Two experiments were conducted to test the efficacy of a novel infectious bursal disease virus (IBDV) vaccine in broiler chickens with maternal IBDV immunity. The IBDV vaccine was formulated by mixing IBDV strain 2512 with bursal disease antibodies (BDA) to produce the IBDV-BDA complex vaccine. In Expt. I, 1-day-old Cobb x Cobb broiler chickens were vaccinated subcutaneously with either IBDV-BDA or commercial live intermediate IBDV vaccine (vaccine A) or were left unvaccinated. In Expt. 2, the vaccine A group was not included; instead, IBDV strain 2512 was included. Chickens were maintained in isolation houses. On day 28 (Expt. 1) and day 32 (Expt. 2) of age, chickens from each group were challenged with a standard USDA IBDV (STC strain) challenge. Challenged and unchallenged chickens were evaluated for their bursa/body weight ratios and antibody titers 3 days post-challenge. Bursae collected from Expt. 2 were examined histologically to evaluate bursal lesions and confirm gross examination. None of the unvaccinated chickens was protected against the challenge virus as evidenced by the presence of acute bursal lesions (edema/hemorrhage). All chickens receiving the IBDV-BDA complex or the IBDV strain 2512 (Expt. 2) were protected from the challenge virus as evidenced by no acute bursal lesions. Additionally, chickens receiving the IBDV-BDA complex vaccine or the IBDV strain 2512 had antibody titers to IBDV, indication the presence of an active immune response. In Expt. 1, chickens vaccinated with vaccine A and challenged had bursal lesions similar to those observed in the unvaccinated, challenged chickens. These chickens also showed no indication of active immunity against the virus. These results suggest that the 1-day-of-age-administered IBDV-BDA complex vaccine can induce active immunity and protection against a standard IBDV challenge in the face of variable levels of maternal IBDV immunity.  相似文献

The feasibility of on-site primary care services and their use by human immunodeficiency virus HIV-seropositive and seronegative injecting drug users within an outpatient methadone maintenance program are examined. A 16-month prospective study was conducted within an ongoing cohort study of HIV infection at a New York City methadone program with on-site primary care services. The study group consisted of 212 seropositive and 264 seronegative drug injectors. A computerized medical encounter data base, with frequencies of primary care visits and with diagnoses for each visit, was linked to the cohort study data base that contained information on patients' demographic characteristics, serologic status, and CD4+ T-lymphocyte counts. Eighty-one percent of the drug injectors in the study voluntarily used on-site primary care services in the methadone program. Those who were HIV-seropositive made more frequent visits than those who were seronegative (mean annual visits 8.6 versus 4.1, P < .001), which increased with declining CD4+ T-lymphocyte counts; 79 percent of those who were seropositive with CD4 counts of less than 200 cells per cubic millimeter received on-site zidovudine therapy or prophylaxis against Pneumocystis carinii pneumonia, or both. Common primary care diagnoses for patients seropositive for HIV included not only conditions specific to the human immunodeficiency virus but also bacterial pneumonia, tuberculosis, genitourinary infections, asthma, dermatologic disease, psychiatric illness, and complications of substance abuse; those who were seronegative were most frequently seen for upper respiratory infection, psychiatric illness, complications of substance abuse, musculoskeletal disease, hypertension, asthma, and diabetes mellitus. Vaginitis and cervicitis,other gynecologic diseases, and pregnancy were frequent primary care diagnoses among both seropositive and seronegative women.  相似文献

BACKGROUND: There is some evidence to suggest that dysfunction in the sensory system of the neck may result in a gamut of signs and symptoms. However, a sound understanding of the somatosensory system in the neck and its normal influence on the central nervous system is essential before signs and symptoms can be identified as representations of ill health or disease arising from the neck. OBJECTIVE: To briefly review current knowledge of the somatosensory system of the neck and to consider its connections and influence on the central nervous system. DATA SOURCES: Information was obtained from peer-reviewed scientific journals and proceedings of scientific meetings that have investigated or considered anatomical and physiological aspects of the sensory system in the necks of human and nonhuman vertebrates. CONCLUSION: Studies involving human and nonhuman vertebrates have provided considerable information about the anatomy of the sensory receptors located in the neck and about where information from these receptors is relayed in the spinal cord and brain. Physiological experiments involving electrical and natural stimulation of the head and neck regions have identified a role for some of these receptors in neck-evoked reflexes. It is clear that in addition to signaling nociception, the somatosensory system of the neck may influence the motor control of the neck, eyes, limbs, respiratory muscles and possibly the activity of some preganglionic sympathetic nerves.  相似文献

The receptor for the vitamin, folic acid, is overexpressed on a number of human tumors, including cancers of the ovary, kidney, uterus, testis, brain, colon, lung, and myelocytic blood cells. Conjugates of folic acid linked via its gamma-carboxyl to either a single drug molecule or assembly of molecules can bind to and enter receptor-expressing cancer cells via folate receptor-mediated endocytosis. Because the affinity of folate conjugates for cell surface folate receptors is high (KD approximately 10(-10) M), folic acid derivatization allows the selective delivery of diagnostic and therapeutic agents to cancer cells in the presence of normal cells. This review will summarize studies aimed at folate-mediated targeting of protein toxins, imaging agents, antisense oligodeoxynucleotides, genes, and liposomes specifically to cancer cells in vitro and in vivo.  相似文献

The interaction of topoisomerase II with its DNA cleavage site is critical to the physiological functions of the enzyme. Despite this importance, the specific enzyme-DNA interactions that drive topoisomerase II-mediated DNA cleavage and religation are poorly understood. Therefore, to dissect interactions between the enzyme and its cleavage site, abasic DNA lesions were incorporated into a bilaterally symmetrical and identical cleavage site. Results indicate that topoisomerase II has unique interactions with each position of the 4-base overhang generated by enzyme-mediated DNA cleavage. Lesions located 2 bases 3' to the point of scission stimulated cleavage the most, whereas those 3 bases from the point of scission stimulated cleavage the least. Moreover, an additive and in some cases synergistic cleavage enhancement was observed in oligonucleotides that contained multiple DNA lesions, with levels reaching >60-fold higher than the wild-type substrate. Finally, topoisomerase II efficiently cleaved and religated a DNA substrate in which apyrimidinic sites were simultaneously incorporated at every position on one strand of the 4-base overhang. Therefore, unlike classical DNA ligases in which base pairing is the driving force behind closure of the DNA break, it appears that for topoisomerase II, the enzyme is responsible for the spatial orientation of the DNA termini for ligation.  相似文献

This research presents experimental results of an investigation aimed at understanding grain size driven mechanical processes in electrodeposited Ni thin films where saccharine additions are commonly used to improve mechanical properties. Ni films were fabricated using salfamate-based electro chemical baths, where it is empirically known that mmol/l concentrations of saccharine will reduce the observed tensile stress in addition to lowering the grain size up to a few nanometer scales. Some previous observations and several theoretical models suggest that saccharine incorporation results in sulfur segregation at grain boundaries. Since grain boundary formation is also associated with tensile stress evolution, a plausible hypothesis is that saccharine additions are directly altering grain boundary energetics. This suggests that saccharine additions should also have an observable effect on intergranular fracture in these films. To test this prediction, in situ stress measurements during film growth and fracture testing of these same films were compared. Lithographically patterned substrates were used to produce films with ordered arrays of uniform islands, which demonstrated island size effects on stress evolution, and enabled a well-defined notch geometry along one of the island boundaries to facilitate fracture experiments. In situ uniaxial tensile testing under in a scanning electron microscope was then used to obtain the fracture strength of such specimens. This technique provided real time recording of microscopic deformation during uniaxial tensile loading. The observed relationships among residual stress, grain size, and fracture strength were then analyzed with detailed models of both film growth and fracture.  相似文献

We have developed a simple method for synthesis of spherical gold nanoparticles (AuNPs) with enhanced surface properties. The polyethyleneimine (PEI) has good potential to minimise the size of the precursor. The UV–vis spectra of synthesised AuNPs with reducing agents (PEI) have been characterised with a peak at 530?nm. The size and shape measurement of AuNPs was confirmed by transmission electron microscopy (TEM) which shows that the mean diameter is 3.9?nm. The optimal concentration of reducing agents was found to be 1% for synthesis of AuNPs. PEI-conjugated AuNP shows binding with arsenic III (0.1?ppm) as confirmed by scanning electron microscope (SEM)/energy dispersive X-rays mapping. TEM revealed the particle shape and size. Zeta potential, zeta deviation, effective particle size, Z-average diameter, polydispersity index and electrophoretic mobility have been observed in order to understand the stability of AuNPs. The image of SEM confirmed that As (III) particles were eventually distributed in PEI-conjugated AuNPs matrix. Further, this study demonstrated that PEI-conjugated AuNPs is a sensing platform of As (III).  相似文献

Streptomyces brunneofungs 118 and S.griseolus 224 were isolated from natural objects and shown to synthesize ammonium specific products belonging to macrotetrolide compounds. Gradient extraction was applied to the mycelium and it was demonstrated that the compounds were rather labile both in the native cells and on synthetic carriers and could be hydrolyzed by aqueous solutions of acetone and ethanol to various linear oligomers of narctinic acids. Acetone mainly stabilized the monomer and dimer fragments whereas in the ethanol extracts a complete set of the oligomers (from the monomer to the tetramer) was detectable. Graident extract of suspension of the microbial intact cells is useful in the study of some properties and the primary identification of biologically active hydrophobic products even at the early stages of their isolation.  相似文献

BACKGROUND: Studies in nonhuman primates provide evidence that intact spatial working memory depends on the integrity of specific areas in the prefrontal cortex. Patients with schizophrenia have been shown to be impaired on spatial working memory tasks. Relatives of schizophrenic patients show a range of cognitive deficits in the absence of clinical symptoms (eg, thought disorder, eye tracking dysfunctions). We predicted that a significant proportion of relatives of schizophrenic patients would show deficits in working memory as measured by a delayed response task. METHODS: In experiment 1, we tested 18 schizophrenic patients, 15 first-degree relatives of schizophrenic patients, and 18 normal control subjects on an oculomotor delayed response task. In experiment 2, we assessed the performance of another group of 12 first-degree relatives of schizophrenic patients and 16 different normal control subjects on a visual-manual delayed response task. RESULTS: Relatives of schizophrenic patients showed significant deficits in working memory on both the oculomotor and visual-manual delayed response tasks. CONCLUSIONS: Some relatives of schizophrenic patients are impaired on tasks that tap spatial working memory and that implicate the prefrontal system. The delayed response paradigm may be useful in elucidating the multidimensionality of the schizophrenic phenotype.  相似文献
