Performance characteristics were evaluated for two lateral-flow test kits, Reveal for Ruminant in Feed (Neogen Corporation) and FeedChek (Strategic Diagnostics Inc.), designed to detect ruminant or terrestrial animal proteins in feeds. The stringent acceptance criteria used were developed by the Center for Veterinary Medicine Office of Research to identify test kits with comparable selectivity and sensitivity to microscopy and PCR assay, the analytical methods used by the U.S. Food and Drug Administration (FDA). Guidelines were developed for evaluating the selectivity, sensitivity, ruggedness, and specificity of these kits. These guidelines further stated that ruggedness and specificity testing would be performed only after a test passed both the selectivity and sensitivity assessments. Acceptance criteria for determining success were developed using a statistical approach requiring 90% probability of achieving the correct response, within a 95% confidence interval. A minimum detection level of 0.1% bovine meat and bone meal, consistent with the sensitivity of the methods used by the FDA, was required. Selectivity was assessed by testing 60 dairy feed samples that contained no added animal proteins; sensitivity was determined by evaluating 60 samples (per level of fortification) of the same feed that contained 0.025, 0.05, 0.1, 0.25, 0.5, 1, or 2% bovine meat and bone meal. The Reveal test passed the selectivity assessment but failed the sensitivity assessment, detecting only samples fortified at the 2% level and then only 17 to 33% of those samples, when read according to the label directions. The FeedChek test passed the sensitivity assessment but failed the selectivity assessment, with rates for false-positive results ranging from 34 to 38%, depending on the user. The sensitivity of the Reveal test was affected by the concentration of trace minerals present in the feed; concentrations toward the high end of the normal range prevented the detection of true positive feed samples containing bovine meat and bone meal. Better sensitivity assessments were obtained when lamb meal was used either alone or in combination with bovine meat and bone meal. The FeedChek test was not affected by the concentration of trace minerals or by the type of animal meal used. The reactive aldehyde acrolein is a ubiquitous environmental pollutant and is also generated endogenously. It is a strong electrophile and reacts rapidly with nucleophiles including thiolates. This review focuses on the effects of acrolein on thioredoxin reductase (TrxR) and thioredoxin (Trx), which are major regulators of intracellular protein thiol redox balance. Acrolein causes irreversible effects on TrxR and Trx, which are consistent with the formation of covalent adducts to selenocysteine and cysteine residues that are key to their activity. TrxR and Trx are more sensitive than some other redox-sensitive proteins, and their prolonged inhibition could disrupt a number of redox-sensitive functions in cells. Among these effects are the oxidation of peroxiredoxins and the activation of apoptosis signal regulating kinase (ASK1). ASK1 promotes MAP kinase activation, and p38 activation contributes to apoptosis and a number of other acrolein-induced stress responses. Overall, the disruption of the TrxR/Trx system by acrolein could be significant early and prolonged events that affect many aspects of redox-sensitive signaling and oxidant stress. 相似文献

Legume residues have been credited with supplying mineral nitrogen (N) to the associated cereal crop and improving soil fertility in the long term. Few studies using15N have reported the fate of legume N and fertilizer N in the presence of legume residues in soil-plant systems over periods of two years or longer. A field experiment was conducted in microplots to evaluate: (1) the residual value of the15N added in leucaena residues; (2) the residual value of fertilizer15N applied in the presence of unlabelled leucaena residues in the first year to maize over three subsequent years; and (3) the long-term fate of residual fertilizer and leucaena15N in a leucaena alley cropping system.There was a significant increase in maize production over three subsequent years after addition of leucaena residues. The residual effect of fertilizer N increased maize yield in the second year when N fertilizer was applied at 36 kg N ha–1 in the first year in the presence of leucaena residues. Of the leucaena15N applied in the first year, the second, third and fourth maize crop recovered 2.6%, 1.8% and 1.4%, respectively. The corresponding values for the residual fertilizer15N were 0.7%, 0.4% and 0.3%. About 12–14% of the fertilizer15N added in the first year was found in the 200 cm soil profile over the following three years. This differed from the 38–41% of leucaena15N detected in the soil over the same period. Most of the residual fertilizer and leucaena15N in the soil was immobilized in the top 25 cm with less than 1% leached below 100 cm. More than 36% of the leucaena15N and fertilizer15N added in the first year was apparently lost from the soil-plant system in the first two years. No further loss of the residual leucaena and fertilizer15N was detected after two years. 相似文献

Urea–formaldehyde-bonded wood products are limited to interior nonstructural applications because of their poor durability under cyclic moisture or humid environments. The stability of solid-wood joints and particleboards can be enhanced by bonding with urea–formaldehyde adhesives modified with di- and trifunctional amines at an effective resin formaldehyde-to-urea mol ratio (F/U) of 1.6; however, particleboard formaldehyde emissions were not improved over those from boards made with unmodified adhesives and were unacceptably high. The relative effectiveness of selected modifications was investigated at resin form aldehyde-to-urea (F/U) molar ratios of 1.4 and 1.2 Solid-wood joints and particleboards made with modified adhesives, an unmodified adhesive, and a phenol formaldehyde (PF) resin were subjected to cyclic soak-dry (cyclic stress) treatments and moist-heat aging. Formaldehyde emissions from particleboards were also determined. At F/U 1.4, the resistance of solid-wood joints made with modified adhesives to cyclic stress and moist-heat aging was equal to that of PF-bonded joints and superior to that of joints bonded with unmodified adhesive. The resistance of particleboards made with modified adhesives was greater than that of boards made with unmodified adhesive but less than that of PF-bonded board. Solid-wood joints and particleboards made with F/U 1.4 resins performed better than did those made with F/U 1.2 resins. The Percus-Yevick equation of state is applied to a two-dimensional adsorbed fluid. Calculation of the adsorption isotherm is divided into two parts: the Henry's constant for gas-solid interactions, and the configurational contribution of the two-dimensional film. The theory is compared with experimental data for adsorption of argon and krypton on graphitized carbon black and agreement is excellent. Tobacco use is prevalent among youth with alcohol and other drug problems, yet this issue has received limited research and clinical attention. This study reports on a controlled evaluation of a cigarette smoking intervention with 54 adolescents in treatment for substance abuse, ages 13-18 (22% female). Participants were assessed at 4 time points. A greater proportion of participants in the treatment condition (n = 26) reported cessation attempts and point abstinence than did control participants (n = 28) at all time points. However, significant differences were found only for point abstinence at a 3-month follow-up. These findings provide initial support for the efficacy of a smoking cessation intervention delivered in the context of adolescent substance abuse treatment. Recent studies support the concept that pulmonary granulomatous inflammation directed by interferon (IFN)-gamma, interleukin (IL)-12, and nitric oxide usually resolves in the absence of fibrosis. To determine whether nitric oxide participates in modulating the fibrotic response during the development of pulmonary granulomas in response to purified protein derivative (PPD), mice presensitized to PPD received daily intraperitoneal injections of N(G)-nitro-D-arginine-methyl ester (D-NAME), N(G)-nitro-L-arginine-methyl ester (L-NAME), or aminoguanidine after delivery of PPD-coated beads to the lungs. Eight days later, morphometric analysis of lung granulomas revealed that L-NAME-treated mice when challenged with PPD in vitro for 36 hours had the largest pulmonary granulomas and the greatest collagen deposition among the treated groups. In addition, equivalent numbers of dispersed lung cells from L-NAME- and aminoguanidine-treated mice produced significantly higher levels of IL-4, monocyte chemoattractant protein (MCP)-1, and macrophage inflammatory protein (MIP)-1alpha and significantly lower levels of eotaxin compared with D-NAME-treated mice. Cultures of dispersed lung cells from L-NAME-treated mice also produced significantly more IL-10 and less IL-12 compared with similar numbers of dispersed lung cells from D-NAME-treated mice. Cultures of isolated lung fibroblasts from L-NAME-treated mice expressed higher levels of C-C chemokine receptor 2 (CCR2) and CCR3 mRNA and contained less MCP-1 and eotaxin protein than a similar number of fibroblasts from D-NAME-treated mice. Thus, nitric oxide appears to regulate the deposition of extracellular matrix in lung granulomas through the modulation of the cytokine and chemokine profile of these lesions. Alterations in the cytokine, chemokine, and procollagen profile of this lesion may be a direct effect of nitric oxide on the pulmonary fibroblast and provide an important signal for regulating fibroblast activity during the evolution of chronic lung disease. 相似文献

This study assessed the responsiveness of newborn breast- and bottle-fed infants to presentations of maternal odor. Maternal odor was presented for 1 min to crying, sleeping or awake newborns. The odors were: (1) own mother's odor - presentation of a hospital gown worn by the baby's mother, (2) other mother's odor - presentation of a hospital gown of another newborn baby's mother, (3) clean gown - presentation of a clean hospital gown and (4) no gown - no gown presented. The results indicated that crying babies stopped crying when either own mother or other mother odor was presented. Awake babies responded specifically to their own mother's odor by increasing mouthing. These results suggest that the practice of presenting the mother's odor to a distressed infant is of clinical usefulness since it was capable of attenuating crying. The results also characterized a role for maternal odor with respect to feeding since presentation of the infant's own mother odor increased mouthing. Thus, presentation of maternal odor may also be useful in enhancing nipple acceptance and feeding in newborns. 相似文献
