There is much concern regarding the genesis of acidic leachate by mine tailings and its potential impact on local water systems. Genesis of the leachate and its subsequent escape into the surrounding watershed is controlled by the primary distribution of facies within the tailings pond. Leachate generation and percolation is enhanced in high-sulphide coarse-grained sands, while percolation is restricted in fine-grained clays formed from bentonitic slimes. The Kidd Copper deposit, located on the South Range of the Sudbury Basin, was a typical offset dike nickel deposit, where mining took place between 1970 and 1990. Using a sequence of temporally separate aerial photographs dating from 1945 to 2002, it is possible to document the initial development of the mine, its closure and the subsequent degradation of the mine tailings waste that was produced during mining activity. Since the mine's closure, the resulting tailings deposit has progressively oxidized, generating acid-rich leachate. Leachate can escape from the tailings via surface run-off and subsurface flow. The spatial distribution of possible flow pathways was mapped using a combination of sub-centimetre-resolution real-time kinematic (RTK) GPS and individual, high-resolution, oriented photographs of subsurface pits. Lithologic mapping information of the Kidd Copper tailings deposit can provide an insight into the distribution of local aquifers and aquitards. Subsurface lithologic mapping was achieved using a combination of sub-centimetre-resolution RTK GPS and individual, high-resolution, oriented photographs of subsurface pits. The photograph of each pit provides lithologic information of a specific point in the tailings deposit, while the high-resolution GPS locates the individual photographs in three-dimensional (3D) geographic space. Combining these data allows us to identify continuous lithologic surfaces between pits, leading to the construction of a 3D model of the deposition of the tailings pond. Grain size and textural details extracted from the images of each lithology permit discrimination between aquifers and aquitards. The final geometric model could be used as input for estimating the hydrologic and chemical evolution of the tailings.  相似文献

Efforts to generate detectable concentrations of free radicals in explosives, binders, and their mixtures are described. Radicals were readily produced in polycaprolactone and polyethylene glycol binders at liquid nitrogen temperature using stresses as low as 0.4 kbar. These radicals were all of the peroxy type, and presumably formed by reaction of mechano-radicals with oxygen present in the polymer. No mechano-radicals were observed from HMX or RDX using samples cooled to liquid nitrogen temperature and applied stresses up to 4 kbar. In neither impacted samples that failed to explode nor the residues remaining after a partial explosion were radicals detected by ESR. Low temperature γ-irradiation of these materials was also carried out. Free radical signals originating in both the polymer and the explosive could be identified. The reactivity of NO2 radicals from γ-irradiated HMX is enhanced in the presence of binder. In γ-irradiated HMX/polycaprolactone mixtures, the NO2 radical anneals rapidly at 150 K, versus 240 K for HMX alone. Evidence is presented to suggest that the relatively stable NO2 radical (such as produced by γ-irradiation) in HMX does not play a dominant role in mechanical initiation.  相似文献

Following the observation of a high correlation between the reciprocal of the speed of ultrasound and lean proportion in beef carcasses, commercial equipment (VOSI) was developed and its performance alongside a smaller assembly (Mini G) is reported. The speed of ultrasound was measured through six sites on beef carcasses within 1 h of stunning: three sites with VOSI and three sites with Mini G. Sides were also scored for fatness and conformation using the EAAP system, and subcutaneous fat depth was measured at 25,50 and 75% of the width of M. longissimus thoracis from the midline at the last rib. All sides were dissected into subcutaneous fat, intermuscular fat, muscle and bone. The reciprocal speed averaged over the three VOSI sites (RV346) was the best single predictor of the proportion of lean in the side. Multiple regression analyses, in predicting per cent lean, showed that addition of conformation score improved the precision of fat score and fat depth for a group of 26 sides. It did not provide a significant improvement over RV346. However, in another group of 50 sides showing more variation in side mass and conformation, conformation score did significantly improve RV346. The best prediction of per cent lean, involving RV346 and tissue thicknesses, gave a residual standard deviation of 1·35%, and conformation score did not significantly improve this regression.  相似文献

A chimeric protein consisting of CXC-chemokine receptor 4 (CXCR4) and the green fluorescent protein (GFP) was used for studying receptor localization and trafficking in real time in stably transduced HeLa, U-937, CEM, and NIH/3T3 cells. CXCR4-GFP was fully active as a co-receptor in mediating human immunodeficiency virus (HIV) entry. Both CXCR4 and CXCR4-GFP were found to undergo significant spontaneous endocytosis. Only 51.5 +/- 7.8% of receptor molecules were found on the plasma membrane in CD4-positive cells, 43.9 +/- 8.5% were found in CD4-negative HeLa cells, 75.6 +/- 9.7% were found in U-937 cells, 72.5 +/- 7.9 were found in CEM cells, and almost none were found in in NIH/3T3 cells. Stromal cell-derived factor-1alpha induced rapid endocytosis of cell surface receptor molecules. A significant part of CXCR4 was targeted to lysosomes upon binding of the ligands, and recycling of internalized CXCR4 was not efficient. Only about 30% of receptor molecules recycled back to the cell surface in HeLa cells, 5% recycled in U937, and 10% recycled in CEM cells, suggesting that the protective effect of chemokines against HIV infection can be attributed not only to competition for binding but also to depletion of the co-receptor molecules from the cell surface. Envelope glycoprotein gp120 of syncytia-inducing/lymphocyte tropic HIV-1 strains induced rapid internalization of CXCR4 in both CD4-negative and CD4-positive cells, suggesting that gp120 is a high affinity ligand of CXCR4.  相似文献

Tyr114 and Tyr197 are highly conserved residues in the active site of human glutathione reductase, Tyr114 in the glutathione disulfide (GSSG) binding site and Tyr197 in the NADPH site. Mutation of either residue has profound effects on catalysis. Y197S and Y114L have 17% and 14% the activity of the wild-type enzyme, respectively. Mutation of Tyr197, in the NADPH site, leads to a decrease in Km for GSSG, and mutation of Tyr114, in the GSSG site, leads to a decrease in Km for NADPH. This behavior is predicted for enzymes operating by a ping-pong mechanism where both half-reactions partially limit turnover. Titration of the wild-type enzyme or Y114L with NADPH proceeds in two phases, Eox to EH2 and EH2 to EH2-NADPH. In contrast, Y197S reacts monophasically, showing that excess NADPH fails to enhance the absorbance of the thiolate-FAD charge-transfer complex, the predominant EH2 form of glutathione reductase. The reductive half-reactions of the wild-type enzyme and of Y114L are similar; FAD reduction is fast (approximately 500 s-1 at 4 degreesC) and thiolate-FAD charge-transfer complex formation has a rate of 100 s-1. In Y197S, these rates are only 78 and 5 s-1, respectively. The oxidative half-reaction, the rate of reoxidation of EH2 by GSSG, of the wild-type enzyme is approximately 4-fold faster than that of Y114L. These results are consistent with Tyr197 serving as a gate in the binding of NADPH, and they indicate that Tyr114 assists the acid catalyst His467'.  相似文献

Myocardial and pulmonary beta-adrenoceptors can be imaged with 2-(S)-(-)-(9H-carbazol-4-yl-oxy)-3-[1-(fluoromethyl)ethyl]amino-2- propanol (S-1'-[18F]fluorocarazolol, I). Quantification of unmodified fluorocarazolol in plasma is necessary for analysis of PET images in terms of receptor densities. We have determined I and its radioactive metabolites in rat, sheep and human plasma, using (1) solid-phase extraction (C18) followed by reversed-phase HPLC and (2) direct injection of untreated plasma samples on an internal-surface reversed-phase (ISRP) column. The two methods were in good agreement. Unmodified I decreased from over 99% initially to less than 5%, 5-10% and 20% at 60 min post-injection in rats, sheep and human volunteers, respectively. Protein binding in sheep and human plasma was determined by ultrafiltration. The fraction of total plasma radioactivity bound to protein and the fraction representing unmodified radioligand were linearly correlated, suggesting that fluorocarazolol was more than 70% protein-bound, whereas its metabolites showed negligible protein binding. Direct injection of plasma on an ISRP column seems a convenient method for quantification of lipophilic radioligands such as fluorocarazolol.  相似文献
