Fourier transform infrared (FTIR) spectrometry has been evaluated as a method for determining the sulfate concentration of ambient aerosol particulate samples. Samples were collected on Teflon filters. The filters were analyzed for sulfate by both FTIR and ion chromatography (IC). There is good agreement between sulfate analysis by IC and analysis of the FTIR transmission spectra of the air filters during the first 5 1/2 days of the Carbonaceous Species Methods Intercomparison Study in which the ambient sulfate concentrations were above the 5.8 μg/cm2 lower limit of detection of the FTIR technique. A method to improve the FTIR lower limit of detection is discussed. The difficulties incurred during background subtraction of the infrared spectra are described. 相似文献

Internationally, pressure is being exerted on governments and regulators to develop modern forms of regulation that deliver more for less, and in better ways. We discuss the ways in which one large regulator, the Environment Agency for England and Wales, has responded to such pressures by implementing risk-based approaches to regulation. After exploring the context for key elements of risk-based environmental regulation, we consider the evolving influence of such approaches. We discuss the impacts against the UK Government's principles for better regulation and the key criteria for policy evaluation, before considering some of the challenges: the need to i) understand the best practice and promote consistency in risk-based regulation; ii) develop reliable, responsive forms of risk assessment and monitoring; iii) build capacities for responsive risk regulation; iv) evaluate the influence of different regulatory styles; and v) better understand the potential role of the private sector. 相似文献

This paper examines relationships between transmembrane potential (Vm), [Ca2+]i dependent membrane ionic currents, and [Ca2+]i handling by the sarcoplasmic reticulum (SR) in a two-dimensional model of cardiac tissue. Luo-Rudy dynamic (LRd) membrane equations were used because they include detailed formulations for triggered SR Ca2+ release dependent on membrane Ca2+ influx (CICR) and for spontaneous SR Ca2+ release following calsequestrin buffer overload (SCR). Reentry's rapid rate (110-ms cycle length) elevated [Ca2+]i and limited CICR, which in turn promoted SCR that occurred at intervals of 320-350 ms, was preferential at sites located inside the functional center, and destabilized the reentrant activation sequence. Although adjustment of LRd parameters for SR Ca2+ modified SCR interval and peak [Ca2+]i in voltage clamp simulations with a command waveform representing Vm time course within the functional center, SCR persisted. Using the same command waveform, SCR also occurred with an alternate SR Ca2+ formulation that represented subcellular details underlying CICR. LRd parameter adjustments to promote CICR and limit SCR in subsequent reentry simulations failed to eliminate SCR completely, as they modulated SCR intervals in a manner consistent with the voltage clamp simulations. Taken together, our findings support a destabilizing influence of functional reentry on [Ca2+]i handling. However, [Ca2+]i instabilities did not always fractionate depolarization wavefronts during reentry. A combination of immuno-electron microscopy and electron energy-loss spectrum-imaging was used to map the distributions of endocrine polypeptide hormones and proteins in mouse pancreatic islet of Langerhans. Tissue was analyzed from control animals and from mice that were heterozygous for the Anx7 gene, which defines a Ca2+/GTP-dependent membrane fusion and ion channel protein. The heterozygous Anx7 (+/-) mouse displays defects in IP3 receptor mediated Ca2+ signaling and insulin secretion. Therefore, information was obtained about the distributions of the hormones insulin and glucagon, as well as the proteins ANX7 and the IP3 receptor. Insulin secretion appears to be defective in the mutants. It was found from immunolabeling experiments that expression of the IP3 receptor is reduced in mutant islets compared to control islets. Subcellular distributions of sulfur and nitrogen obtained by electron energy-loss spectrum-imaging showed that the insulin concentrations of beta granules were essentially the same in control and mutant islets. By contrast, immunogold labeling of mutant islets shows more insulin immunoreactivity in the beta granules. It follows that insulin may be packaged differently in mutant islets, making antigenic determinants more available to the labeling antibody. The increased rate of insulin secretion in the hyperplastic mutant islets can be explained by compensatory increases in islet size, rather than by an increased insulin concentration in the beta cells. The results indicate that reduced ANX7 expression leads to defects in the IP3 receptor expression in the endocrine cells of the mutant mouse. Increased size of the islet or of adrenal medulla may be a compensatory mechanism for secretion defect by individual endocrine cells. Defects in IP3 receptor expression, and documented consequences of a Ca2+ signaling defect, lead to other changes in organelles such as the mitochondrial number in islet beta-cells. We present a generalized exposure assessment of 28 disposal options for poultry carcasses in the event of a highly pathogenic avian influenza (HPAI) outbreak. The analysis supports a hereto unverified disposal hierarchy for animal carcasses, placing waste processing (e.g., incineration and rendering) above controlled disposal (e.g., landfill), above uncontrolled disposal (e.g., burial on-farm). We illustrate that early stages of the disposal chain (on-farm) pose greater opportunities for exposure to hazardous agents than later stages, where agents are generally contained, wastes are treated, and residues are managed by regulated processes. In selecting carcass disposal options, practitioners are advised to consider the full range of hazards rather than focusing solely on the HPAI agent, and to give preference to technologies that (i) offer high destruction efficiencies for target pathogens; (ii) do not give rise to significant releases of other pathogenic organisms; and (iii) do not release unacceptable concentrations of toxic chemicals. The approach offers an exposure assessment perspective for carcass disposal, thus providing a risk-informed basis for contingency planning and operational intervention. Spacing of edge fractures in layered materials 总被引:1,自引:0,他引:1
Opening-mode fractures developed from a free surface in a layered material often terminate at the interface that divides the fractured layer and the underlying layer. They also display regular spacing that is of the same order of magnitude as the thickness of the fractured layer. We have investigated the stress distribution between two adjacent edge fractures as a function of the ratio of fracture spacing to thickness of the fractured layer using a two-layer elastic model with a fractured top layer. The results show that when the ratio of fracture spacing to the layer thickness changes from greater than to less than a critical value the normal stress acting perpendicular to the fractures near the free surface changes from tensile to compressive. This stress state transition precludes further infilling of fractures unless they are driven by mechanisms other than a pure extension, or there are flaws that significantly perturb the local stress field between the fractures. Hence, the critical fracture spacing to layer thickness ratio defines a lower limit for fractures driven by extension, which also defines the condition of fracture saturation. The critical value of the fracture spacing to layer thickness ratio is independent of the average strain of the fractured layer, and it increases with increasing ratio of Young's modulus of the fractured layer to that of the underlying layer. The critical value increases with increasing Poisson's ratio of the fractured layer, but it decreases with increasing Poisson's ratio of the underlying layer. For the case with the same elastic constants for the fractured layer and the underlying layer, the critical spacing to layer thickness ratio is about 3.1. Delamination between the fractured layer and the underlying layer makes the critical spacing to layer thickness ratio much greater. Infilling fractures grow more easily from flaws located near the bottom of the fractured layer than from those located near the free surface when the spacing to layer thickness ratio is less than the critical value. The propagation of an edge flaw between adjacent edge fractures is unstable, but for the flaw to propagate to the interface, its height has to be greater than a critical size, that decreases with increasing fracture spacing to layer thickness ratio. The propagation behavior of an internal flaw with its lower tip at the interface depends on the edge fracture spacing to layer thickness ratio. The propagation is unstable, when the fracture spacing to layer thickness ratio is greater than a critical value; stable, when the fracture spacing to layer thickness ratio is less than another critical value; and first unstable, then stable, and/or unstable again, when the fracture spacing to layer thickness ratio is between these two critical values. 相似文献
