Objectives: The aim of this study was to evaluate the effect of bioactive glass–ceramic particles (Biosilicate®) addition on surface nanoroughness and topography of Resin-modified glass ionomer cements (RMGICs).

Methods: Experimental materials were made by incorporating 2 wt% of Biosilicate® into Fuji II LC® (FL) and Vitremer® (VT) powders. Disks of RMGICs (with and without Biosilicate®) measuring 0.5 cm (diameter) × 0.5 mm (thickness) were fabricated and polished. Samples were stored at 37 °C in dry or immersed in distilled water for 30 days. Digital images (20 × 20 μm) from the surfaces were obtained by means of an atomic force microscopy. Three images were acquired for each sample, and four nanoroughness measurements were performed in each image. Nanoroughness (Ra, nm) was assessed by Nanoscope Software V7. Data were analyzed with ANOVA and Student–Newman–Keuls multiple comparisons (p < 0.05). SEM images were obtained for surface topography analysis.

Results: FL was significantly rougher than VT (p < 0.05) in wet and dry conditions. The addition of Biosilicate® increased the surface roughness in VT and decreased in FL, regardless of the storage media (p ≤ 0.05). No differences existed between materials and storage conditions after Biosilicate® addition. Significance: The Biosilicate® particles addition produced changes on the surface nanoroughness of the RMGICs. These changes depended on the particles size of the original cements in dry conditions. In water storage, dissolution of the Biosilicate® particles, a silica-rich gel formation, and a hydroxyl carbonate apatite precipitation on the surface of the materials changed the nanoroughness surface. FL was the roughest in both conditions.

The onset of lactation in dairy cows is characterized by severe negative energy and protein balance. Methionine availability during this time for milk production, hepatic lipid metabolism, and immune function may be limiting. Supplementing Met to peripartal diets with adequate Lys in metabolizable protein (MP) to fine-tune the Lys:Met ratio may be beneficial. Fifty-six multiparous Holstein cows were fed the same basal diet from 50 d before expected calving to 30 d in milk. From −50 to −21 d before expected calving, all cows received the same diet [1.24 Mcal/kg of dry matter (DM), 10.3% rumen-degradable protein, and 4% rumen-undegradable protein] with no Met supplementation. From −21 d to expected calving, the cows received diets (1.54 Mcal/kg of DM, 10% rumen-degradable protein, and 5.1% rumen-undegradable protein) with no added Met (control, CON; n = 14), CON plus MetaSmart (MS; Adisseo Inc., Antony, France; n = 12), or CON plus Smartamine M (SM; Adisseo Inc.; n = 12). From calving through 30 d in milk, the cows received the same postpartum diet (1.75 Mcal/kg of DM and 17.5% CP; CON), or the CON plus MS or CON plus SM. The Met supplements were adjusted daily and top-dressed over the total mixed ration at a rate of 0.19 or 0.07% (DM) of feed for MS or SM. Liver tissue was collected on −10, 7, and 21 d, and blood samples more frequently, from −21 through 21 d. Data were analyzed using the MIXED procedure of SAS (SAS Institute Inc., Cary, NC) with the preplanned contrasts CON versus SM + MS and SM versus MS. No differences in prepartal DM intake (DMI) or body condition score were observed. After calving, body condition score was lower (2.6 vs. 2.8), whereas DMI was greater (15.4 vs. 13.3 kg/d) for Met-supplemented cows. Postpartal diet × time interactions were observed for milk fat percentage, milk fat yield, energy-corrected milk:DMI ratio, and energy balance. These were mainly due to changes among time points across all treatments. Cows supplemented with either Met source increased milk yield, milk protein percentage, energy-corrected milk, and milk fat yield by 3.4 kg/d, 0.18% units, 3.9 kg/d, and 0.18 kg/d, respectively. Those responses were associated with greater postpartum concentration of growth hormone but not insulin-like growth factor 1. There was a diet × time effect for nonesterified fatty acid concentration due to greater values on d 7 for MS; however, liver concentration of triacylglycerol was not affected by diet or diet × time but increased postpartum. Blood neutrophil phagocytosis at 21 d was greater with Met supplementation, suggesting better immune function. Supplemental MS or SM resulted in a tendency for lower incidence of ketosis postpartum. Although supplemental MS or SM did not decrease liver triacylglycerol, it improved milk production-related traits by enhancing voluntary DMI.  相似文献

Samples of virgin olive oils (105) from seven Extremaduran olive varieties (Cacereña, Carrasqueña, Cornezuelo, Corniche, Morisca, Picual, and Verdial de Badajoz) in three stage of maturity (green, semi-ripe, and ripe) were collected and the alkane, alkene, and sesquiterpene contents determined. There were significant differences at the 0.01 probability level in most of the hydrocarbons, both by variety and by state of maturity. Discriminant analysis applied to 70 samples explained 72.3% of the variance between the different groups of varieties and allowed 90% of the samples to be classified according to their variety. The acceptability of the model was verified against the remaining 35 samples, giving a mean level of correct classification of 94%.  相似文献

In food fermentation, many types of immobilization systems are used, such as hydrogel entrapment, where alginate is the main biopolymer. One of the important problems in industrial processes is the quantifications of biomass, since the traditional system of direct cell counting cannot be used. In this study, a simple digital imaging method to determine the biomass of yeasts immobilized into alginate capsules was developed. Important evidence of the yeasts growing inside the alginate was the change in the surface color of the capsule. Digital images were taken with different biomass concentration, and the RGB-analysis showed significant differences in the blue field. The histogram of the blue channel was used to develop a PLS multivariate calibration to predict biomass concentration. The method was validated in primary beer fermentation with good efficiency.  相似文献

Multispectral images of natural scenes were collected from both forests and coral reefs to represent typical, complex scenes that might be viewed by modern animals. Both reflectance spectra and modeled visual color signals in these scenes were decorrelated spectrally by principal-component analysis. Nearly 98% of the variance of reflectance spectra and color signals can be described by the first three principal components for both forest and coral reef scenes, which implies that three well-designed visual channels can recover almost all of the spectral information of natural scenes. A variety of natural illuminants affects color signals of forest scenes only slightly, but the variation in ambient irradiance spectra that is due to the absorption of light by water has dramatic influences on the spectral characteristics of coral reef scenes.  相似文献

Glyceraldehyde-3-phosphate dehydrogenase (G3PDH) is often used as a control gene for mRNA expression, however it has been proposed to be overexpressed in all hepatocellular carcinomas (HCC). Equal amounts of tumor and paired normal (T/N) RNA, based on OD260/280 nm, were compared using ethidium bromide staining, poly-T probing, gene-specific dot blot and Northern blots using control probes G3PDH, actin and histone H4. Using mRNA blots 13/20 surgical HCC pairs did not overexpress G3PDH. Those 7/20 intact samples which did appear to overexpress G3PDH on Northern blot could not be detected by poly-T probing of dot blots. The apparent overexpression was not specific for the control gene G3PDH nor for the malignancy HCC. It may represent partial mRNA degradation, or the presence of as yet unknown substances which interfere with absorption at 260/280 nm. We advise caution in selecting human T/N pairs for differential gene expression studies. For HCC, no clinicopathological variables, including cirrhosis, predicted whether a T/N sample pair was likely to be balanced or not.  相似文献

OBJECTIVE: To investigate the roles of brain angiotensin II and C-type natriuretic peptide (CNP) in the hypertensive mechanism of deoxycorticosterone acetate (DOCA)-salt hypertension. METHODS: We injected 50 microg/kg CV-11 974, an angiotensin II type-1 receptors antagonist, 30 nmol/kg CNP-22, or the vehicle (artificial cerebrospinal fluid) into the cerebral ventricle or intravenously 5 min before the intracerebroventricular infusion of 1.5 mol/I NaCl solution for 30 min into either male normotensive Wistar rats or DOCA-salt hypertensive rats anesthetized with urethane, and their arterial pressures and heart rates were continuously recorded. Blood (2 ml) was collected at the end of the infusion for the measurement of plasma concentration of arginine vasopressin. We infused 10 or 50 microg/kg per day CV-11 974, 10 or 50 nmol/kg per day CNP-22, or the vehicle (1 microl/h) into the cerebral ventricles of DOCA-salt hypertensive rats for 7 days by using osmotic minipumps, and measured their systolic arterial pressures, pulse rates, and urinary excretions of vasopressin. RESULTS: Intracerebroventricular pre-administrations of CV-11 974 and of CNP-22 inhibited increases in mean arterial pressure, heart rate, and plasma vasopressin concentration induced by intracerebroventricular infusion of 1.5 mol/l NaCl into normotensive rats; increases in hemodynamics and plasma level of vasopressin induced by intracerebroventricular infusion of 1.5 mol/l NaCl were suppressed by intracerebroventricular pre-injections of CV-11 974, but not of CNP-22, into DOCA-salt hypertensive rats. Continuous intracerebroventricular infusions of 50 microg/kg per day CV-11 974 attenuated hypertension in DOCA-salt treated rats, accompanied by a reduction in urinary excretion of vasopressin. Continuous intracerebroventricular infusions of 50 nmol/kg per day CNP-22, however, affected neither hypertension nor urinary excretion of vasopressin in DOCA-salt hypertensive rats. CONCLUSION: Brain angiotensin II could play a role in the pressor mechanism of DOCA-salt hypertension by increasing release of vasopressin via type 1 receptors. That brain CNP has an inhibitory effect on release of vasopressin in acute experiments indicates that the impairment of this inhibitory effect of brain CNP on secretion of vasopressin could be involved in the pathogenesis of DOCA-salt hypertension in rats.  相似文献

A 54 year-old man had symptoms of acute right hemispheric cerebral ischemia. He was initially considered for participation in a trial of early thrombolysis in stroke, but an innominate artery embolus was found with no apparent arterial source. The embolus was removed by means of a combined brachial and carotid bifurcation approach to protect the cerebral vasculature from embolic fragmentation during extraction. Further investigation revealed deep venous thrombosis, evidence of pulmonary emboli, and a patent foramen ovale, supporting a diagnosis of paradoxic embolus. Additional treatment included anticoagulation and placement of an inferior vena caval filter. The unusual condition of paradoxic embolus is reviewed, and the management of this patient is discussed.  相似文献

The ability of a peptide hormone to affect many different intracellular targets is thought to be possible because of the modular organization of signal transducing molecules in the cell. Evidence for the presence of signaling modules in metazoan cells, however, is incomplete. Herein we show, with morphology and cell fractionation, that all the components of a mitogen-activated protein kinase pathway are concentrated in caveolae of unstimulated human fibroblasts. Addition of platelet-derived growth factor to either the intact cell or caveolae isolated from these cells stimulates tyrosine phosphorylation and activates mitogen-activated protein kinases in caveolae. The molecular machinery for kinase activation, therefore, is preorganized at the cell surface of quiescent cells.  相似文献
