Flow cytometry assays, which measure CD69 activation and intracellular cytokine production, have been used to measure peripheral blood lymphocyte (PBL) responses to in vitro antigen exposure. In the present study, we show that, in healthy individuals and immunosuppressed kidney transplant recipients, CD69 expression and intracellular cytokine production by peripheral blood T cells compare favorably to thymidine uptake as a measure of PBL response to alloantigen in mixed leukocyte culture (MLC). Heparinized whole blood from 23 healthy individuals was incubated for 24-48 h with 3rd party allogeneic monocytes; blood from twelve kidney transplant recipients was incubated with monocytes from their kidney donor and with monocytes from unrelated individuals. The percentage of T cells expressing surface CD69 or intracellular IL-2 or IL-4 was determined by 3-color flow cytometry. We identified 5 donor-specific response patterns in our kidney transplant group. One transplant recipient was hyporesponsive; his cells did not express CD69 or produce IL-2 in response to either donor or 3rd party allogeneic cells. All other transplant recipients expressed CD69 and IL-2 in response to 3rd party allogeneic cells. Two had no response to donor cells (donor-specific hyporesponsiveness), three had donor-specific anergy (CD69 expression without cytokine production in response to donor cells), five had a donor-specific Thl response (CD69 expression and IL-2 production in response to donor cells), and one had a donor-specific Th2 response (CD69 expression and IL-4 but not IL-2 production in response to donor cells). Rapid measures of donor-specific hyporesponsiveness such as CD69 activation antigen expression and intracellular cytokine production may prove valuable in monitoring lymphocyte function and aid in the long-term management of kidney transplant recipients.  相似文献

Window opening in passive solar dwellings Discussions of the user influence on the heating energy consumption of buildings are often controversial. A particularly significant issue is the opening of windows. However, concrete data about user behaviour are rarely available, and the actual consequences of open windows on the heating energy consumption are not understood very well. Based on measured data, this paper discusses the duration of additional window opening in passive solar dwellings, typical behaviour patterns, influence factors and the consequences on the heating energy consumption. Window opening during the summer is also considered. The study is based on measurements carried out at the passive solar housing development in Wiesbaden‐Dotzheim [1], [2] during 2001 and 2002.  相似文献

We present an extension of the random walker segmentation to images with uncertain gray values. Such gray-value uncertainty may result from noise or other imaging artifacts or more general from measurement errors in the image acquisition process. The purpose is to quantify the influence of the gray-value uncertainty onto the result when using random walker segmentation. In random walker segmentation, a weighted graph is built from the image, where the edge weights depend on the image gradient between the pixels. For given seed regions, the probability is evaluated for a random walk on this graph starting at a pixel to end in one of the seed regions. Here, we extend this method to images with uncertain gray values. To this end, we consider the pixel values to be random variables (RVs), thus introducing the notion of stochastic images. We end up with stochastic weights for the graph in random walker segmentation and a stochastic partial differential equation (PDE) that has to be solved. We discretize the RVs and the stochastic PDE by the method of generalized polynomial chaos, combining the recent developments in numerical methods for the discretization of stochastic PDEs and an interactive segmentation algorithm. The resulting algorithm allows for the detection of regions where the segmentation result is highly influenced by the uncertain pixel values. Thus, it gives a reliability estimate for the resulting segmentation, and it furthermore allows determining the probability density function of the segmented object volume.  相似文献

Due to their unique morphology‐related properties, yolk@shell materials are promising materials for catalysis, drug delivery, energy conversion, and storage. Despite their proven potential, large‐scale applications are however limited due to demanding synthesis protocols. Overcoming these limitations, a simple soft‐templated approach for the one‐pot synthesis of yolk@shell nanocomposites and in particular of multicore metal nanoparticle@metal oxide nanostructures (MNP@MOx) is introduced. The approach here, as demonstrated for AuNP@ITOTR (ITOTR standing for tin‐rich ITO), relies on polystyrene‐block‐poly(4‐vinylpyridine) (PS‐b‐P4VP) inverse micelles as two compartment nanoreactor templates. While the hydrophilic P4VP core incorporates the hydrophilic metal precursor, the hydrophobic PS corona takes up the hydrophobic metal oxide precursor. As a result, interfacial reactions between the precursors can take place, leading to the formation of yolk@shell structures in solution. Once calcined these micelles yield AuNP@ITOTR nanostructures, composed of multiple 6 nm sized Au NPs strongly anchored onto the inner surface of porous 35 nm sized ITOTR hollow spheres. Although of multicore nature, only limited sintering of the metal nanoparticles is observed at high temperatures (700 °C). In addition, the as‐synthesized yolk@shell structures exhibit high and stable activity toward CO electrooxidation, thus demonstrating the applicability of our approach for the design of functional yolk@shell nanocatalysts.  相似文献

Atomically precise engineering of the position of molecular adsorbates on surfaces of 2D materials is key to their development in applications ranging from catalysis to single‐molecule spintronics. Here, stable room‐temperature templating of individual molecules with localized electronic states on the surface of a locally reactive 2D material, silicene grown on ZrB2, is demonstrated. Using a combination of scanning tunneling microscopy and density functional theory, it is shown that the binding of iron phthalocyanine (FePc) molecules is mediated via the strong chemisorption of the central Fe atom to the sp3‐like dangling bond of Si atoms in the linear silicene domain boundaries. Since the planar Pc ligand couples to the Fe atom mostly through the in‐plane d orbitals, localized electronic states resembling those of the free molecule can be resolved. Furthermore, rotation of the molecule is restrained because of charge rearrangement induced by the bonding. These results highlight how nanoscale changes can induce reactivity in 2D materials, which can provide unique surface interactions for enabling novel forms of guided molecular assembly.  相似文献

A major clinical problem within synthetic, large-scaled scaffolds is the insufficient nutrient supply resulting in inhomogeneous cell proliferation and differentiation. The aim of this study was to analyse pH value, oxygen consumption and migration of human osteoblasts within a 3D tantalum scaffold, clinically used for larger bone defects. After 24 h the oxygen concentration within the scaffold decreased significantly and remained low during incubation. Monitoring of the pH value inside the tantalum scaffold showed a slightly acidification under static culture conditions. However, cell migration within the 3D scaffold was detected. Hence, in clinical application it can be assumed that porous tantalum scaffolds can be settled by osteoblasts under critical oxygen and nutrient supply. In general, monitoring of cell migration, oxygen consumption and acidification can be a suitable instrument for creating advanced 3D bone scaffolds.  相似文献

High‐refresh‐rate displays (e. g., 120 Hz) have recently become available on the consumer market and quickly gain on popularity. One of their aims is to reduce the perceived blur created by moving objects that are tracked by the human eye. However, an improvement is only achieved if the video stream is produced at the same high refresh rate (i. e. 120 Hz). Some devices, such as LCD TVs, solve this problem by converting low‐refresh‐rate content (i. e. 50 Hz PAL) into a higher temporal resolution (i. e. 200 Hz) based on two‐dimensional optical flow. In our approach, we will show how rendered three‐dimensional images produced by recent graphics hardware can be up‐sampled more efficiently resulting in higher quality at the same time. Our algorithm relies on several perceptual findings and preserves the naturalness of the original sequence. A psychophysical study validates our approach and illustrates that temporally up‐sampled video streams are preferred over the standard low‐rate input by the majority of users. We show that our solution improves task performance on high‐refresh‐rate displays.  相似文献

The cambial meristem is responsible for bark and wood formation in woody plants. The activity of the cambial meristem is controlled by various factors; one of them is the plant hormone cytokinin. Here, we have explored different approaches to genetically engineering cambial activity in poplar plants by the ectopic expression of a cytokinin biosynthesis gene with enhanced activity (named ROCK4) or of a gene encoding a constitutively active cytokinin receptor variant (ROCK3). Both genes are derived from Arabidopsis thaliana and were expressed in poplar trees under the control of their own promoter or the cambium-specific pHB8 promoter. pIPT3:ROCK4- and pHB8:ROCK4-expressing plants were smaller than wild-type plants and formed more lateral branches; pHB8:ROCK4 transgenic plants additionally showed an increased stem diameter. In contrast, pAHK3:ROCK3- and pHB8:ROCK3-expressing plants grew taller than wild type without an altered branching pattern and formed more cambial cells, leading to increased radial stem growth. The effectivity of ROCK3 when expressed in either secondary phloem cells or in cambial cells is consistent with a dual, tissue-autonomous and non-autonomous activity of cytokinin in regulating cambial activity. We propose ROCK3 as a novel gene to enhance biomass formation in woody plants.  相似文献
