Species adsorbed at the surfaces of soot particles sampled at different locations in a low-pressure methane flame have been analyzed. The analysis method is laser desorption/laser ionization/time-of-flight mass spectrometry (LD/LI/TOF-MS) applied to soot particles deposited on a filter after probe extraction in the flame. In order to fully characterize the experimental apparatus, a strategy of systematic investigations has been adopted, beginning with the study of less complex systems constituted by model soot (standard polycyclic aromatic hydrocarbons, PAHs, adsorbed on black carbon), and then natural soot sampled from a literature reference ethylene flame. This characterization allowed a good understanding of the analytical response of PAHs to the desorption and ionization processes and the definition of the optimal experimental conditions. The soot PAH content was then investigated on a low-pressure methane/oxygen/nitrogen premixed flat flame (? = 2.32) as a function of the sampling height above the burner (HAB). The obtained mass spectra are reproducible, fragment-free, well resolved in the analyzed m/z range and they are characterized by an excellent signal-to-noise ratio. They all feature regular peak sequences, where each signal peak has been assigned to the most stable high-temperature-formed PAHs. The structure of the mass spectra depends on the sampling HAB into the flame, i.e., on the reaction time. An original contribution to the data interpretation comes from the development of a new sampling method that makes it possible to infer hypotheses about the PAH partition between the gas phase and the soot particles. This method highlights the presence of high-mass PAHs in the soot nucleation zone, and it suggests the importance of heterogeneous reactions occurring between flame PAHs and soot particles. 相似文献

Background
The exposure to pollutants such as diesel exhaust particles (DEP) is associated with an increased incidence of respiratory diseases. However, the mechanisms by which DEP have an effect on human health are not completely understood. In addition to their action on macrophages and airway epithelial cells, DEP also modulate the functions of dendritic cells (DC). These professional antigen-presenting cells are able to discriminate unmodified self from non-self thanks to pattern recognition receptors such as the Toll like Receptors (TLR) and Scavenger Receptors (SR). SR were originally identified by their ability to bind and internalize modified lipoproteins and microorganisms but also particles and TLR agonists. In this study, we assessed the implication of SR in the effects of DEP associated or not with TLR agonists on monocyte-derived DC (MDDC). For this, we studied the regulation of CD36, CXCL16, LOX-1, SR-A1 and SR-B1 expression on MDDC treated with DEP associated or not with TLR2, 3 and 4 ligands. Rapid computation of the QR factorization of a matrix is fundamental to many scientific and engineering problems. Weight gain after smoking cessation was studied in a naturalistic setting where (1) all smokers quit and (2) risk factors for postcessation weight gain were modified. Participants were 332 military recruits (227 men, 105 women), 86 of whom were smokers who quit during 6 weeks of basic training. Results showed no significant weight changes for smokers who quit. Pretest smoking rates and feat of weight gain were unrelated to changes in weight. Results suggest that an intensive program that limits access to alcohol and foods that are high in fat and that increases physical activity can attenuate weight gain after smoking cessation. The objective of this study was to determine i) if Camembert cheese micro-organisms could be detected in fecal samples after regular consumption by human subjects and ii) the consequence of this consumption on global metabolic activities of the host colonic microbiota. An open human protocol was designed where 12 healthy volunteers were included: a 2-week period of fermented products exclusion followed by a 4-weeks Camembert ingestion period where 2x40 g/day of Camembert cheese was consumed. Stools were collected from the volunteers before consumption, twice during the ingestion period (2nd and 4th week) and once after a wash out period of 2 weeks. During the consumption of Camembert cheese, high levels of Lactococcus lactis and Leuconostoc mesenteroides were measured in fecal samples using real-time quantitative PCR, reaching median values of 8.2 and 7.5 Log(10) genome equivalents/g of stool. For Ln. mesenteroides, persistence was observed 15 days after the end of Camembert consumption. The survival of Geotrichum candidum was also assessed and the fecal concentration reached a median level of 7.1 Log(10) CFU/g in stools. Except a decreasing trend of the nitrate reductase activity, no significant modification was shown in the metabolic activities during this study. 相似文献

The effect of processing on mycotoxin content in milling fractions has been investigated in 10 samples of durum wheat contaminated with T-2 and HT-2 toxins at levels ranging from 97 to 5,954 μg/kg (sum of T-2 and HT-2 toxins). Either naturally contaminated samples or samples artificially inoculated with Fusarium sporotrichioides under field conditions were used. A method based on liquid chromatography-tandem mass spectrometry coupled with immunoaffinity column cleanup was validated in-house for the simultaneous analysis of both toxins in a variety of matrices, including uncleaned wheat, cleaned wheat, screenings, bran, red dog, fine middlings, and semolina. Mean recoveries from samples spiked with T-2 and HT-2 toxins at levels of 100 μg/kg ranged from 85 to 107%, with relative standard deviations (RSDs) lower than 14%. The milling process led to an increase of T-2 and HT-2 toxin contents up to 13- and 5-fold in screenings and bran, respectively, compared with occurrence in the uncleaned wheat; however, an overall reduction of T-2 and HT-2 toxins by 54% (RSD, 20%) and 89% (RSD, 3%) was observed in cleaned wheat and in semolina, respectively. 相似文献
