BACKGROUND: Alveolar macrophages are thought to play an important part in regulating lung immune responses. While it is clear that human alveolar macrophages suppress T cell proliferation in vitro, the mechanisms by which this is achieved are not clear, nor is it known whether alveolar macrophages also inhibit other aspects of T cell function. METHODS: Peripheral blood mononuclear cells were stimulated with phytohaemagglutinin or house dust mite allergen, and cultured with variable numbers of autologous alveolar macrophages obtained by bronchoalveolar lavage from 20 normal subjects. RESULTS: Alveolar macrophages induced a reversible inhibition of T cell proliferation in response to both mitogen and allergen stimulation, with the latter being considerably more susceptible to inhibition. This was achieved via heterogenous mechanisms, involving both soluble factors derived from alveolar macrophages and cell-cell contact. Despite inhibiting proliferation, alveolar macrophages had little or no effect on T cell calcium flux, the characteristic changes in CD3, CD2, CD28 and interleukin-2 (IL-2) receptor expression which accompany normal T cell activation, and IL-2 and interferon gamma secretion. In contrast, alveolar macrophages inhibited the tyrosine phosphorylation of proteins which may be involved in IL-2 receptor-associated signal transduction. CONCLUSIONS: The immunoregulatory properties of alveolar macrophages are relatively selective, allowing T cell activation and cytokine secretion while inhibiting T cell proliferation within the lung.  相似文献

We investigated the effect of the retinal pigmented epithelium on cell proliferation and differentiation in rosetted retinospheroids, which are retina-like spheres reaggregated in the complete absence of retinal pigmented epithelium from dissociated retinal cells of 6-day-old chick embryos in a rotation culture system. In spheroids raised in the absence of retinal pigmented epithelium (controls), acetylcholinesterase was expressed in cells of an inner nuclear-like layer and their neuropil matrices. Moreover, the ratio between rods and cones was found to be approximately normal throughout the spheroid. When spheroids were cultured in the presence of retinal pigmented epithelium monolayers, cell proliferation in spheroids as determined by BrdU labelling was significantly increased and extended for 1 week, while acetylcholinesterase protein levels and specific activities in homogenates were decreased to approximately 30%. At the same time, opsin immunoreactivity was completely suppressed within the spheroid and appeared slowly in cells around its periphery; i.e. the proportion of rhodopsin-positive cells decreased from 14 to 3%. This study reveals that the retinal pigmented epithelium in vitro sustains cell proliferation but inhibits the differentiation of acetylcholinesterase-positive cells and of photoreceptors.  相似文献

Work-related low-back disorders (LBDs) continue to be one of the single largest sources of compensation costs. The relative contributions of personal, workplace, organizational, and environmental variables to the development and severity of LBDs are not completely understood. The inclusion of personal variables in epidemiologic studies of LBDs has been inconsistent, and different authors have different opinions concerning the importance of such variables. Personal variables either known or suspected to influence outcomes are discussed to elucidate the importance of these variables with respect to understanding LBDs and conducting epidemiological studies in industry. The authors suggest that age, gender, injury history, relative strength, smoking, and psychosocial variables be studied further, and that height, weight, pathologies, genetic factors, maximum oxygen uptake, and absolute strength are unlikely to produce significant effects in industrial populations.  相似文献

Curvature and Bending Moments from Inclinometer Data   总被引:1,自引:0,他引:1  
Bending moments in excavation support walls and deep foundations are frequently estimated using curvature derived from inclinometer surveys. The basic idea involves fitting a curve or a series of curves to the inclinometer data. The curvature is then estimated as the second difference of the displacement profile. Although many methods are available to estimate curvature, there is a lack of a consistent standard in deducing curvature from inclinometer readings. Twelve methods for estimating curvature that lend themselves well to spreadsheet or programmable calculations are reviewed. They are then applied to 60 sets of inclinometer readings obtained from a variety of walls and drilled shafts. Included are six laterally load tested drilled shafts which were monitored using both inclinometers and strain gauges. A direct comparison of curvatures from inclinometers with those from strain gauges was performed. The comparisons show that a piecewise cubic polynomial curve fitting a moving window of five successive inclinometer data points provide curvature values that are in good agreement with those from strain gauges, and is proposed as a reasonable method for estimating bending moments from inclinometer data.  相似文献

The new method for surgical treatment of benign tumors and dysplastic processes in the facial skull bones includes resection of the involved segment of the bone, its mechanical treatment, extracorporeal freezing, sterilization, and replantation. Biophysical studies revealed that 10-min freezing of the preparation in liquid nitrogen followed by slow defrosting kills tumor cells. Animal experiments demonstrated excellent osteoplastic properties of thus treated replants. Based on the results of treatment of 29 patients, the authors defined the indications for such treatment and demonstrated its high efficacy in benign tumors and dysplastic processes in the facial skull.  相似文献

Compared to normal volunteers, coronary vasodilation reserve is reduced in patients with hypertrophic cardiomyopathy but not in rowing athletes with left ventricular hypertrophy. The paper presents several mnemonics to assist clinicians in recalling DSM-IV diagnostic criteria for personality disorders. The mnemonics are acronyms, and each letter is associated with a specific criterion. Each acronym reflects a facet of the related disorder; for example, the acronym for the diagnostic criteria for paranoid personality disorder is SUSPECT, and for histrionic personality disorder it is PRAISE ME. The mnemonics have been used to teach students and residents the conceptual nature of DSM-IV disorders and to help them remember the criteria.  相似文献
