Background

Silica nanoparticles (SiNPs) have numerous beneficial properties and are extensively used in cosmetics and food industries as anti-caking, densifying and hydrophobic agents. However, the increasing exposure levels experienced by the general population and the ability of SiNPs to penetrate cells and tissues have raised concerns about possible toxic effects of this material. Although SiNPs are known to affect the function of the airway epithelium, the molecular targets of these particles remain largely unknown. Given that SiNPs interact with the plasma membrane of epithelial cells we hypothesized that they may affect the function of Transient Receptor Potential Vanilloid 4 (TRPV4), a cation-permeable channel that regulates epithelial barrier function. The main aims of this study were to evaluate the effects of SiNPs on the activation of TRPV4 and to determine whether these alter the positive modulatory action of this channel on the ciliary beat frequency in airway epithelial cells.

Results

Using fluorometric measurements of intracellular Ca2+ concentration ([Ca2+]i) we found that SiNPs inhibit activation of TRPV4 by the synthetic agonist GSK1016790A in cultured human airway epithelial cells 16HBE and in primary cultured mouse tracheobronchial epithelial cells. Inhibition of TRPV4 by SiNPs was confirmed in intracellular Ca2+ imaging and whole-cell patch-clamp experiments performed in HEK293T cells over-expressing this channel. In addition to these effects, SiNPs were found to induce a significant increase in basal [Ca2+]i, but in a TRPV4-independent manner. SiNPs enhanced the activation of the capsaicin receptor TRPV1, demonstrating that these particles have a specific inhibitory action on TRPV4 activation. Finally, we found that SiNPs abrogate the increase in ciliary beat frequency induced by TRPV4 activation in mouse airway epithelial cells.

Conclusions

Our results show that SiNPs inhibit TRPV4 activation, and that this effect may impair the positive modulatory action of the stimulation of this channel on the ciliary function in airway epithelial cells. These findings unveil the cation channel TRPV4 as a primary molecular target of SiNPs.

The stripper section of a fluid‐coker consists of a system of baffles (sheds) that enhances the removal efficiency of entrained and adsorbed hydrocarbons from the fluidised coke‐particles. If the particles contain a thin liquid film layer of heavy hydrocarbons, making them excessively ‘wet’ or ‘sticky’, and if they stay in contact with sheds for too long, solid deposits are formed that lead to stripper fouling. Extensive fouling decreases stripping efficiency and liquid product yield and can shorten run‐times between shutdowns. Because of the fouling, the shape of sheds mostly changes by increasing their surfaces thickness. An early indication of that fouling and the ability to follow its development are essential for choosing optimal parameters of the process. The radioactive particle tracking (RPT) method has been tested to determine its applicability to indicate the change in the shape of internals within a fluidised bed reactor when direct observation is impossible. A single radioactive tracer‐particle has been traced in experiments lasting from 2 to 6 h. The experiments were conducted in a lab‐scale, cold‐flow fluidised bed into which a single shed with walls of different thickness was incorporated. This experimental fluidised bed provides intensive solid phase mixing that allows a single tracer‐particle to be located in any place within the reactor. By registering the frequency of the tracer‐particle appearance within a defined internal space surrounding the shed, the shape of shed was reconstructed. The mathematical formulation of mass transfer in food processes is often based on the diffusion equation with the diffusion coeffcients considered as effective parameters. In some cases, more than one specie can be simultaneously diffusing in opposite directions. Therefore, the movement of one component could affect the movement of the remaining components. The aim of this work is to describe a mathematical procedure to obtain the diffusion coeffcients of different species that simultaneously diffuse in such a situation that each mass flux is affected by the existence of the others. The correspondent microscopic mass balances combined with Fick's law were simultaneously solved using a numerical finite difference method.  相似文献

Fibrous filter pressure drop and aerosol collection efficiency were measured at low air pressures (0.2–0.8 atm) and high face velocities (5–19 m/s) to give fiber Reynolds numbers lying in the viscous-inertial transition flow regime (1–15). In this regime, contemporary filtration theory based on Kuwabara's viscous flow through an ensemble of fibers underpredicts single fiber impaction by several orders of magnitude. Streamline curvature increases substantially as air stream inertial forces become significant. Dimensionless pressure drop measurements followed the viscous-inertial theory of Robinson and Franklin (1972 Robinson, M. and Franklin, H. 1972. The Pressure Drop of a Fibrous Filter at Reduced Ambient Pressures. J. Aerosol Sci., 3: 413427. [Crossref] [Google Scholar]) rather than Darcy's linear pressure–velocity relationship. Sodium chloride and iron nano-agglomerate aerosols were tested to provide a comparison between particles of dissimilar densities and shape factors. Total filter efficiency collapsed when plotted against the particle Stokes number and fiber Reynolds number. Efficiencies were then modeled with an impactor type equation where the cutpoint Stokes number and a steepness parameter described data well in the sharply increasing portion of the curve (20%–80% efficiency). A minimum in collection efficiency was observed at small Stokes numbers and attributed to interception and diffusive effects. The cutpoint Stokes number was a linearly decreasing function of fiber Reynolds number. Single fiber efficiencies were calculated from total filter efficiencies and compared to contemporary viscous flow impaction theory (Stechkina et al. 1969 Stechkina, I. B., Kirsch, A. Existing theories underpredicted measured single fiber efficiencies, although comparison is problematic. The assumption of uniform flow conditions for each successive layer of fibers is questionable; thus, the common exponential relationship between single fiber efficiency and total filter efficiency may not be appropriate in this regime.

The use of epoxidized cottonseed oil as plasticizer for poly(vinyl chloride) was studied. The plasticizer content was set to 70 phr and the optimum isothermal curing conditions were studied in the temperature range comprised between 160 and 220 °C with varying curing times in the 7.5–17.5 min range. The influence of the curing conditions on overall performance of cured plastisols was followed by the evolution of mechanical properties (tensile tests with measurements of tensile strength, elongation at break, and modulus), change in color, surface changes of fractured samples by scanning electron microscopy (SEM), thermal transitions by differential scanning calorimetry, and migration in n‐hexane. The optimum mechanical features of cured plastisols are obtained for curing temperatures in the 190–220 °C range. For these curing conditions, fractography analysis by SEM gives evidences of full curing process as no PVC particles and free plasticizer can be found. © 2016 Wiley Periodicals, Inc. J. Appl. Polym. Different versions of the cluster expansion are explored using the Mo-Ta system as an example. One of the objectives of this work is to establish a clear distinction between phenomenological expansions that express the energy of an alloy in the form of a generalized Ising model, i.e. with constant pair and many body interactions, and cluster expansions that use a set of complete basis functions in configurational space and define the interactions as projections of the energy onto the basis functions. For the latter case, the interactions are functions of concentration and depend, furthermore, on the full state of order of the system. Such dependence is expected since the configurational energy is shown to be a homogeneous function of degree one in the complete set of configurational variables, or correlation functions, with the interactions being the Euler derivatives of the energy with respect to the correlation functions. For the Mo-Ta system we show that, by including the concentration dependence of the interactions either explicitly or through their dependence on volume, the cluster expansion converges significantly faster than the phenomenological Ising-like models commonly used to represent the energies of disordered alloys.  相似文献

Hybrid photocatalysts based on an adsorbent SiMgOx and a photocatalyst TiO2 were developed in a plate shape. The ceramic surface was coated with TiO2 by the slip-casting technique. The effect of the support in the photocatalytic degradation of trichloroethylene (TCE) was analyzed by modifying TiO2 loading and the layer thickness. Photocatalysts were characterised by N2 adsorption–desorption, mercury intrusion porosimetry, SEM, UV–vis spectroscopy and XRD. A direct relationship between the TiO2 content and the photocatalytic activity was observed up to three layers of TiO2 (0.66 wt.%). Our results indicate that intermediate species generated on the TiO2 layer can migrate through relatively long distances to react with the OH surface groups of the support. By increasing the TiO2 loading of the photocatalyst two effects were observed: trichloroethylene conversion is enhanced, while the efficiency of the oxidation process is decreased at expenses of increasing the selectivity to COCl2 and dichloroacetylchloride (DCAC). The results are discussed in terms of the layer thickness, TiO2 amount, TCE conversion and CO2, and COCl2 selectivity.  相似文献

局部腐蚀,包括点腐蚀、应力腐蚀开裂和腐蚀疲劳是导致电厂发电装置损伤的重要原因,它们诱发50%以上的意外停机事故。迄今,腐蚀破坏的预测一直沿用经验和统计的方法,但是,由于已证实该方法的适用性和其结果的准确性欠佳,故不宜作为例行检修规程的基础。本文提出了预测局部腐蚀损伤的决定论方法,并使用这个方法说明点腐蚀如何引发低压蒸汽轮机涡轮盘Wilson线下游部位的应力腐蚀开裂(SCC),我们的计算表明SCC的萌发与扩展对蒸汽小氧含量、冷凝液膜的环境和涡轮盘所承受的应力这三个因素敏感。  相似文献
