The role of peroxisomal processes in the maintenance of neurons has not been thoroughly investigated. We propose using Caenorhabditis elegans as a model organism for studying the molecular basis underlying neurodegeneration in certain human peroxisomal disorders, e.g. Zellweger syndrome, since the nematode neural network is well characterized and relatively simple in function. Here we have identified C. elegans PEX-5 (C34C6.6) representing the receptor for peroxisomal targeting signal type 1 (PTS1), defective in patients with such disorders. PEX-5 interacted strongly in a two-hybrid assay with Gal4p-SKL, and a screen using PEX-5 identified interaction partners that were predominantly terminated with PTS1 or its variants. A list of C. elegans proteins with similarities to well-characterized yeast beta-oxidation enzymes was compiled by homology probing. The possible subcellular localization of these orthologues was predicted using an algorithm based on trafficking signals. Examining the C termini of selected nematode proteins for PTS1 function substantiated predictions made regarding the proteins' peroxisomal location. It is concluded that the eukaryotic PEX5-dependent route for importing PTS1-containing proteins into peroxisomes is conserved in nematodes. C. elegans might emerge as an attractive model system for studying the importance of peroxisomes and affiliated processes in neurodegeneration, and also for studying a beta-oxidation process that is potentially compartmentalized in both mitochondria and peroxisomes.  相似文献

An emerging strategy for creating self-healing materials relies on embedded vascular networks of microchannels to transport reactive fluids to regions of damage. Here we investigate the use of active pumping for the pressurized delivery of a two-part healing system, allowing a small vascular system to deliver large volumes of healing agent. Different pumping strategies are explored to improve the mixing and subsequent polymerization of healing agents in the damage zone. Significant improvements in the number of healing cycles and in the overall healing efficiency are achieved compared with prior passive schemes that use only capillary forces for the delivery of healing agents. At the same time, the volume of the vascular system required to achieve this superior healing performance is significantly reduced. In the best case, nearly full recovery of fracture toughness is attained throughout 15 cycles of damage and healing, with a vascular network constituting just 0.1 vol% of the specimen.  相似文献

Ghosh and Maruska have recently reported a sunlight conversion efficiency for Al-doped single crystal TiO2 of 1.3%. In this work we report conversion efficiencies of 1% for A1 and Y-doped, polycrystalline TiO2 electrodes, and 1.1% for B-doped samples. We also discuss the effect of such doping on the electrical resistivity of the resulting oxides, and compare the efficiencies of homogeneous and differential cell configurations using the same photoanodes and counter electrodes.  相似文献

The ITER remote handling (RH) system has been divided into 7 major equipment system procurements that deliver complete systems (operator interfaces, equipment controllers, and equipment) according to task oriented functional specifications. Each equipment system itself is an assembly of transporters, power manipulators, telemanipulators, vehicular systems, cameras, and tooling with a need for controllers and operator interfaces.From an operational perspective, the ITER RH systems are bound together by common control rooms, operations team, and maintenance team; and will need to achieve, to a varying degree, synchronization of operations, co-operation on tasks, hand-over of components, and sharing of data and resources. The separately procured RH systems must, therefore, be integrated to form a unified RH system for operation from the RH control rooms.The RH system will contain a heterogeneous mix of specially developed RH systems and off-the-shelf RH equipment and parts. The ITER Organization approach is to define a control system architecture that supports interoperable heterogeneous modules, and to specify a standard set of modules for each system to implement within this architecture. Compatibility with standard parts for selected modules is required to limit the complexity for operations and maintenance. A key requirement for integrating the control system modules is interoperability, and no module should have dependencies on the implementation details of other modules.The RH system is one of the ITER Plant systems that are integrated and coordinated through the hierarchical structure of the ITER CODAC system. It is distinguished from other Plant systems by the man-in-the-loop nature of RH operations and the need for control rooms at a level below the main control room. The RH control system architecture has been designed to also support the central monitoring and coordination of the RH activities.  相似文献

We report a simple model describing the solubility of rods in solvents, expressing the final result explicitly in terms of the surface entropy and the enthalpy of mixing. This model can be combined with any expression for the mixing enthalpy depending on the requirements. For example, in one instance it predicts the dispersed concentration of rods to decrease exponentially with the Flory-Huggins parameter of the dispersion. Using a different enthalpy function, it predicts a Gaussian peak when concentration is plotted versus solvent surface energy. The model also suggests specific solvent-rod interactions to be important and shows the dispersed concentration to be very sensitive to ordering at the solvent-rod interface. We have used this model to describe experimental results for the concentration of dispersed nanotubes in various solvents. Qualitative agreement with these predictions is observed experimentally. However, we suggest that the fact that quantitative agreement is not found may be explained by solvent ordering at the nanotube surface.  相似文献

Integrated perovskite/organic bulk heterojunction (BHJ) solar cells have the potential to enhance the efficiency of perovskite solar cells by a simple one‐step deposition of an organic BHJ blend photoactive layer on top of the perovskite absorber. It is found that inverted structure integrated solar cells show significantly increased short‐circuit current (Jsc) gained from the complementary absorption of the organic BHJ layer compared to the reference perovskite‐only devices. However, this increase in Jsc is not directly reflected as an increase in power conversion efficiency of the devices due to a loss of fill factor. Herein, the origin of this efficiency loss is investigated. It is found that a significant energetic barrier (≈250 meV) exists at the perovskite/organic BHJ interface. This interfacial barrier prevents efficient transport of photogenerated charge carriers (holes) from the BHJ layer to the perovskite layer, leading to charge accumulation at the perovskite/BHJ interface. Such accumulation is found to cause undesirable recombination of charge carriers, lowering surface photovoltage of the photoactive layers and device efficiency via fill factor loss. The results highlight a critical role of the interfacial energetics in such integrated cells and provide useful guidelines for photoactive materials (both perovskite and organic semiconductors) required for high‐performance devices.  相似文献

Thermal regime is strongly associated with hydrodynamics in water, and it plays an important role in the dynamics of water quality and ecosystem succession of stratified reservoirs. Changes in both climate and hydrological conditions can modify thermal regimes. Liuxihe Reservoir (23°45′50″N; 113°46′52″E) is a large, stratified and deep reservoir in Guangdong Province, located at the Tropic of Cancer of southern China. The reservoir is a warm monomictic water body with a long period of summer stratification and a short period of mixing in winter. The vertical distribution of suspended particulate material and nutrients are influenced strongly by the thermal structure and the associated flow fields. The hypolimnion becomes anoxic in the stratified period, increasing the release of nutrients from the bottom sediments. Fifty-one years of climate and reservoir operational observations are used here to show the marked changes in local climate and reservoir operational schemes. The data show increasing air temperature and more violent oscillations in inflow volumes in the last decade, while the inter-annual water level fluctuations tend to be more moderate. To quantify the effects of changes in climate and hydrological conditions on thermal structure, we used a numerical simulation model to create scenarios incorporating different air temperatures, inflow volumes, and water levels. The simulations indicate that water column stability, the duration of the mixing period, and surface and outflow temperatures are influenced by both natural factors and by anthropogenic factors such as climate change and reservoir operation schemes. Under continuous warming and more stable storage in recent years, the simulations indicate greater water column stability and increased duration of stratification, while irregular large discharge events may reduce stability and lead to early mixing in autumn. Our results strongly suggest that more attention should be focused on water quality in years of extreme climate variation and hydrological conditions, and selective withdrawal of deep water may provide an efficient means to reduce internal loading in warm years.  相似文献

In many environmental contaminant situations selenium has become the primary element of concern because of its bioaccumulative nature in food webs. Initial concerns about selenium were related to fish kills at Belews Lake, NC, Martin Lake, TX, and Kesterson Reservoir, CA, and to bird deformities at Kesterson Reservoir. Additional concerns were identified under the National Irrigation Water Quality Program at Salton Sea, CA, Kendrick, WY, Stewart Lake, UT, and Grand Valley and Uncompahgre Valley, CO. Recent studies have raised concerns about selenium impacts on aquatic resources in Southeastern Idaho and British Columbia. The growing discomfort among the scientific community with a waterborne criterion has lead the US Environment Protection Agency to consider a tissue-based criterion for selenium. Some aquatic ecosystems have been slow to recover from selenium contamination episodes. In recent years, non-governmental researchers have been proposing relatively high selenium thresholds in diet and tissue relative to those proposed by governmental researchers. This difference in opinions is due in part to the selection of datasets and caveats in selecting scientific literature. In spite of the growing selenium literature, there are needs for additional research on neglected organisms. This review also discusses the interaction of selenium with other elements, inconsistent effects of selenium on survival and growth of fish, and differences in depuration rates and sensitivity among species.  相似文献

Benefit/cost methodologies are well accepted for project analysis from a national perspective. Recent pressure to develop local decisionmaking and local funding for water projects has focused attention on the regional costs and benefits of such development. This paper examines some methodological issues that emerge as one attempts to estimate these regional costs and benefits. Uncritical use of regional economics procedures and concepts such as input-output analysis and value added to measure the primary and secondary impacts of these projects carries the risk of seriously overestimating regional project benefits. The paper concludes that new approaches to regional project analysis must be developed that will reconcile and incorporate regional economics procedures with benefit/cost methodology.  相似文献
