The widely used pretest–posttest design traditionally consists of a pretest and a posttest of equal lengths. However, greater statistical power can usually be achieved by using a posttest that is somewhat longer than the pretest. This article provides a method for calculating the optimal allocation of pretest and posttest assessment units so as to maximize power and presents the accompanying reduction in required sample size for a range of parameter values. When insufficient information is available to estimate the optimal location precisely, the effects of allocating 75% of the assessment units to the posttest are evaluated. The safety and pharmacokinetics of L-627, a new injectable carbapenem antibiotic, were evaluated in healthy volunteers. In single-dose studies, 20, 40, 80, 150, 300 and 600 mg of L-627 were administered by i.v. infusions over 1 hour. Plasma concentration-time profiles were well described with a two-compartment open model. The half-life of elimination from plasma was 1.3 +/- 0.8 (mean +/- SD) hour, and the Cmax and AUC paralleled the doses given. The mean urinary recovery of unchanged L-627 within the first 12 hours was 63.1 +/- 2.7% of the dose. In the multiple-dose studies, 300 mg of L-627 (i.v. over 1 hour) was administered every 12 hours, 11 times in total and 600 mg of L-627 was administered every 12 hours, 9 times in total. No discernible accumulation of the drug in plasma was observed. There were no subjective or objective abnormal findings definitely attributable to the drug except that one subject in one of the multiple-dose regimens (300 mg b.i.d.) showed only a slight elevation of transaminase value, although the elevated value promptly recovered after completion of dosing. No abnormality was observed in the other multiple-dose regimen (600 mg b.i.d.). From these results, L-627 was concluded to be safe and well tolerated.  相似文献

Comments on the article by S. T. Fiske et al (see record 1992-07509-001) praising the American Psychological Association's (APA's) amicus brief intended to inform the Supreme Court of current scientific thinking regarding gender stereotyping. A balanced perspective, it is argued, should include both research supporting an argument and findings that are inconsistent with that argument. The APA brief gave no indication of contradictory findings. Moreover, review of the research cited indicates that more than half of the studies do not support the argument for which they were cited. A model has been developed relating wearout to breakdown in thin oxides. Wearout has been described in terms of trap generation inside of the oxide during high voltage stressing prior to breakdown. Breakdown occurred locally when the local density of traps exceeded a critical value and the product of the electric field and the higher leakage currents through the traps exceeded a critical energy density. The measurement techniques needed for determining the density of high-voltage stress generated traps have been described along with the method for coupling the wearout measurements to breakdown distributions. The average trap density immediately prior to breakdown was measured to be of the order of low-1019/cm3 in 10 nm thick oxides fabricated on p-type substrates stressed with negative gate voltages. The model has been used to describe several effects observed during measurements of time-dependent-dielectric-breakdown distributions. The area dependence of breakdown distributions, the differences in the breakdown distributions during constant current and constant voltage stressing, and the multi-modal distributions often observed were simulated using the model. Fuel cell-based automobiles have gained attention in the last few years due to growing public concern about urban air pollution and consequent environmental problems. From an analysis of the power and energy requirements of a modern car, it is estimated that a base sustainable power ofca. 50 kW supplemented with short bursts up to 80 kW will suffice in most driving requirements. The energy demand depends greatly on driving characteristics but under normal usage is expected to be 200 Wh/km. The advantages and disadvantages of candidate fuel-cell systems and various fuels are considered together with the issue of whether the fuel should be converted directly in the fuel cell or should be reformed to hydrogen onboard the vehicle. For fuel cell vehicles to compete successfully with conventional internal-combustion engine vehicles, it appears that direct conversion fuel cells using probably hydrogen, but possibly methanol, are the only realistic contenders for road transportation applications. Among the available fuel cell technologies, polymer-electrolyte fuel cells directly fueled with hydrogen appear to be the best option for powering fuel cell vehicles as there is every prospect that these will exceed the performance of the internal-combustion engine vehicles but for their first cost. A target cost of $ 50/kW would be mandatory to make polymer-electrolyte fuel cells competitive with the internal combustion engines and can only be achieved with design changes that would substantially reduce the quantity of materials used. At present, prominent car manufacturers are deploying important research and development efforts to develop fuel cell vehicles and are projecting to start production by 2005.  相似文献

Intravascular perfluorochemical (PFC) emulsions together with a high oxygen (O2) tension may increase the delivery of dissolved O2 to useful levels. A severely anemic model of cardiopulmonary bypass (CPB) was used to test the hypothesis that a novel PFC emulsion (PFCE; Oxygent [Alliance Pharmaceutical Corp., San Diego, CA] 90% w/v perflubron) used at a high PO2 during bypass delivers sufficient O2 to ameliorate hypoxic myocardial contractile dysfunction. Acutely anemic dogs (N = 42; hematocrit = 15.8 +/- 0.6% [mean +/- SEM] before CPB and 10.9 +/- 0.1% during CPB) were divided into four groups. Group 1 was a control (n = 12). As CPB was initiated, groups 2 (n = 10), 3 (n = 10), and 4 (n = 10) had 1.35 g PFC.kg-1, 2.7 g PFC.kg-1, or 5.4 g PFC.kg-1 added via the venous return cannula. Pre-CPB and post-CPB cardiac function was measured by the first derivative of left ventricular pressure (dP/dtmax). The dP/dtmax on separation from CPB was: group 1, 619 +/- 96; group 2, 738 +/- 56; group 3, 782 +/- 101; and group 4, 828 +/- 100 (p < 0.05 groups 3 and 4 versus group 1). Mortality during the first hour after separation from CPB was higher in group 1 than in PFCE treated dogs; however, this trend did not attain statistical significance (p < 0.065). The PFC dose was higher in survivors than in nonsurvivors (2.6 +/- 0.4 g PFC.kg-1 versus 1.2 +/- 0.5 g PFC.kg-1; p < 0.05). A PFCE used at a high PO2 provides sufficient physically dissolved O2 to relieve myocardial hypoxic injury in a severely anemic model of CPB. Current PFCEs are effective O2 carriers. This finding suggests that they can be used as a temporary erythrocyte substitute to diminish the need for allogeneic transfusions during cardiac operations.  相似文献

Multisystemic therapy (MST) delivered through a community mental health center was compared with usual services delivered by a Department of Youth Services in the treatment of 84 serious juvenile offenders and their multiproblem families. Offenders were assigned randomly to treatment conditions. Pretreatment and posttreatment assessment batteries evaluating family relations, peer relations, symptomatology, social competence, and self-reported delinquency were completed by the youth and a parent, and archival records were searched at 59 wks postreferral to obtain data on rearrest and incarceration. In comparison with youths who received usual services, youths who received MST had fewer arrests and self-reported offenses and spent an average of 10 fewer weeks incarcerated. In addition, families in the MST condition reported increased family cohesion and decreased youth aggression in peer relations. This paper presents a relatively straightforward approach to the modelling of electrochemical reactors operated in batch or continuous modes. The models are based on ideal flow assumptions of either well-mixed or plug flow and incorporate reaction rate models based on electrochemical kinetics and mass transport at one electrode. General characteristics of the reactor models are described, particularly with regard to the need for good mass transport in metal recovery applications. An example is given on the use of the model in the recovery of a heavy metal (Cd2+) from an acidified solution containing Cd(II) and Fe(III) ions. The concept of Motif Combination was introduced in the French Automotive industry's R&W standard. There has however been no formal mathematical theory for the technique of motif combination. This paper sets out to develop this theory. The main result of the paper is a set of four properties that the motif combination rules must satisfy in order to have certain desirable metrological properties.Motif methods may well provide useful techniques which complement rather than replace the traditional approach based on a mean line. It was found that many functional problems could be simulated very efficiently using motif methods. Examples of functional simulation via motif combination are illustrated in the paper.  相似文献
