Skin tumour development was studied in groups of mice painted once with 125 mug of 3-methylcholanthrene (MCA) either at 12:00 or at 24:00 MET. Eight animals were kept in each box. The animals were observed weekly for 20 months and all tumours were registered. There was no difference between the two groups of mice as regards tumour induction time or number of papilloma-bearing mice. In the groups of mice treated at 24:00 the number of skin tumours to develop was 9 per cent higher than in groups of mice treated at 12:00. This difference in papilloma yields is not statistically significant. Among female mice painted at 24:00 carcinoma-bearing animals were significantly more numerous (50 per cent) than among those painted at 12:00, whereas there was no difference between the groups of male mice. Considering the groups collectively (males + females), the intergroup difference (17 per cent) in advantage of painting at 24:00 was barely significant (0.5 less than p less than 0.10). There was no difference between the groups as regards the total number of carcinomas to occur. When the tumour yields in individual boxes were found to vary greatly. The slight increase in tumour yield after night painting correlates with the circadian variation in proliferative activity of the epidermidis. Previous reports in the literature have shown similar differences. Further investigations and better methods seem necessary before a definite conclusion can be drawn concerning a possible diurnal variation in the susceptibility of mouse skin to chemical carcinogenesis. It is also emphasized that it is necessary to exercise great caution when the results of classical epidermal chemical carcinogenesis experiments are to be interpreted. It seems necessary to observe animals for at least 15 months before any conclusion can be drawn.  相似文献

Ribonucleic acid (RNA) metabolism of the normal and copper-treated ( Cu-T200 IUD) human endometrium was investigated. The relative concentration of total, messenger, ribosomal and transfer RNA was measured in normal and Cu-treated endometrium using the technique of affinity chromatography in polysepharose. The transition from the proli ferative to the secretory endometrium in normal women was accompanied by significant increases (p less than .05) in total RNA, messenger RNA and in ribosomal RNA. The relative proportions of bound and free messenger RNA were also modified by endometrial maturation changing from 70% bound messenger RNA in the proliferative to 83% in the secretory phase. Cu-T200 Cu release appeared to particularly affect RNA metabolism in the secretory phase. During the proliferative phase only the concentration of transfer RNA and the proportion of bound to free messenger RNA were modified by the Cu-T200. The Cu-T200 induced significant decreases (p less than .01 and p less than .05) in all RNA parameters, with the exception of the RNA/deoxyribonucleic acid ratio.  相似文献

Conducted a study with 32 13-17 yr old male chronic truants from a low-income, inner-city area. Ss were given counseling aimed at returning them to regular attendance. Optimistic vs threat appeal and presence vs absence of supportive instruction were manipulated in a 2 * 2 design, and persuasion was assessed both as verbal intention to attend school regularly and as actual attendance. Supportive instruction included the continuing availability of a tutor. Results show that threat appeal was sufficient to change intention to return to school, but supportive instruction was necessary to change actual attendance. Milk was collected from six mothers during their 8th wk of lactation to determine the diurnal variation of bile salt-stimulated lipase and serum-stimulated lipase. On the day of collection, one breast was completely emptied at 0600, 1000, 1400, 1800, and 2000 h. Activities of bile-stimulated lipase and serum-stimulated lipase in the milk were determined. Activity of bile-stimulated lipase was constant throughout the day, whereas serum-stimulated lipase activity varied significantly with time. Both activities differed significantly in milk from different women. Time of collection must be considered when measuring activity of serum-stimulated lipase but is not a major variable when measuring activity of bile-stimulated lipase in human milk. These results may be helpful when designing future studies involving these enzymes.  相似文献

Diacetyl, noted for its appealing butter‐like aroma, is present naturally in many foods, and humans have been exposed to it since the beginning of civilization. The advent of microwave (MW) cooking technology has led to the development of a significant market for MW popcorn, to which diacetyl and other flavoring compounds have been frequently added. Based upon reported associations between diacetyl inhalation and lung disease in employees of MW popcorn processing facilities, a very conservative safe level of occupational exposure to diacetyl has been proposed by the American Conference of Governmental Industrial Hygienists. Yet there is conflicting evidence that diacetyl causes lung disease in workers, and no evidence to condemn diacetyl as the cause of lung problems in MW popcorn consumers. Consumer dietary exposure to diacetyl in foods is below levels of health concern while common airborne levels of diacetyl from MW popcorn are far below the conservatively established limit to protect workers.  相似文献

Mammalian vocalizations can encode contextual information in both the spectrographic components of their individual vocal units and in their temporal organization. Here we observed 23 Holstein-Friesian dairy cows immediately after birth during interactions with their calf and when their calf was separated to the other side of a fence line. We investigated whether the vocalizations emitted in these postpartum contexts would vary temporally. We also described the maternal and stress behaviors preceding and following postpartum vocal production using kinematic diagrams and characterized call sequence structure. The kinematic diagrams highlight the disruption of maternal responses caused by calf separation and show that behavioral and vocal patterns varied according to the cows' emotional states and proximity to the calf in both contexts. During calf interactions, cows mainly produced closed-mouth calls simultaneous to licking their calf, whereas an escalation of stress responses was observed during calf separation, with the cows approaching the fence line, becoming alert to the calf, and emitting more mixed and open-mouth calls. Call sequences were similarly structured across contexts, mostly containing repetitions of a single call type, with a mean interval of 0.57 s between calls and a greater cumulative vocalization duration, attributed to an increased number of vocal units per sequence. Overall, calf separation was associated with a greater proportion of calls emitted as a sequence (inverse of single isolated calls), a shorter interval between separate call sequences, and a greater number of vocal units per sequence, compared with calf interactions. These temporal vocal features varied predictably with the high stress expression from cows during calf separation and may represent temporal modulations of emotional expression. Despite the noisy farm soundscape, empirical call type and temporal vocal features were easy to measure; thus, findings could be applied to future cattle studies wishing to analyze vocalizations for on-farm welfare assessments.  相似文献

Inconsistent evidence of inflammatory immune cell infiltrates in adipose tissues with extensive triglyceride mobilization raises the possibility that regulatory or anti-inflammatory immune cell populations reside within the mesenteric adipose tissue (MAT) and mesenteric lymph nodes (MLN). These resident immune cell populations may be involved in attenuating the inflammatory response. We explored the immune cell population of MAT and MLN collected from lean, lactating Holstein cows without apparent disease in an abattoir (n = 42). Lean cows had a body condition score of 2.6 ± 0.1 (mean ± SD) with a greater frequency of adipocyte area occurring in small rather than large adipocytes. Cells were labeled with monoclonal antibodies specific to bovine leukocyte antigens for enumeration by flow cytometry. Within both lymph node and adipose tissues, relatively large subpopulations of cells expressed the β2 integrins CD11b and CD11c, class II major histocompatibility antigens (MHCII), and the SIIRP-1α receptor (CD172a) typical of dendritic cells and macrophages. Macrophage/dendritic cell heterogeneity was marked by β2 integrin expression alone or in conjunction with CD172a or MHCII across subpopulations from both tissues; CD209, the DC-SIGN c-type lectin receptor of dendritic cells, was not detected by fluorescence-activated cell sorting in either tissue. Lymphocytes comprised 74.1 ± 3.7% and 13.7 ± 3.7% of the MLN and MAT cell populations, respectively, and CD3+CD4+ lymphocytes accounted for 49.8 ± 9.9% of the MLN and 6.13 ± 1.23% of the MAT cells. Fox P3+ regulatory lymphocytes comprised 15.3 ± 1.1% and 6.73 ± 0.52% of the MLN and MAT cells, whereas γδ+ lymphocytes accounted for 6.65 ± 0.74% and 3.91 ± 0.43% of the MLN and MAT cells, respectively. Subpopulations of CD3+CD8+ cytotoxic T cells and CD3+CD11c+ innate lymphocytes were present in MLN but not MAT. These results show that subpopulations of resident tissue macrophages, dendritic cells, T helper lymphocytes, regulatory T lymphocytes (Tregs), and γδ lymphocytes reside in mesenteric lymph nodes and adipose tissues. Balance in the innate and adaptive immune functions embedded in these tissues could support metabolic health.  相似文献

Liver biopsies from 38 multiparous Holstein cows were used to determine rates of peroxisomal beta-oxidation and total beta-oxidation of palmitate in liver homogenates and contents of total lipid, triglyceride, and glycogen during the lactation cycle. Cows were assigned to one of four diets from wk 4 through wk 42 of lactation: control, control plus nicotinic acid (12 g/d), supplemental fat, or supplemental fat plus nicotinic acid. Liver biopsies were obtained at wk 3 (covariate), 6, 12, 24, and 42 of lactation. Neither supplemental fat nor nicotinic acid affected palmitate oxidation in liver homogenates or liver composition. Peroxisomal beta-oxidation capacity and the ratio of peroxisomal to total beta-oxidation decreased from wk 3 to 12 and then increased at wk 42. Contents of total lipid and triglyceride decreased, and content of glycogen increased, from wk 3 to 12. Total oxidation capacity in liver homogenates was correlated negatively with total lipid and triglyceride in liver, yields of milk and solids-corrected milk (SCM), and plasma nonesterified fatty acids (NEFA), and was correlated positively with liver glycogen, dry matter intake (DMI), energy balance, and plasma glucose. Peroxisomal beta-oxidation was correlated negatively with yields of milk and SCM. The ratio of peroxisomal to total beta-oxidation was correlated positively with liver total lipid, liver TG, and plasma NEFA and negatively with DMI and energy balance. When only data from wk 3 postpartum were considered, both total and peroxisomal beta-oxidation were correlated negatively with hepatic concentrations of total lipid and TG. Peroxisomal beta-oxidation in liver of dairy cows is not affected by feeding supplemental fat or nicotinic acid during wk 4 to 42 of lactation but may be a part of the hepatic adaptations to negative energy balance.  相似文献

Membrane filtration (microfiltration and ultrafiltration) has become an accepted process for drinking water treatment, but membrane fouling remains a significant problem. The objective of this study was to systematically investigate the mechanisms and components in natural waters that contribute to fouling. Natural waters from five sources were filtered in a benchtop filtration system. A sequential filtration process was used in most experiments. The first filtration steps removed specific components from the water, and the latter filtration steps investigated membrane fouling by the remaining components. Particulate matter (larger than 0.45 microm) was relatively unimportant in fouling as compared to dissolved matter. Very small colloids, ranging from about 3-20 nm in diameter, appeared to be important membrane foulants based on this experimental protocol. The colloidal foulants included both inorganic and organic matter, but the greatest fraction of material was organic. When the colloidal fraction of material was removed, the remaining dissolved organic matter (DOM), which was smaller than about 3 nm and included about 85-90% of the total DOM, caused very little fouling. Thus, although other studies have identified DOM as a major foulant during filtration of natural waters, this work shows that a small fraction of DOM may be responsible for fouling. Adsorption was demonstrated to be an important mechanism for fouling by colloids.  相似文献
