol acyl transferase,sn-glycerol-3-phosphate acyl transferase and phosphatidylate phosphohydrolase. Tissue lipid levels of liver and small intestine also were reduced by the agent. The rat serum lipoprotein lipid content was modulated by the drug, which should be favorable for the removable of cholesterol from peripheral tissue for conduction to the liver for clearance from the body. Low density lipoprotein (LDL) cholesterol levels were reduced after treatment, which suggests that the agent potentially reduces deposition of cholesterol in plaques. If chemotherapy for atherosclerosis is to be successful, then the high density lipoprotein (HDL) cholesterol level needs to be elevated more than 16% to 25%, the level produced by current hypolipidemic agents. 2-(2,4-Dimethylphenyl)-indan-1,3-dione offers a 75% increase in HDL cholesterol levels and a 30% reduction of LDL cholesterol levels with a suppression of de novo synthesis of lipids and a reduction of tissue cholesterol deposition.  相似文献

We have determined the sequence requirements for a protein hinge in triosephosphate isomerase. The codons encoding the hinge at the C-terminus of the active-site lid of triosephosphate isomerase were replaced with a genetic library of all possible 8,000 amino acid combinations. The most active of these 8,000 mutants were selected using in vivo complementation of a triosephosphate isomerase deficient strain of E. coli, DF502. Approximately 3% of the mutants complement DF502 with an activity that is above 70% of wild-type activity. The sequences of these hinge mutants reveal that the solutions to the hinge flexibility problem are varied. Moreover, these preferences are sequence dependent; that is, certain pairs occur frequently. They fall into six families of similar sequences. In addition to the hinge sequences expected on the basis of phylogenetic analysis, we selected three new families of 3-amino-acid hinges: X(A/S)(L/K/M), X(aromatic/beta-branched)(L/K), and XP(S/N). The absence of these hinge families in the more than 60 known species of triosephosphate isomerase suggests that during evolution, not all of sequence space is sampled, perhaps because there is no neutral mutation pathway to access the other families.  相似文献

This paper examines the view that productive teaching in the operating theatre requires a recognition of the unique set of factors which may influence the process of communication. For tutor-student interaction to occur in a productive manner, the tutor's behaviour will need to be modified to suit the demands of the other tasks in which both tutor and student are involved. The particular value of recognizing the role of nonverbal behaviours during interaction is discussed, with the use of specific examples related to the positioning of the tutor and student, and their use of facial expression, the eyes and voice.  相似文献

Experimental data on the diffusion of fluorescent dye plumes were obtained in the nearshore zone of the Great Lakes. The transport and diffusion of continuous dye plumes may be studied either in a frame of reference moving with the centre of gravity of the plume or in a frame of reference fixed to the stationary source. Conventionally, the former is referred to as “relative” diffusion and the latter as “absolute” diffisuion. The inter-link between the two concepts is explained by the random movements of the centre of gravity of the diffusing dye plume usually referred to as the “meandering”.In the past, experimental data from continuous dye plumes in coastal currents have been used to study “relative” diffusion, neglecting the random movements of the centre of gravity. In reality, however, “meandering” appears to be a more efficient agency than “relative” diffusion from the practical point of view of dispersing effluents over larger volume of the water body.In the present analysis, particular attention is given to delineate the two component physical processes of “relative” diffusion and “meandering” diffusion, by extended sampling of the dye plume to cover time scales of both the processes. Calculated horizontal eddy diffusivity Ky in “absolute” diffusion is greater by a factor of 2 as compared to “relative” diffusion. Very high peak/mean concentration ratios observed as the centre of the plume are a direct result of “meandering” due to large scale turbulent eddies.  相似文献

We studied in 10 supine anesthetized dogs diaphragm contraction produced by electrical activation with intramuscular electrodes surgically implanted in the ventral surface of the diaphragm and compared this with activation of the ipsilateral phrenic nerve (C5, 6, and 7) before it entered the thorax. Repetitive 40-Hz pulse trains with supramaximal current stimulus were used after hyperventilation of the animals to apnea. A single intramuscular electrode within 1 to 2 cm of the site of phrenic nerve entry into the diaphragm produced a mean transdiaphragmatic pressure of 12.0 cm H2O +/- 0.97 SE and mean tidal volume of 0.27 L +/- 0.04 SE. Mean values observed with phrenic nerve stimulation were not statistically different, and both electrode systems produced equivalent outward abdominal motion and upper rib cage paradox, as monitored by inductive plethysmography. There was no difference in gas exchange during stimulation with a single hemidiaphragm electrode and mechanical ventilation compared at the same tidal volume and respiratory rate. Blockade of neuromuscular transmission with curare eliminated intramuscular and phrenic nerve stimulation proportionately, suggesting that activation of the diaphragm is dependent in both cases on the phrenic nerve. This technique does not entail manipulation of the phrenic nerve and may have clinical application as an alternative technique for diaphragm pacing.  相似文献

An accurate estimation of reference evapotranspiration (ET0) is of paramount importance for many studies such as hydrologic water balance, irrigation system design and management, crop yield simulation, and water resources planning and management. In the present study, Blaney-Criddle, Jensen-Haise and Hargreaves (temperature based), Priestley-Taylor, Radiation and Makkink (radiation based) and, Pan Evaporation and Christiansen (pan evaporation based) methods have been evaluated and recalibrated with respect to FAO-56 Penman-Monteith method for estimating daily ET0 in the semi-arid Tirupati, Nellore, Rajahmundry, Anakapalli and Rajendranagar sites of Andhra Pradesh, India. Recalibrated Blaney-Criddle (temperature based), Radiation (Radiation based) and Christiansen (Pan evaporation based) methods showed a satisfactory performance at the sites. Further, recalibrated Blaney- Criddle method showed relatively better performance than Radiation and Christiansen methods in the daily ET0 estimation. Recalibrated Blaney- Criddle method may therefore be adopted at the sites selected for the present study and also at the sites with similar climatic conditions for satisfactory daily ET0 estimation.  相似文献

In this paper, parameter optimization of FSW of cryorolled AA2219 alloy was carried out to obtain defect free weld joint with maximum weld strength. To achieve this, artificial neural network (ANN) was used to model the relationship between the input parameters and the mechanical and corrosion properties (output) of the weld joints. The optimal FSW parameters were determined by genetic algorithm (GA). The feasible solution of the GA was tool rotational speed of 1005 rpm, tool travel speed of 20 mm/min and tool tilt angle of 3°. The feasible parameter was used to weld and check the ability of the parameter to produce better weld joint than the L9 orthogonal array parameters. The weld, subjected to the confirmation test, was investigated by means of metallurgical, mechanical, and corrosion testing. This process reduces the costs associated with trial runs to obtain optimal parameters and also the production cost of the cryorolled (CR) plate which is high.  相似文献
