Abstract : A process for the micropropagation of Amorphophallus rivierii (elephant yam or konjac) and for the extraction and purification of the glucomannan polysaccharide from fresh konjac plant corms has been developed. This process involves extraction with 2-propanol, which has the additional benefit of extracting carotenoids as a potentially valuable side-product. Starch granules with an unusually high and homogeneous gelatinisation temperature range are normally present in the corms, particularly immature ones, and this greatly reduces the strength of gels formed using the glucomannan. Therefore, the extraction process also involves the selective hydrolysis of the starch, by α- and β-amylases that have been specially selected for an absence of contaminating β-mannanase or β-glucanase activity that would depolymerise the glucomannan and render it nonfunctional. Bacillus lichenformis α-amylase was preferred. Using this process pure glucomannan could be extracted which, when mixed with K -carrageenan, forms a gel almost twice as strong as locust bean gum-K -carrageenan gels of the same concentration.  相似文献

Mycotoxin analysis is usually carried out by high performance liquid chromatography after immunoaffinity column cleanup or in enzyme-linked immunosorbent assay tests. These methods normally involve determination of single compounds only. EU legislation already exists for the aflatoxins, ochratoxin A and patulin in food, and legislation will come into force for deoxynivalenol, zearalenone and the fumonisins in 2007. To enforce the various legal limits, it would be preferable to determine all mycotoxins by routine analysis in different types of matrices in one single extract. This would also be advantageous for HACCP control purposes. For this reason, a multi-method was developed with which 33 mycotoxins in various products could be analysed simultaneously. The mycotoxins were extracted with an acetonitrile/water mixture, diluted with water and then directly injected into a LC–MS/MS system. The mycotoxins were separated by reversed-phase HPLC and detected using an electrospray ionisation interface (ESI) and tandem MS, using MRM in the positive ion mode, to increase specificity for quality control. The following mycotoxins could be analysed in a single 30-min run: Aflatoxins B1, B2, G1 and G2, ochratoxin A, deoxynivalenol, zearalenone, T-2 toxin, HT-2 toxin, α-zearalenol, α-zearalanol, β-zearalanol, sterigmatocystin, cyclopiazonic acid, penicillic acid, fumonisins B1, B2 and B3, diacetoxyscirpenol, 3- and 15-acetyl-deoxynivalenol, zearalanone, ergotamin, ergocornin, ergocristin, α-ergocryptin, citrinin, roquefortin C, fusarenone X, nivalenol, mycophenolic acid, alternariol and alternariol monomethyl ether. The limit of quantification for the aflatoxins and ochratoxin A was 1.0 µg kg?1 and for deoxynivalenol 50 µg kg?1. The quantification limits for the other mycotoxins were in the range 10–200 µg kg?1. The matrix effect and validation data are presented for between 13 and 24 mycotoxins in peanuts, pistachios, wheat, maize, cornflakes, raisins and figs. The method has been compared with the official EU method for the determination of aflatoxins in food and relevant FAPAS rounds. The multi-mycotoxin method has been proven by the detection of more than one mycotoxin in maize, buckwheat, figs and nuts. The LC–MS/MS technique has also been applied to baby food, which is subject to lower limits for aflatoxin B1 and ochratoxin A, ergot alkaloids in naturally contaminated rye and freeze-dried silage samples.  相似文献

Nonlinear sorption by natural organic matter may have a significant impact on the behavior of organic contaminants in soils and sediments. This study presents a molecular probe approach based on linear solvation energy relationships (LSERs) to identify and quantify the molecular interactions causing concentration-dependent sorption and proposes estimation methods for sorption nonlinearities. Sorption isotherms ranging over concentrations of more than 4 orders of magnitude were determined in batch systems for 23 and 16 chemically diverse probe compounds in a lignite sample and a peat soil, respectively. Each sorbent showed characteristic nonlinear sorption with Freundlich exponents (1/n) being 0.7-1. The LSER-based analysis revealed that the strength of nonspecific interactions did not vary with concentration for both sorbents. In lignite, specific interactions did not affect sorption nonlinearity either, suggesting that compound-independent factors of lignite were responsible for the nonlinear sorption. In the peat soil, by contrast, the specific interactions related to the solute polarizability/dipolarity parameter (S) decreased with increasing concentration. Consequently, compounds of higher S values were more susceptible to nonlinear sorption in the peat soil. Phenol probes have shown that hydrogen bond donating properties of sorbate compounds have a substantial impact on the overall strength of sorption with organic matter, but no significant influence on sorption nonlinearity. Heterocyclic aromatic compounds appear to undergo additional interactions that are not accounted for by the LSER. These additional interactions considerably enhance both sorption capacity and nonlinearity.  相似文献

Despite growing concerns about cross-contamination of ready-to-eat foods with Listeria monocytogenes, our knowledge about the ecology and transmission of L. monocytogenes in retail establishments has remained limited. We conducted a cross-sectional study to characterize the prevalence, distribution, and subtype diversity of L. monocytogenes in 120 New York State retail deli establishments that were hypothesized to present an increased risk for environmental L. monocytogenes contamination (i.e., small establishments and establishments with a history of failed New York State Agriculture and Markets inspections). Analysis of these data along with previously reported data for 121 predominantly larger retail establishments in New York State identified establishment size, geographic location, and inspection history as significant predictors of L. monocytogenes presence and prevalence. The odds of an establishment being L. monocytogenes positive were approximately twice as high for large establishments, establishments located in New York City, or establishments with poor inspection history (as compared with establishments without these attributes), even though correlation between location and inspection history complicated interpretation of results. Within an establishment, L. monocytogenes was significantly more prevalent on nonfood contact surfaces than on food contact surfaces; prevalence was particularly high for floors and in floor drains, sinks, the dairy case, and milk crates. L. monocytogenes subtype diversity differed between sites, with lineage I isolates significantly associated with nonfood contact surfaces and lineage II isolates significantly associated with food contact surfaces. Isolates belonging to the same ribotype were often found dispersed across multiple sites within an operation.  相似文献

Lead (Pb) exposure induces severe nephrotoxic effects in humans and animals. Herein, we compare the effects of two chelating agents, salinomycin and deferiprone, on Pb-induced renal alterations in mice and in the homeostasis of essential elements. Adult male mice (Institute of Cancer Research (ICR)) were randomized into four groups: control (Ctrl)—untreated mice administered distilled water for 28 days; Pb-exposed group (Pb)—mice administered orally an average daily dose of 80 mg/kg body weight (BW) lead (II) nitrate (Pb(NO3)2) during the first two weeks of the experimental protocol followed by the administration of distilled water for another two weeks; salinomycin-treated (Pb + Sal) group—Pb-exposed mice, administered an average daily dose of 16 mg/kg BW salinomycin for two weeks; deferiprone-treated (Pb + Def) group—Pb-exposed mice, administered an average daily dose of 20 mg/kg BW deferiprone for 14 days. The exposure of mice to Pb induced significant accumulation of the toxic metal in the kidneys and elicited inflammation with leukocyte infiltrations near the glomerulus. Biochemical analysis of the sera revealed that Pb significantly altered the renal function markers. Pb-induced renal toxicity was accompanied by a significant decrease in the endogenous renal concentrations of phosphorous (P), calcium (Ca), copper (Cu) and selenium (Se). In contrast to deferiprone, salinomycin significantly improved renal morphology in Pb-treated mice and decreased the Pb content by 13.62% compared to the Pb-exposed group. There was also a mild decrease in the renal endogenous concentration of magnesium (Mg) and elevation of the renal concentration of iron (Fe) in the salinomycin-treated group compared to controls. Overall, the results demonstrated that salinomycin is a more effective chelating agent for the treatment of Pb-induced alterations in renal morphology compared to deferiprone.  相似文献

This work reports on the features of a sorption processes for the ultimate removal and recovery of reactive phosphorus from aquaculture and poultry production wastewater effluents. The sorbent used was a cross-linked polyamine (PAA-HCl) polymeric hydrogel. The PAA-HCl hydrogels were prepared by chemically cross-linking aqueous solutions of linear PAA-HCl chains with epichlorohydrin (EPI). The phosphorus binding capacity of the gels was measured in standard aqueous solutions as a function of ionic strength. Equilibrium PO4(3-), loadings of 100 mg anion/g gel were obtained. The regeneration ability of the gels was demonstrated by release of the bound phosphorus anions upon washing with 1-2 M NaOH solution, providing opportunities to recover and reuse the gel over multiple cycles. The ionic polyamine gels have been demonstrated to be appropriate materials for treating poultry and aquaculture wastewater effluents. Upon treatment phosphorus anion concentrations were reduced to levels suitable for discharge into natural surface waters.  相似文献

The paper presents design and verification of regression models for prediction of pipe conveyor belt contact forces on idler rolls. Their advantage is in the fact that they have been created according to the real experimental measurement. The obtained models correspond with real operational conditions. The concept of the experimental rig at the Technical University of Košice is designed so that it represents 8 m long section of pipe conveyor. The conveyor belt is an object with action of tension force and at the same time it takes the initiative in the contact force formation. Several criteria have been used to verify the presented regression models. The tensioning force 9000, 12,000 and 15,000 N have MPE of prediction error less than 5%. About 120,000 measured values have been processed during the evaluations which correspond to approximately 15,000 results from experimental measurements.  相似文献

Risk assessments related to use of water and safety of fresh produce originate from both water and food microbiology studies. Although the set‐up and methodology of risk assessment in these 2 disciplines may differ, analysis of the current literature reveals some common outcomes. Most of these studies from the water perspective focus on enteric virus risks, largely because of their anticipated high concentrations in untreated wastewater and their resistance to common wastewater treatments. Risk assessment studies from the food perspective, instead, focus mainly on bacterial pathogens such as Salmonella and pathogenic Escherichia coli. Few site‐specific data points were available for most of these microbial risk assessments, meaning that many assumptions were necessary, which are repeated in many studies. Specific parameters lacking hard data included rates of pathogen transfer from irrigation water to crops, pathogen penetration, and survival in or on food crops. Data on these factors have been investigated over the last decade and this should improve the reliability of future microbial risk estimates. However, the sheer number of different foodstuffs and pathogens, combined with water sources and irrigation practices, means that developing risk models that can span the breadth of fresh produce safety will be a considerable challenge. The new approach using microbial risk assessment is objective and evidence‐based and leads to more flexibility and enables more tailored risk management practices and guidelines. Drawbacks are, however, capacity and knowledge to perform the microbial risk assessment and the need for data and preferably data of the specific region.  相似文献

Kuka is an important vegetable in the Nigerian food chain. The moisture sorption characteristics of the vegetable studied at 34, 37 and 45°C between aw 0.10–0.96, revealed a BET type II behaviour. The Kuka exhibited hysteresis and an increase in EMC with increase in aw and decrease in temperature. Four sorption models (Oswin, Halsey, Kuhn, and GAB) were studied and the Oswin model was the most suitable. The constants in the Oswin model and GAB monolayer moisture contents were obtained and found to be temperature-dependent. Heats of sorption were greater for desorption than adsorption and in either mode, they reduced with an increase in moisture content. An exponential equation was obtained to relate heat of sorption with moisture content.  相似文献

The underlying basis for the behaviour of a population of cells is the cell growth and division cycles of the individual cells. Under certain special circumstances, oscillations in cell cultures can be observed that are a reflection of metabolic oscillations, such as in glycolysis, or cell cycle oscillations from a partially synchronized population. These oscillations can sometimes occur spontaneously, as in the chemostat culture of Saccharomyces cerevisiae at low dilution rates, or can be induced using chemical blockers or entrainment by a periodic nutrient environment. Continuous methods for inducing a synchronized culture include the pulsed chemostat, continuous phasing and self-cycling fermentation. Results obtained by studying both spontaneously oscillating cultures and synchronized cultures have provided insight into the mechanisms responsible for the progress and control of the cell cycle as well as the intimate relationship that exists between the cellular dynamics and the dynamics of the environment.  相似文献
