Breast cancer is a leading cause of death among women in the USA. The efficacy of existing anticancer therapeutics can be improved by targeting them through conjugation with ligands binding to cellular receptors. Recently, we developed a novel drug targeting strategy based on the use of pre-selected cancer-specific 'fusion pVIII proteins' (fpVIII), as targeting ligands. To study the efficiency of this approach in animal models, we developed a panel of breast cancer cell-binding phages as a source of targeted fpVIIIs. Two landscape phage peptide libraries (8-mer f8/8 and 9-mer f8/9) were screened to isolate 132 phage variants that recognize breast carcinoma cells MCF-7 and ZR-75-1 and internalize into the cells. When tested for their interaction with the breast cancer cells in comparison with liver cancer cells HepG2, human mammary cells MCF-10A cells and serum, 16 of the phage probes selectively interacted with the breast cancer cells whereas 32 bound both breast and liver cancer cells. The most prominent cancer-specific phage DMPGTVLP, demonstrating sub-nanomolar Kd in interaction with target cells, was used for affinity chromatography of cellular membrane molecules to reveal its potential binding receptor. The isolated protein was identified by direct sequencing as cellular surface nucleolin. This conclusion was confirmed by inhibition of the phage-cell interaction with nucleolin antibodies. Other prominent phage binders VPTDTDYS, VEEGGYIAA, and DWRGDSMDS demonstrate consensus motifs common to previously identified cancer-specific peptides. Isolated phage proteins exhibit inherent binding specificity towards cancer cells, demonstrating the functional activity of the selected fused peptides. The selected phages, their peptide inserts and intact fusion proteins can serve as promising ligands for the development of targeted nanomedicines and their study in model mice with xenograft of human cells MCF-7 and ZR-75-1. 相似文献

The objective of this study was to determine effects of pre rigor stretching on beef tenderness development. Beef loins (M. longissimus dorsi; n=24) were assigned to either stretching or non-stretching treatments and aged for 14days at -1.5°C. Sarcomere length, shear force, water-holding capacity and proteolysis were determined for the loins at 1, 7 and 14days of ageing. Stretching increased the length of the loins by 33% resulting in a trend of increasing sarcomere length (P=0.19). However, no significant differences for shear force values, purge, drip and cooking losses between treatments were found. Shear force values decreased with ageing times (P<0.05). Western blot assay found that stretching did not affect the extent of desmin degradation. The findings from the present study suggest that pre rigor stretching with the current increase in length will not contribute to tenderness improvement of the beef loins. 相似文献

ABSTRACT: Certain reactions that occur in food during storage, such as nonenzymatic browning and lipid oxidation, form compounds that have been shown to be mutagenic. It is possible that over long storage periods, significant amounts of these products could be formed. Although some research has been published concerning the mutagenicity of foods due to processing or cooking, little research has been done regarding mutagenicity of foods stored for an extended time. The objective of this research was to determine the potential mutagenicity of white rice held in accelerated and long-term storage using the Ames Salmonella /microsome assay. Fresh long-grain white rice was packaged in foil laminate pouches and held at 60 °C for 18 wk. Rice stored for > 25 y in an oxygen-free environment at or below room temperature in size number 10 cans was obtained from residential storage. The standard plate-incorporation method was used to evaluate the mutagenic potential of all treatments using Salmonella typhimurium tester strains TA97a, TA98, TA100, and TA102. Samples were plated at 5 dilutions with and without rat liver S9 enzyme. A solvent control was also plated for each strain. Treatments yielding counts at least double the solvent control level were considered mutagenic. Plate counts for all treatments fell well below the required doubling of the solvent control value. White rice held in accelerated and long-term storage appears not to increase in mutagenic compounds as measured by the Ames assay, supporting its use for long-term storage purposes such as emergency preparedness and humanitarian food aid. 相似文献

We present an analysis of the combustion synthesis mechanisms for the preparation of hexaboride materials using three compounds as model systems: EuB(6), YbB(6), and YB(6). These three hexaborides were chosen because of the differences in ionic radii between Eu(3+), Yb(3+), and Y(3+), which is a factor in their stability. The powders were prepared using metal nitrates, carbohydrazide, and two different boron precursor powders. The resulting materials were analyzed by X-ray diffraction, which showed that combustion synthesis is effective for the synthesis of EuB(6), since the Eu(3+) ion has an ionic radius greater than ~1 ?. The synthesis of YbB(6) and YB(6) is not as effective because of the small size of the Yb(3+) and Y(3+) ions, making the hexaborides of these metals less stable and resulting in the synthesis of borates due to the presence of oxygen during the combustion process. Scanning electron microscopy and dynamic light scattering of the EuB(6) powders shows that the particle size of the hexaboride product is dependent on the particle size of the boron precursor. The magnetic susceptibility of our EuB(6) powders manifests irreversible behavior at low applied fields, which disappears at higher fields. This behavior can be attributed to the increase in size and number of magnetic polarons with increasing magnetic field. 相似文献

Intensity modulated photocurrent (IMPS) and photovoltage (IMVS) spectroscopies were used to study the mechanism of photoprocesses in P3HT:PCBM bulk heterojunction organic solar cells at various light intensities. The use of the frequency domain techniques allowed us to separate the bulk and interfacial processes and gain a valuable insight into the mechanism of losses in these devices. The results provide direct evidence that interfacial nongeminate recombination is one of the dominant loss and aging mechanisms in bulk heterojunction organic solar cells. The trapping of photoexcited holes in the P3HT phase was found to contribute to the increased recombination rate. The results suggest that promising ways of improving the efficiency of bulk heterojunction solar cells may be reducing the charge trapping both at and near the P3HT:PCBM interface, as well as improving the efficiency of charge extraction at contacts. 相似文献

The production of thermally concentrated fruit juices uses temperatures high enough to achieve at least a 5-log reduction of pathogenic bacteria that can occur in raw juice. However, the transportation and storage of concentrates at low temperatures prior to final packaging is a common practice in the juice industry and introduces a potential risk for postconcentration contamination with pathogenic bacteria. The present study was undertaken to evaluate the likelihood of Escherichia coli O157:H7, Listeria monocytogenes and Salmonella surviving in cranberry, lemon, and lime juice concentrates at or above temperatures commonly used for transportation or storage of these concentrates. This study demonstrates that cranberry, lemon, and lime juice concentrates possess intrinsic antimicrobial properties that will eliminate these bacterial pathogens in the event of postconcentration recontamination. Bacterial inactivation was demonstrated under all conditions; at least 5-log Salmonella inactivation was consistently demonstrated at -23 degrees C (-10 degrees F), at least 5-log E. coli O157:H7 inactivation was consistently demonstrated at -11 degrees C (12 degrees F), and at least 5-log L. monocytogenes inactivation was consistently demonstrated at 0 degrees C (32 degrees F). 相似文献

The functional properties of whole, diced or minced high and normal pH beef were determined after 0, 1, 2, 3 and 7 month’s storage. There was no interaction between pH, degree of comminution and storage time for most of the attributes measured. Regardless of storage time or comminution method, high pH meat had significantly (P < 0.001) higher sarcoplasmic protein solubility, water holding capacity, cooked batter torsion stress and strain, yield and emulsion stability than normal pH meat. Normal pH meat had significantly (P < 0.01) higher myofibrillar protein solubility and hue angle than high pH meat. Water holding capacity significantly (P < 0.01) decreased and hue angle increased with the increase in the degree of comminution. Protein solubility in raw meat and the cooked batter stress, strain, yield and emulsion stability of both high and normal pH meat deteriorated with time. However, this deterioration was faster in the normal pH meat compared to high pH meat except for total and myofibrillar protein solubility. Within the parameters of the present study, it is concluded that frozen high pH meat possesses superior functional attributes compared to normal pH meat regardless of the degree of comminution or storage time. 相似文献

In this report, the influence of the intrinsic transitions between bound-to-delocalized states (crossed states or quasicontinuous density of electron-hole states) on photoluminescence excitation (PLE) spectra of InAs quantum dots (QDs) was investigated. The InAs QDs were different in size, shape, and number of bound states. Results from the PLE spectroscopy at low temperature and under a high magnetic field (up to 14 T) were compared. Our findings show that the profile of the PLE resonances associated with the bound transitions disintegrated and broadened. This was attributed to the coupling of the localized QD excited states to the crossed states and scattering of longitudinal acoustical (LA) phonons. The degree of spectral linewidth broadening was larger for the excited state in smaller QDs because of the higher crossed joint density of states and scattering rate. 相似文献
