Hematocrits (PCV) were measured at 29 and 106 d of age (PCV1 and PCV2, respectively) in male and female White Plymouth Rocks. Four lines were used, two of which had undergone 40 generations of divergent selection for 8-wk BW (HWS, LWS), and two respective sublines (HWR, LWR), in which selection had been relaxed for five generations. At both ages, males and females did not differ for PCV in lines HWR, LWR, and LWS. For line HWS there was an age by sex interaction that resulted from an age effect for males but not for females, and from a sex effect at each age. At both ages, PCV was higher for the HW than the LW lines. Initially, there was no difference between the selected and their respective relaxed lines, but by 106 d, HWR chickens had a higher PCV than HWS chickens. In lines HWR and LWR, PCV increased with age. There was a negative correlation in HWS males for PCV1 with 28 and 56 d BW. The HWR males also had a negative correlation for PCV1 with BW at 28 d, but not between PCV2 and BW. The correlation for PCV1 with PCV2 was high and positive for HWR males and females. 相似文献

The yeast gene, YTA10, encodes a member of a novel family of putative ATPases. Yta10p, as deduced from the nucleotide sequence, is 761 amino acids in length (predicted molecular mass 84.5 kDa). The amino acid sequence of Yta10p exhibits high similarity to two other yeast proteins, Yta11 and Yta12, and to E. coli FtsH. Several features of Yta10p are compatible with its localization in mitochondria. We report here that Yta10p is a yeast mitochondrial protein and that import is dependent on a membrane potential and accompanied by processing to a protein of approximately 73 kDa. Disruption of YTA10 leads to a nuclear petite phenotype and to a loss of respiratory competence, as shown by spectrophotometric measurement of the activities of respiratory complexes I-III and IV, respectively. These findings together with the high similarity of Yta10p to several ATP-dependent proteases suggest that Yta10p is a mitochondrial component involved, directly or indirectly, in the correct assembly and/or maintenance of active respiratory complexes. 相似文献

The role of 4-hydroxynonenal (HNE), a major lipid peroxidation product, in oxidative damage to mitochondrial cytochrome c oxidase (COX) was examined. Oxidative stress was induced in mitochondria isolated from livers of male Sprague-Dawley rats by tert-butylhydroperoxide (t-BHP). COX activity was inhibited, with a concomitant increase in endogenous HNE level in mitochondria. COX activity was also inhibited following incubation of mitochondria with 50-450 microM HNE. Blocking HNE degradation intensified COX inhibition by HNE and by t-BHP-induced oxidative stress, the latter accompanied by a simultaneous increase in endogenous HNE production. On the other hand, COX inhibition by HNE was markedly reduced by potentiating HNE degradation via enhancing conjugation of HNE with reduced glutathione (GSH). Incubation of purified COX with 10-400 microM HNE resulted in HNE adduct formation with specific subunits of COX, correlated with inhibition of the enzyme activity. These data suggest that HNE may inhibit mitochondrial COX by forming adducts with the enzyme, and that this could be one mechanism underlying mitochondrial damage caused by oxidative stress. Prothymosin alpha (ProTalpha) is an abundant acidic nuclear protein that may be involved in cell proliferation. In our search for its cellular partners, we have recently found that ProTalpha binds to linker histone H1. We now provide further evidence for the physiological relevance of this interaction by immunoisolation of a histone H1-ProTalpha complex from NIH 3T3 cell extracts. A detailed analysis of the interaction between the two proteins suggests contacts between the acidic region of ProTalpha and histone H1. In the context of a physiological chromatin reconstitution reaction, the presence of ProTalpha does not affect incorporation of an amount of histone H1 sufficient to increase the nucleosome repeat length by 20 bp, but prevents association of all further H1. Consistent with this finding, a fraction of histone H1 is released when H1-containing chromatin is challenged with ProTalpha. These results imply at least two different interaction modes of H1 with chromatin, which can be distinguished by their sensitivity to ProTalpha. The properties of ProTalpha suggest a role in fine tuning the stoichiometry and/or mode of interaction of H1 with chromatin. 相似文献

Six consultation cases of mullerian adenosarcoma of the uterus were encountered in women who were receiving adjuvant tamoxifen therapy for carcinoma of the breast. To our knowledge, only one previous similar case has been reported. The women, who were 48-76 years of age, had received tamoxifen for periods of 6 months to 4 years (mean 2.7 years) in five of the cases; the duration of tamoxifen therapy in the sixth case is unknown. All of the tumors were polypoid endometrial masses that superficially invaded the myometrium in two cases. The microscopic appearance of the tumors was similar to that of previously described uterine mullerian adenosarcomas. These and other recent observations indicate that tamoxifen treatment may be complicated by uterine neoplasms other than endometrial adenocarcinoma. These findings also support previous observations that prolonged exogenous or endogenous hyperestrinism may lead to the development of mesenchymal and mixed epithelial-mesenchymal tumors of the uterus. 相似文献

Gene transfer with recombinant murine leukemia viruses (MuLV) provides the potential to permanently correct inherited lung diseases, such as cystic fibrosis (CF). Several problems prevent the application of MuLV-based recombinant retroviruses to lung gene therapy: (i) the lack of cell proliferation in mature pulmonary epithelia, (ii) inefficient gene transfer with a vector applied to the apical surface, and (iii) low titers of many retroviral preparations. We found that keratinocyte growth factor (KGF) stimulated proliferation of differentiated human tracheal and bronchial epithelia. Approximately 50% of epithelia divided in response to KGF as assessed by bromodeoxyuridine histochemistry. In airway epithelia stimulated to divide with KGF, high-titer ampho- and xenotropic enveloped vectors preferentially infected cells from the basal side. However, treatment with hypotonic shock or EGTA transiently increased transepithelial permeability, enhancing gene transfer with the vector applied to the mucosal surfaces of KGF-stimulated epithelia. Up to 35% of cells expressed the transgene after gene transfer. By using this approach, cells throughout the epithelial sheet, including basal cells, were targeted. Moreover, the Cl- transport defect in differentiated CF airway epithelia was corrected. These findings suggest that barriers to apical infection with MuLV can be overcome. 相似文献

As materials with high resistance to brittle fracture, fine-grained structural steels are very important for high-grade weldments in steel construction, mechanical engineering and the fabrication of apparatus and vessels. Besides good weldability they are distinguished by good toughness behaviour in the residual welding stress range. Through fixing the annealing limit at 30 mm initially (today 38 or 50 mm as the case may be) and dimensioning according to the yield strength, development during the course of recent years has led to the high-strength hardened and tempered grades. Finally the gap between 400 and 500 N/nm2 yield strength was closed by the microalloyed steels, some of them air-hardening.Today a large number of grades are available also for hot or cold-working components, throughout the strength range. In view of their various merits, the designer does not always find it easy to make the correct choice. Despite extensive literature, setbacks are repeatedly sustained. Often insufficient allowance is made for the properties of the material in the design, fabrication, inspection and trials. Pattern techniques in the semiconductor industry are driven by the need to improve cost/performance per bit. The key to reducing
cost per bit and improving performance is increased density. Tighter photolithography ground rules (smaller images with reduced
line width and registration tolerances) are primary sources of increased density. Enhanced ground rules coupled with larger
wafers are certain to form the basis for the next generation of VLSI chips. The trend toward tighter ground rules and larger
wafers is causing many semiconductor manufacturers to migrate away from IX projection tools, the workhorses of present-day
production lines, to step and repeat exposure tools. The higher numerical aperture of step and repeat tools, coupled with
chip-by-chip focus, guarantees improved resolution compared to today’s IX projection tools. The price paid for increased numerical
aperture is reduced field size and, subsequently, reduced throughput and higher exposure cost per wafer. Ideally, it would
be preferable to improve the resolution of the IX projection tool/process to make it competitive with the step and repeat
tool, while maintaining the high throughput of IX systems. Effective use of shorter exposure (< 350 nm) wavelengths in IX

A new method is presented for extracting statistical potentials dependent on the relative side chain and backbone orientations in proteins. Coarse-grained, anisotropic potentials are constructed for short-, medium-, and long-range interactions using the Boltzmann method and a database of non-homologous protein structures. The new orientation-dependent potentials are analyzed using a spherical harmonics decomposition method with real eigenfunctions. This method permits a more realistic, continuous angular representation of the coarse-grained potentials. Results of tests for discriminating the native protein conformations from large sets of decoy proteins, show that the new continuous distance- and orientation-dependent potentials present significantly improved performance. Novel graphical representations are developed and used to depict the orientational dependence of the interaction potentials. These new continuous anisotropic statistical potentials could be instrumental in developing new computational methods for structure prediction, threading and coarse-grained simulations. 相似文献
