Kernels from two samples of 18 ears of corn were hand-shelled into three fractions (Fl, F2 and F3) relative to the site (F3) where the ears had previously been inoculated with one of four isolates of Fusarium graminearum (Schwabe). F3 and Fl were from the nearest and remotest parts of the ear, respectively, to the inoculation site. Ears of uninoculated control corn were processed in the same manner. The resulting 30 samples were analysed, after grinding (20 mesh), for deoxynivalenol (DON), zearalenone (ZEA), ergosterol (EGS), dry matter (DM), 200 kernel weight, Kjeldahl nitrogen (N), ether extract, ash, sugar, starch, acid and neutral detergent fibre (ADF, NDF, respectively), cellulose, hemicellulose and lignin. The true metabolisable energy (TME), TME corrected to zero N balance (TMEn), true available amino acid (TAAA) and true DM digestibility (TDMD) contents of these samples and corn to which 4, 16 or 64 mg purified DON kg?1 were added, were determined using adult White Leghorn chickens. The DON, ZEA, EGS, N and ash contents were higher (P<0.001), and ether extract, TME and TMEn values were lower (p<0.01) in F3 than in Fl. Higer avilabilites accured for aspartic and glutamic acids, isoleucine, leucine and histidine in F2 and F3 than F1 (P<0.05). Carbohydrate and fibre values varied little (p>0.05) among fractions either within or among incoulation treatments. Sources of Fusarium inoculate influenced (P<0.05) the DON, ZEA, N, fat and TDMD of the corn. The TMEn and total TAAA of corn decreased (P<0.001) when purified DON was added. Moderate relationships (r=0.57?0.73) occurred between DON and ZEA, EGS and N. In general, these results are similar to those for wheat, and indicate that chemical and nutritive content of feedingstuffs may be increased when they are infected by toxigenic fungi. 相似文献

Clinical and histopathological factors fail to adequately predict outcomes in children with high-grade gliomas, indicating a need to identify relevant biological markers of tumor behavior to guide therapeutic decision-making. Basic fibroblast growth factor (bFGF) is a mitogenic and angiogenic factor that has been observed to be overexpressed in a significant percentage of malignant gliomas, although the prognostic significance of this expression is unknown. To address this issue, the expression status of bFGF was examined immunohistochemically in a series of 27 archival pediatric malignant non-brainstem gliomas treated consecutively at our institution between 1975 and 1992. Tumors were categorized based on expression levels, and the association between expression status and outcome was examined. Sixteen cases showed high levels of expression of bFGF, and 11 showed low levels. There was no correlation between expression status and either tumor histology, patient age, or tumor location. However, there was a significant difference in outcome between patients with high levels of bFGF immunoreactivity and those with low expression. Median progression-free survival was >66 months in the low bFGF group as compared to 6 months in the high bFGF group (P = 0.006). Median overall survival was >66 months in the low bFGF group as compared to 18 months in the high bFGF group (P = 0.03). Tumor bFGF expression seems to be strongly associated with outcome in children with high-grade gliomas and, consequently, may serve as a biological correlate of patient prognosis in conjunction with other prognostic variables. 相似文献

Based on the concept of the common mucosal immune system, immunization at various inductive sites can induce an immune response at other, remote mucosal surfaces. The immune responses elicited through rectal and oral routes of antigen delivery were compared with respect to (i) measurement of antibody responses in serum and various external secretions of the vaccinees and (ii) characterization of the nature and homing potentials of circulating antibody-secreting cells (ASC). Specific ASC appeared in the circulation in 4 of 5 volunteers after oral and 9 of 11 volunteers after rectal immunization with Salmonella typhi Ty21a. The kinetics, magnitude, and immunoglobulin isotype distribution of the ASC responses were similar in the two groups. In both groups, almost all ASC (99 or 95% after oral or rectal immunization, respectively) expressed alpha4 beta7, the gut homing receptor (HR), whereas L-selectin, the peripheral lymph node HR, was expressed only on 22 or 38% of ASC, respectively. Oral immunization elicited a more pronounced immune response in saliva and vaginal secretion, while rectal immunization was more potent in inducing a response in nasal secretion, rectum, and tears. No major differences were found in the abilities of the two immunization routes to induce a response in serum or intestinal secretion. Thus, the rectal antigen delivery should be considered as an alternative to the oral immunization route. The different immune response profiles found in various secretions after oral versus rectal antigen administration provide evidence for a compartmentalization within the common mucosal immune system in humans. 相似文献

Conductivity gradient focusing (CGF) is one member of a family of gradient focusing techniques, characterized by two opposing forces which produce a dynamic equilibrium and which are able to simultaneously separate and concentrate proteins. In CGF, the two counteracting forces result from a constant convective flow of buffer opposed by an electric field gradient. This gradient in the electric field is formed by gradually decreasing buffer conductivity, i.e., when a slow-moving, relatively high conductivity buffer is dialyzed against a low conductivity purge buffer. This paper presents the design of an analytical-scale CGF device and the results of several experiments with colored proteins, both in free solution and with the use of a 45 micron size-exclusion (SEC) packing to decrease dispersion. Experimental results with hemoglobin suggest that CGF may one day be capable of resolving proteins with small charge differences. A linear computer model of conductivity gradient focusing is derived, and some suggestions are made for further development of this new electrophoretic method. 相似文献

The theophylline-binding RNA aptamer contains a 15 nucleotide motif that is required for high-affinity ligand binding. One residue within this RNA motif is only semiconserved and can be an A or C. This residue, C27, was disordered in the previously determined three-dimensional structure of the complex, suggesting that it is dynamic in solution. 13C Relaxation measurements are reported here, demonstrating that C27 is highly dynamic in the otherwise well-ordered RNA-theophylline complex. A synthetic complex with an abasic residue at position 27 was found to exhibit wild-type binding affinity (Kd approximately 0.2 microM), indicating that the base of residue 27 is not directly involved with theophylline binding. Surprisingly, the U27 and G27 RNAs were found to bind theophylline with low affinity (Kd values > 4 microM). NMR spectroscopy on the U27 RNA revealed the presence of an A7-U27 base pair in the free RNA that prevents formation of a critical base-platform structural motif and therefore blocks theophylline binding. Similarly, a protonated A7H+-C27 base pair forms in the absence of theophylline at low pH, which explains the unusual pH dependence of theophylline binding of the C27 RNA aptamer. Thus the weak binding for various nucleotides at position 27 arises not from unfavorable interactions in the RNA-theophylline complex but instead from stable interactions in the free state of the RNA that inhibit theophylline binding. 相似文献
