We have recently reported that thymic B lymphocytes (TBL) are the first B-cell subpopulation undergoing isotype switching to IgG and IgA during embryonic life. The aim of this study is to analyze the influence of antigenic stimulation on TBL location and activity using a germ-free (GF) newborn pig model, in which maternal antibodies and antigens do not affect B-cell development. Immunohistological analysis showed that TBL were disseminated mainly in the thymic medulla. There were no differences in the distribution of TBL, both in GF newborn piglets before and after colonization with Escherichia coli and in older conventionally reared (CONV) piglets. The number of immunoglobulin (Ig)-secreting cells measured by the ELISPOT method was not influenced by microflora and food antigens. IgM-positive cells secreting IgM and CD45RC-positive cells spontaneously producing IgM, IgG, and IgA were detected in newborn thymus. Our findings suggest that TBL differentiation and Ig switching to IgG and IgA-secreting cells is not influenced by external antigens and that the thymic microenvironment plays an important role in this process.  相似文献

Distributed interactive simulation (DIS) is a revolutionary development that is changing permanently many features of Defense Department work. The field of warfare modeling and simulation (M&S) is quite large, however, and this paper reviews its evolution and how DIS fits within it. A few things are clear. First, DIS is exceedingly valuable for training. Second, DIS could be a powerful means for improving the quality of planning and analysis if used wisely for occasional well designed experiments. These could 1) provide insights about real-world processes involving human performance and behavior (including decision making), 2) help inform and calibrate models, and 3) help test plans in a quasioperational environment. Third, distributed war gaming, which depends on DIS technology, is already lowering boundaries among developers, planners, and warfighters. This can shorten development processes and improve the results through virtual prototyping; it can also improve operational readiness. To achieve these potential benefits, however, will require a holistic approach, conceptual breakthroughs, and profoundly difficult model development efforts. The challenges include developing integrated hierarchies of models, developing adaptive decision models and other models of human behavior, developing and using new forms of uncertainty-sensitive analysis, and learning how to use DIS experiments effectively  相似文献

Process capability indices (PCIs) are used in industry to assess percentages of nonconforming parts. An underlying assumption is that the output process measurements are distributed as normal random variables. When normal distributions are assumed, but different distributions are present - such as skew, heavy-tailed, and short-tailed distributions - the percentages of nonconforming parts are significantly different than the computed PCIs indicate. Data arising from nonnormal distributions can sometimes be transformed to conform to the normality assumption and the PCI's computed for the transformed data. In this paper, the effect of the transformation on the estimate of nonconforming parts is examined for three examples of nonnormal distributions - gamma, lognormal, and Weibull. The results of this experimental analysis suggest that data transformation can be useful for estimating an interval for Cpk values and the number of nonconforming parts.  相似文献

Plasma HIV-1 RNA testing was used to monitor 43 HIV-1 infected patients newly placed on antiretroviral therapy or whose therapy had been recently changed. A polymerase chain reaction kit was used to measure HIV-1 RNA in clinical samples or frozen plasma. The cutoff of this test was 200 RNA copies/ml. The first group (11 patients) was stable on long-term zidovudine monotherapy when switched to stavudine. The HIV-1 RNA of three patients who had a regular decline in CD4+ T cell count did not change despite this switch, with a mean follow-up of 630 days. The HIV-1 RNA copy numbers of eight patients whose CD4+ T cell counts were stable declined an average of 0.53 log10 between days 90 and 650. The second group (14 patients) was on long-term zidovudine monotherapy and had declining CD4+ T cell counts over the past 6 months. Lamivudine was added to this regimen on day 0. HIV-1 RNA copy number decreased rapidly within 30 d, reaching -0.86 log10 on day 90, and this effect was maintained thereafter, with a mean follow-up of 161 days. There was a concomitant mean gain of +33 CD4+ T cells on day 90. The third group (nine patients) had never received anti-retroviral therapy and was given zidovudine+didanosine. HIV-1 RNA copy number decreased in all cases but one, reaching -1.31 log10 on day 150. This decrease was transient in three cases. The last group (nine patients) had also not had previous anti-retroviral therapy and was given zidovudine + didanosine + lamivudine in combination. HIV-1 RNA copy numbers declined rapidly in all cases, to below the cutoff in eight cases within a mean period of 50.5 days. The CD4+ cell counts increased by 164 cells/microliter on day 14 and by 201 cells/microliter on day 180. The response to therapy of the total population of 43 patients varied according to cases. The relative changes in p24 antigen compared to HIV-1 RNA also differed between patients. Measurement of HIV-1 viremia appears to be a valuable tool in current practice for individualizing therapy.  相似文献

Cyclic voltammetric and galvanostatic experiments have been carried out with both synthetic and commercial bleach solutions with a view to regenerating the same. Maximum efficiency could be achieved if the oxidation is carried out below an anodic potential of 0·80 V vs SCE. According to our common medical culture, some facts are simply unquestionable, for instance La?nnec invented the stethoscope. But was he the first one? On a recent trip to Egypt we visited the temple of Kom Ombo, built prior to the roman period and renowned as a medical care center. Today, the tourist is fascinated by the magnificent hieroglyphics on the well-preserved walls testifying to significant advances in various fields of medicine including ophthalmology and gynecology. We were particularly interested by the basreliefs presenting vivid drawings of some of the first medical instruments. We easily identified curettes, scissors, a balance, forceps for dental extraction, and a surgical saw, but were captivated by two other instruments. The first one looked a lot like what La?nnec invented around 1820. The second one was amazingly similar to the instrument we use everyday, with a distal opening and flexible tubes (woven papyrus?) leading to proximal ear pieces. Our Egyptian guide was formal: the stethoscope was invented in Egypt. The scientific impact of our observations leaves something to be desired, but did make us think about the huge gap between the advancement of medical knowledge in ancient Egypt and La?nnec's (re)-invention. Exposed to a similar gap in history, what would our documents stored on CD and video tapes have to say to future touring doctors?  相似文献
