The removal of albumin and other high abundance proteins is a routine first step in the analysis of serum and plasma proteomes. However, as albumin can bind proteins and peptides, there is a universal concern as to how the serum proteome is changed by the removal of albumin. To address this concern, the current study was designed to identify proteins and peptides removed from the serum during albumin depletion; to determine which of these are bound to albumin (rather than copurified) and whether the bound proteins are intact proteins or peptide fragments. Sequential, independent analyses including both anti‐albumin antibody (anti‐HSA) affinity chromatography and SEC were used to isolate albumin‐bound proteins. RP‐HPLC and 1‐D SDS‐PAGE were then used to further separate the proteins prior to identification by MS/MS. Finally, whole protein molecular weight (MW) MS measurements coupled with protein coverage obtained by MS were combined to assess whether the bound proteins were intact or peptide fragments. Combining the results from multiple approaches, 35 proteins, of which 24 are intact, were found to be associated with albumin, and they include both known high and low abundance proteins. 相似文献

Inattentional blindness is the failure to detect unexpected events when attention is otherwise engaged. Previous research indicates that inattentional blindness increases as perceptual demands intensify. The authors present 6 cuing experiments that manipulated both the perceptual demands of a primary letter-naming task and the expectations of the individual. Inattentional blindness was greatest for individuals who held a numerical expectation that was consistent with the number of primary-task items presented. Expectation also affected detection differentially at various levels of perceptual load: Detection at moderate and high perceptual load was significantly affected by expectation, whereas detection at low perceptual load was not. The authors suggest that at moderate to high levels of perceptual load, individuals whose numerical expectations are fulfilled terminate processing when the primary task is complete, at the expense of the unexpected visual event. These experiments provide compelling evidence that expectations do affect detection of an unexpected stimulus, and they are the first to demonstrate that individuals set their attention for the number of items to be detected and are vulnerable to inattentional blindness whenever their primary-task numerical expectation is fulfilled. To investigate the trophic transfer of nanomaterials along the food chain, we examined the potential trophic transfer and biomagnification of CdSe/ZnS quantum dots (QDs) in a simple freshwater food chain. Our results indicate that QDs can transfer from zooplankton to Danio rerio (zebrafish) by dietary exposure. No significant biomagnification of QDs was observed and the biomagnification factors for both adult and juvenile zebrafish were both less than one (0.04 and 0.004 respectively). The assimilation efficiency was 8% and 4% for adult and juvenile zebrafish respectively. This study is the first to examine the potential food chain transfer and biomagnification of QDs from zooplankton to zebrafish. 相似文献

Zinc spinel ferrite, ZnFe2O4 (ZFO), is an emerging photoanode material for photoelectrochemical (PEC) solar fuel production. However, a lack of fundamental insight into the factors limiting the photocurrent has prevented substantial advance in its performance. Herein, it is found that ZFO nanorod array photoelectrodes with varying crystallinity exhibit vastly different PEC properties. Using a sacrificial hole scavenger (H2O2), spatially defined carrier generation, and electrochemical impedance spectroscopy, it is shown that ZFO with a relatively poor crystallinity but a higher spinel inversion degree (due to cation disorder) exhibits superior photogenerated charge separation efficiency and improved majority charge carrier transport compared to ZFO with higher crystallinity and a lower inversion degree. Conversely, the latter condition leads to better charge injection efficiency. Optimization of these factors, and the addition of a nickel–iron oxide cocatalyst overlayer, leads to a new benchmark solar photocurrent for ZFO of 1.0 mA cm?2 at 1.23 V versus reversible hydrogen electrode (RHE) and 1.7 mA cm?2 at 1.6 V versus RHE. Importantly, the observed correlation between the cation disorder and the PEC performance represents a new insight into the factors important to the PEC performance of the spinel ferrites and suggests a path to further improvement. 相似文献

Human exposure to nanoparticles is inevitable as nanoparticles become more widely used and, as a result, nanotoxicology research is now gaining attention. However, while the number of nanoparticle types and applications continues to increase, studies to characterize their effects after exposure and to address their potential toxicity are few in comparison. In the medical field in particular, nanoparticles are being utilized in diagnostic and therapeutic tools to better understand, detect, and treat human diseases. Exposure to nanoparticles for medical purposes involves intentional contact or administration; therefore, understanding the properties of nanoparticles and their effect on the body is crucial before clinical use can occur. This Review presents a summary of the in vitro cytotoxicity data currently available on three classes of nanoparticles. With each of these nanoparticles, different data has been published about their cytotoxicity due to varying experimental conditions as well as differing nanoparticle physiochemical properties. For nanoparticles to move into the clinical arena, it is important that nanotoxicology research uncovers and understands how these multiple factors influence the toxicity of nanoparticles so that their undesirable properties can be avoided. 相似文献

Traditional urban water management systems are ill-equipped to address future challenges. Despite policy rhetoric supporting Total Water Cycle Management and the diverse water supply approach, there are many social and institutional barriers to effective implementation. A quantitative online survey with over 1000 practitioners in three Australian capital cities identified practitioners' level of receptivity to the diverse water source approach, and their experience of the scope and priority of such barriers to their implementation. The analysis revealed a high level of practitioner support, yet a critical lack of institutional tools and incentives for supporting implementation. Organoids are 3D cultures that to some extent reproduce the structure, composition and function of the mammalian tissues from which they derive, thereby creating in vitro systems with more in vivo-like characteristics than 2D monocultures. Here, the ability of human organoids derived from normal gastric, pancreas, liver, colon and kidney tissues to metabolise the environmental carcinogen benzo[a]pyrene (BaP) was investigated. While organoids from the different tissues showed varied cytotoxic responses to BaP, with gastric and colon organoids being the most susceptible, the xenobiotic-metabolising enzyme (XME) genes, CYP1A1 and NQO1, were highly upregulated in all organoid types, with kidney organoids having the highest levels. Furthermore, the presence of two key metabolites, BaP-t-7,8-dihydrodiol and BaP-tetrol-l-1, was detected in all organoid types, confirming their ability to metabolise BaP. BaP bioactivation was confirmed both by the activation of the DNA damage response pathway (induction of p-p53, pCHK2, p21 and γ-H2AX) and by DNA adduct formation. Overall, pancreatic and undifferentiated liver organoids formed the highest levels of DNA adducts. Colon organoids had the lowest responses in DNA adduct and metabolite formation, as well as XME expression. Additionally, high-throughput RT-qPCR explored differences in gene expression between organoid types after BaP treatment. The results demonstrate the potential usefulness of organoids for studying environmental carcinogenesis and genetic toxicology. 相似文献

A second-generation enantiospecific synthesis of spiroleucettadine is described. The original reported antibacterial activity was not observed when the experiment was repeated on the synthetic samples; however, significant anti-proliferative activity was uncovered for both enantiomers of spiroleucettadine. Comparison of the optical rotational data and ORD-CD spectra of both enantiomers and the reported spectrum from the natural source have not provided a definitive answer regarding the absolute stereochemistry of naturally occurring spiroleucettadine. Efforts then focussed on alteration at the C-4 and C-5 positions of the slightly more active (−)-spiroleucettadine. Ten analogues were synthesised, with three analogues found to possess similar anti-proliferative profiles to spiroleucettadine against the H522 lung cancer cell line. 相似文献

The continuation of Moore's law for semiconductor fabrication envisages the introduction of extreme ultraviolet lithography (EUVL) based on a source wavelength of 13.5?nm for high-volume manufacturing within the next few years. While exposure tools have already been developed and the feasibility of the technology well demonstrated, the key source requirement in terms of power output remains to be achieved. Currently, sources based on laser-produced plasmas from tin droplet targets appear to be the most promising and are being deployed in manufacturing tools. Progress in CO2 laser design aimed at increasing conversion efficiency to close to 5% should make possible the attainment of greater than 200?W of in-band optical power. Recently, research has commenced on the development of sources operating at a wavelength near 6.7?nm for beyond 13.5?nm lithography and gadolinium has been identified as the fuel of choice. The results of these experiments are described and show many similarities to the behavior of tin plasmas as essentially the same atomic and plasma processes are involved, albeit at an electron temperature close to a factor of three higher. 相似文献

This study compared the effects of implanting two interpenetrating polymer networks (IPNs) into rabbit corneas. The first (Implant 1) was based on PEG-diacrylate, the second (Implant 2) was based on PEG-diacrylamide. There were inserted into deep stromal pockets created using a manual surgical technique for either 3 or 6 months. The implanted corneas were compared with normal and sham-operated corneas through slit lamp observation, anterior segment optical coherence tomography, in vivo confocal scanning and histological examination. Corneas with Implant 1 (based on PEG-diacrylate) developed diffuse haze, ulcers and opacities within 3 months, while corneas with Implant 2 (based on PEG-diacrylamide) remained clear at 6 months. They also exhibited normal numbers of epithelial cell layers, without any immune cell infiltration, inflammation, oedema or neovascularisation at post-operative 6 month. Morphological studies showed transient epithelial layer thinning over the hydrogel inserted area and elevated keratocyte activity at 3 months; however, the epithelium thickness and keratocyte morphology were improved at 6 months. Implant 2 exhibited superior in vivo biocompatibility and higher optical clarity than Implant 1. PEG-diacrylamide-based IPN hydrogel is therefore a potential candidate for corneal inlays to correct refractive error. 相似文献
