Arsenic pollution at the industrial site of Reppel-Bocholt (north Belgium)   总被引:1,自引:0,他引:1  
An industrial site, polluted with As and heavy metals, was investigated by combining chemical (sequential extractions and pHstat leaching tests), physical and mineralogical characterization of soil samples and slag fragments, and by the analysis of soil porewater aimed at assessing the distribution, speciation and mobility of heavy metals and As. On the site itself, arsenic concentrations up to 3.6% in surficial soil samples and up to 22% in slag fragments were found, together with elevated concentrations (percentage level) of Cu, Co, Ni, Zn and Pb. High concentrations of arsenic (up to 38,000 microg/l) and heavy metals (up to 1700 microg/l Cu and 4700 microg/l Zn) were also found in the in situ sampled soil porewater, highlighting the considerable availability of As, Zn and Cu for uptake by plants and leaching to the ground water. Sequential extractions also indicated a high availability of arsenic and copper in most samples and slag fragments of the industrial site, although poorly reactive phases were encountered as well. pHstat leaching tests confirmed that the present leaching of contaminants is alarming. Moreover, soil acidification will enhance the leaching of contaminants, emphasizing that remediation of the industrial site is urgent. Small scale variability of total metal concentrations and metal speciation, both in the horizontal and vertical direction, and the occurrence of a camouflage layer underline the importance of elaborate sampling for pollution assessment on an industrial site.  相似文献

Sources of heavy metals in urban wastewater in Stockholm   总被引:4,自引:0,他引:4  
The sources of heavy metals to a wastewater treatment plant was investigated. Sources can be actual goods, e.g. runoff from roofs, wear of tires, food, or activities, e.g. large enterprises, car washes. The sources were identified by knowing the metals content in various goods and the emissions from goods to sewage or stormwater. The sources of sewage water and stormwater were categorized to enable comparison with other research and measurements. The categories were households, drainage water, businesses, pipe sediment (all transported in sewage water), atmospheric deposition, traffic, building materials and pipe sediment (transported in stormwater). Results show that it was possible to track the sources of heavy metals for some metals such as Cu and Zn (110 and 100% found, respectively) as well as Ni and Hg (70% found). Other metals sources are still poorly understood or underestimated (Cd 60%, Pb 50%, Cr 20% known). The largest sources of Cu were tap water and roofs. For Zn the largest sources were galvanized material and car washes. In the case of Ni, the largest sources were chemicals used in the WTP and drinking water itself. And finally, for Hg the most dominant emission source was the amalgam in teeth. For Pb, Cr and Cd, where sources were more poorly understood, the largest contributors for all were car washes. Estimated results of sources from this study were compared with previously done measurements. The comparison shows that measured contribution from households is higher than that estimated (except Hg), leading to the conclusion that the sources of sewage water from households are still poorly understood or that known sources are underestimated. In the case of stormwater, the estimated contributions are rather well in agreement with measured contributions, although uncertainties are large for both estimations and measurements. Existing pipe sediments in the plumbing system, which release Hg and Pb, could be one explanation for the missing amount of these metals. Large enterprises were found to make a very small contribution, 4% or less for all metals studied. Smaller enterprises (with the exception of car washes) have been shown to make a small contribution in another city; the contribution in this case study is still unknown.  相似文献

White storks (Ciconia ciconia) fed in contaminated waters resulting from the Aznacollar acid mining-sludge spillage into the R. Guadiamar, which feeds the eastern flank of the Guadalquivir marshes (Do?ana), S.W. Spain. The sludge was rich in a range of toxic elements, and in organic pollutants such as the aromatic amines. Storks did not exhibit elevated metals in their blood immediately following the accident, but chick blood collected the year following the accident showed genotoxic damage compared to the controls. In this study lead isotope analysis was used to assess if the storks had ingested sludge-derived contaminants. The sludge lead isotope ratio was distinct from that of the Do?ana sediments. The stork blood lead isotope ratios exactly matched that of the sludge. It was concluded that the storks had ingested sludge-derived contaminants. A detailed study of the lead contamination along the R. Guadiamar and the R. Guadalquivir (of which the Guadiamar is a tributary) was also conducted to place the white stork colony lead exposure in the context of the spatial contamination of the storks' habitat.  相似文献

The objective of this study was to estimate the daily fat yield and fat percentage from one sampled milking per cow per test day in an automatic milking system herd, when the milking times and milk yields of all individual milkings are recorded by the automatic milking system. Multiple regression models were used to estimate the 24-h fat percentage when only one milking is sampled for components and milk yields and milking times are known for all milkings in the 24-h period before the sampled milking. In total, 10,697 cow test day records, from 595 herd tests at 91 Dutch herds milked with an automatic milking system, were used. The best model to predict 24-h fat percentage included fat percentage, protein percentage, milk yield and milking interval of the sampled milking, milk yield, and milking interval of the preceding milking, and the interaction between milking interval and the ratio of fat and protein percentage of the sampled milking. This model gave a standard deviation of the prediction error (SE) for 24-h fat percentage of 0.321 and a correlation between the predicted and actual 24-h fat percentage of 0.910. For the 24-h fat yield, we found SE = 90 g and correlation = 0.967. This precision is slightly better than that of present a.m.-p.m. testing schemes. Extra attention must be paid to correctly matching the sample jars and the milkings. Furthermore, milkings with an interval of less than 4 h must be excluded from sampling as well as milkings that are interrupted or that follow an interrupted milking. Under these restrictions (correct matching, interval of at least 4 h, and no interrupted milking), one sampled milking suffices to get a satisfactory estimate for the test-day fat yield.  相似文献

The objective was to test whether calves with the Leu/Leu genotype release more growth hormone (GH) than calves with Leu/Val and Val/Val genotypes. Danish Holstein (n = 286), Danish Red (n = 68), and Danish Jersey (n = 61) calves were genotyped for the Leu/Val polymorphism in the GH gene and assessed for GH release following inducement by the growth hormone releasing hormone (GHRH). Three GH traits were assessed for each calf: BASELINE, PEAK, and RATE. BASELINE and PEAK are the mean concentration of GH in blood sampled before and after GHRH inducement. RATE is the disappearance rate of GH in blood sampled after GHRH inducement. Danish Jersey calves with Leu/Leu genotype had a higher PEAK and RATE than calves with the Val/Val genotype, whereas the Leu/Val genotype had an intermediate response. The contribution of the Leu/Val polymorphism to the total genetic variation of the BASELINE, PEAK, and RATE traits was 5, 30, and 27%, respectively. By contrast, the amount of GH released by the Danish Holstein and Danish Red calves was not influenced by their GH genotype. Further studies involving calves with all three genotypes are required to further elucidate whether this polymorphism has a functional role or whether it works through a linked-gene effect specific to certain cattle breeds.  相似文献

The effectiveness of three carbohydrate sources, high-moisture ear corn (HMEC), cracked shelled corn (CSC), and a 50:50 mixture of HMEC plus dried citrus pulp (DCP), fed with or without supplemental rumen-undegraded protein as expeller soybean meal (ESBM), was assessed in 48 multiparous dairy cows. All diets contained (dry mater [DM] basis) 50% alfalfa silage, 10% ryegrass silage, 28% NDF, and one of six concentrates: A) 38% HMEC; B) 38% CSC; C) 19% DCP plus 19% HMEC; D) 27% HMEC plus 12% ESBM; E) 27% CSC plus 12% ESBM; or F) 13% DCP, 13% HMEC, and 12% ESBM. Diets A, B, and C averaged 19% crude protein, of which 53% was nonprotein nitrogen (NPN), and diets D, E, and F averaged 22% crude protein, of which 40% was NPN. Cows were fed a high-energy covariate diet for 2 wk, blocked into eight groups of six, based on covariate protein yield, then randomly assigned to diets that were fed for 12 wk. Feeding ESBM increased DM intake, yields of milk, fat-corrected milk, fat, protein, SNF, and milk and blood urea concentration and decreased weight loss. There were no production differences between HMEC and CSC. However, DM intake, yields of milk, fat-corrected milk, fat, protein, lactose, SNF, and milk SNF content all were lower on the diets containing DCP versus HMEC and CSC. A 6 x 6 Latin square trial conducted at the same time with six ruminally cannulated cows showed similar effects of diet on DM intake and milk production. Ruminal ammonia was elevated by ESBM but not ruminal total amino acids and branched-chain volatile fatty acids. Ruminal propionate was highest on HMEC diets and lowest on DCP diets; acetate, butyrate and acetate-to-propionate ratio were lowest on HMEC diets and highest on DCP diets. These results indicated that, compared to HMEC and CSC, feeding the pectin-rich carbohydrate source DCP altered ruminal fermentation but depressed intake and milk production in lactating cows.  相似文献

In order to obtain a better understanding of the biochemical events taking place in Saccharomyces cerevisiae during the lag phase, the proteins expressed during the first hours after inoculation were investigated by two-dimensional (2-D) gel electrophoresis and compared to those expressed in late respiratory growth phase. The studies were performed on a haploid strain (S288C) grown in defined minimal medium. Some of the abundant proteins, whose expression relative to total protein expression was induced during the lag phase, were identified by MALDI MS, and the expression of the corresponding genes was assessed by Northern blotting. The rate of protein synthesis was found to increase strongly during the lag phase and the number of spots detected on 2-D gels increased from 502 spots just after inoculation to 1533 spots at the end of the lag phase. During the first 20 min, the number of detectable spots was considerably reduced compared to the number of spots detected from the yeast in respiratory growth just prior to harvest and inoculation (747 spots), indicating an immediate pausing or shutdown in synthesis of many proteins just after inoculation. In this period, the cells got rid of most of their buds. The MALDI MS-identified, lag phase-induced proteins were adenosine kinase (Ado1p), whose cellular role is presently uncertain, cytosolic acetaldehyde dehydrogenase (Ald6p) and (DL)-glycerol-3-phosphatase 1, both involved in carbohydrate metabolism, a ribosomal protein (Asc1p), a fragment of the 70-kDa heat shock protein Ssb1, and translationally controlled tumour protein homologue (Yk1056cp), all involved in translation, and S-adenosylmethionine synthetase I involved in biosynthesis reactions. The level of mRNA of the corresponding genes was found to increase strongly after inoculation. By pattern matching using previously published 2-D maps of yeast proteins, several other lag phase-induced proteins were identified. These were also proteins involved in carbohydrate metabolism, translation, and biosynthesis reactions. The identified proteins together with other, yet unidentified, lag phase-induced proteins are expected to be important for yeast growth initiation and could be valuable biological markers for yeast performance. Such markers would be highly beneficial in the control and optimisation of industrial fermentations.  相似文献

The present study evaluated the ability to isolate Listeria from foods, using shortened procedure of sample enrichment followed by immunomagnetic separation or filtration methods, and serological identification of isolated bacteria by colony-blot and Western blot methods with anti-p60 antibodies. By these rapid methods, identification of Listeria was achieved in much shorter time (40-48 h) than with standard cultivation and biochemical identification procedures. The rapid methods used are easy to perform and, what is most important, their specificity is very high and fulfills the expectations. The possibility to select Listeria colonies growing on non-selective media by blotting with anti-p60 antiserum seems to be particularly valuable in examination of food samples containing/not too many Listeria (1-10 CFU/25 g). However, the blot method using anti-PepD mAb specific to unique region of L. monocytogenes p60 is necessary to distinguish L. monocytogenes from other Listeria species.  相似文献

Forty-eight crossbred growing-finishing pigs were used to study the effects of polyunsaturated fatty acids (PUFA 31%= low and 50%= high) and fish oil (0, 0.2, and 0.4% capelin) diets on fatty acid composition, chemical traits, and sensory properties of the longissimus muscle, fat, and sausages. High levels of PUFA, independent of the level of fish oil, increased oxidation and rancidity for whole muscle (stored at 1 and 8 months at -23?°C) and sausages (TBARS 0.6-1.3). Fish oil at 0.4% in the diet increased TBA values of loin, but did not affect sensory evaluation scores. An interaction between PUFA and fish oil occurred for TBARS values and rancid odour in sausage, where the 0.4% fish oil and high PUFA level showed highest oxidation (TBARS 1.9). Although fish oil and high PUFA levels might contribute to a more healthy meat, their undesirable affects on palatability would limit their use.  相似文献
