Recent insight into the molecular mechanisms of biological mineral formation (biomineralization) has enabled biomimetic approaches for the synthesis of functional organic‐inorganic hybrid materials under mild reaction conditions. Here we describe a novel method for enzyme immobilization in thin (nanoscale) conformal mineral coatings using biomimetic layer‐by‐layer (LbL) mineralization. The method utilizes a multifunctional molecule comprised of a naturally‐occurring peptide, protamine (PA), covalently bound to the redox enzyme Glucose oxidase (GOx). PA mimics the mineralizing properties of biomolecules involved in silica biomineralization in diatoms, and its covalent attachment to GOx does not interfere with the catalytic activity. Highly efficient and stable incorporation of this modified enzyme (GOx‐PA) into nanoscale layers (~5–7 nm thickness) of Ti‐O and Si‐O is accomplished during protamine‐enabled LbL mineralization on silica spheres. Depending on the layer location of the enzyme and the type of mineral (silica or titania) within which the enzyme is incorporated, the resulting multilayer biocatalytic hybrid materials exhibit between 20–100% of the activity of the free enzyme in solution. Analyses of kinetic properties (Vmax, KM) of the immobilized enzyme, coupled with characterization of physical properties of the mineral‐bearing layers (thickness, porosity, pore size distribution), indicates that the catalytic activities of the synthesized hybrid nanoscale coatings are largely determined by substrate diffusion rather than enzyme functionality. The GOx‐PA immobilized in these nanoscale layers is substantially stabilized against heat‐induced denaturation and largely protected from proteolytic attack. The method for enzyme immobilization described here enables, for the first time, the high yield immobilization and stabilization of enzymes within continuous, conformal, and nanoscale coatings through biomimetic LbL mineralization. This approach will likely be applicable to a wide variety of surfaces and functional biomolecules. This paper discusses the systematization effort undertaken to generate an equipment specification of an extremely detailed and new nature for DCMEs in accordance with the INTELSAT specification IESS-501. The main areas of importance addressed are additional to what has already been defined within the IESS-501. In this paper a new rotor position observer for permanent magnet synchronous machines (PMSM) based on an Extended-Kalman-Filter (EKF) is presented. With this method, just one single EKF is sufficent to evaluate the position information from electromotive force (EMF) and anisotropy. Thus, the PMSM can be controlled for the entire speed range without a position sensor and without the need to switch or synchronize between different observers. The approach covers online estimation of permanent magnetic field and mechanical load. The resulting EKF-based rotor position estimator is embedded in the existing cascaded control concept of the PMSM without need of additional angle trackers or signal filters. The experimental validation for the position sensorless control shows optimized dynamic behaviour.
AbstractIn this day and age, it is widely argued that stakeholder engagement in water-related decision-making processes yields many benefits, including legitimacy, acceptance and trust. Key legal frameworks, such as the European Water Framework Directive and the Aarhus Convention, have spurred the emergence of formal forms of stakeholder engagement. On the other hand, many engagement processes are spontaneous and self-organized. This article investigates the strategies used in formal (government-led) and informal (bottom-up) engagement processes in search of a middle ground. To this end, case studies in the Netherlands, the United States, Uganda and Ethiopia are analyzed using the OECD’s stakeholder engagement checklist. We conclude with reflection on the ways forward to make formal and informal stakeholder engagement complementary. 相似文献
Reaction-bonded Al2O3 (RBAO) ceramics were fabricated starting from mechanically alloyed Al2O3/Al, Al2O3/ Al/ZrO2, and Al2O3/Al/ZrO2/Zr mixtures. Isopressed compacts were heat-treated in air up to 1550°C. Reaction-bonding mechanisms, kinetics, and the influence of ZrO2 and Zr additions are investigated. Independent of additive, oxidation of Al proceeds both as solid/gas and liquid/gas reaction, and the reaction kinetics follow a parabolic rate law. The reaction rate depends strongly on the particle size of Al. The activation energy of the reaction depends essentially on green density. Below the melting temperature of Al, in samples containing 45 vol% Al and 55 vol% Al2O3, it is 112 and 152 kJ/mol at ∼64% and ∼74% TD, respectively, while above the melting temperature, it lies in the range ∼ 26–33 kJ/mol. Zr additions reduce the activation energy to some extent. Samples with only ZrO2 additions exhibit nearly the same activation energies as ZrO2-free samples, though ZrO2 has a very positive effect on the microstructural development in RBAO ceramics. Microstructure evolution and some strength data of RBAO bodies are also reported. 相似文献
High performance liquid chromatography-electrospray tandem mass spectrometry was used to elucidate the phospholipids in krill oil extracted from Euphausia superba, an emerging source for human nutritional supplements. The study was carried out in order to map the species of the choline-containing phospholipid classes: phosphatidylcholine and lyso-phosphatidylcholine. In addition, the prevalent phosphatidylcholine class was quantified and the results compared with prior analysis. The qualification was performed with separation on a reverse phase chromatography column, while the quantification was obtained with class separation on a normal phase chromatography column. An Orbitrap system was used for the detection, and pulsed-Q dissociation fragmentation was utilized for the identification of the species. An asymmetrical exclusion list was applied for detection of phospholipid species of lower concentration, significantly improving the number of species observed. A total of 69 choline-containing phospholipids were detected, whereof 60 phosphatidylcholine substances, among others seven with probable omega-3 fatty acids in both sn-1 and sn-2. The phosphatidylcholine concentration was estimated to be 34 ± 5 g/100 g oil (n = 5). These results confirm the complexity of the phospholipid composition of krill oil, and the presence of long chained, heavily unsaturated fatty acids. 相似文献
