Phospholipase D (PLD), isolated from Indian mustard seeds and purified to homogeneity, has recently been identified as typical α‐type PLD with a high activity toward phosphatidylcholine (PC) in an aqueous mixed micellar system (Khatoon et al. 2014, Phytochemistry 117, 65–75). In light of biocatalytical application, we have now studied the enzyme in aqueous‐organic two‐phase systems and compared the results with the properties of the enzyme in aqueous mixed micellar systems. n‐Hexane containing 5–10 mol% of 1‐ or 2‐octanol proved to be optimal as an organic solvent in the two‐phase system, whereas anionic detergents such as sodium dodecyl sulfate or sodium deoxycholate were favorable components in the mixed micellar system. The optimum pH value (5.5–5.6) and the optimum Ca2+ ion concentration (70 mM) were independent of the reaction system. The head group selectivity in terms of activity toward different phospholipids (PC > phosphatidylethanolamine > phosphatidylglycerol) was similar in both types of reaction systems. Also, the KM values toward PC were on the same order of magnitude. In contrast, the Vmax value in the two‐phase system was 20‐fold lower than in the mixed micellar system. The enzyme was able to catalyze the substitution of choline in PC by ethanolamine, glycerol, and ethylene glycol with high efficiency. l ‐Serine was exchanged for choline with low activity. Myo‐inositol was not an alcohol acceptor, but promoted the hydrolysis of PC.  相似文献

Recent research has indicated that the younger generations may be disconnected from nature and even show some dislike for natural conditions. This study addressed adolescents’ preferences for riverscapes with varying fluvial dynamics as recreational settings. A stated choice survey with digitally calibrated river scenarios asked Austrian pupils (N = 281) about their preferences for several physical and social characteristics of a floodplain landscape. Pupils perceived riverscapes with a high water dynamic as best for recreation, while water bodies with low water levels and algae, dry river sites, floods and littered trails were disliked. Heterogeneity among the pupils was found with one smaller segment preferring settings with high water dynamic and low human impact, while the larger one preferred settings with more human impact and low water dynamic. This segment that shows some dislike for natural conditions had less experience with rivers and scored lower on natural river-related attitudes and perceptions. Implications for recreational river planning and aesthetic assessments are derived.  相似文献

Electroceramic support materials can help reducing the noble-metal loading of iridium in the membrane electrodes assembly (MEA) of proton exchange membrane (PEM) electrolyzers. Highly active anodes containing Ir-black catalyst and submicronic Ti4O7 are manufactured through screen printing technique. Several vehicle solvents, including ethane-1,2-diol; propane-1,2-diol and cyclohexanol are investigated. Suitable functional anodic layer with iridium loading as low as 0.4 mg cm?2 is obtained. Surface properties of the deposited layers are investigated by atomic force microscopy (AFM). The most homogeneous coating with the highest electronic conductivity is obtained using cyclohexanol. Tests in PEM electrolyzer operating at 1.7 V and 40 °C demonstrate that the CCM with anode coated with cyclohexanol presents a 1.5-fold higher Ir-mass activity than that of the commercial CCM.  相似文献

This paper describes an ongoing effort focused on combined research and curriculum development for multidisciplinary, geographically distributed architecture∕engineering∕construction (A∕E∕C) teamwork. It presents a model for a distributed A∕E∕C learning environment and an Internet-based Web-mediated collaboration tool kit. The distributed learning environment includes six universities from Europe, Japan, and the United States. The tool kit is aimed to assist team members and owners (1) capture and share knowledge and information related to a specific project; (2) navigate through the archived knowledge and information; and (3) evaluate and explain the product's performance. The A∕E∕C course offered at Stanford University acts as a testbed for cutting-edge information technologies and a forum to teach new generations of professionals how to team up with practitioners from other disciplines and take advantage of information technology to produce a better, faster, more economical product. The paper presents new assessment metrics to monitor students' cross-disciplinary learning experience and track programmatic changes. The paper concludes with challenges and quandaries regarding the impact of information technologies on team performance and behavior.  相似文献

Organic Phosphorus Compounds. XII. The Hydrolytic Behaviour of O,O-Dimethyl O-(1,2-Dibromo-2,2-dichloroethyl) Phosphate (Naled) The hydrolysis of Naled 2 , an organophosphorus pesticide accessible by bromination of the enolphosphate Dichlorvos 1 , in unbuffered aqueous solution does not only lead — as hitherto described — to dimethyl phosphate 3 and bromodichloroacetaldehyde 4 (path A), but also via intermediate formation of Desmethylnaled 5 to monomethyl phosphate 6 (path B). Kinetic measurements show that in the temperature range of 20–30°C both reactions proceed with similar velocities, and the primary step 2 → 5 determines the rate of path B. In comparison to Dichlorvos 1 the hydrolytic stability of Naled 2 has been found to be markedly lower. — The information on the degradation of Naled in unbuffered aqueous solution is completed by studies on the pH-dependence of the hydrolysis and on the alkylation capacity of this pesticide.  相似文献

The occurrence, distribution, and temperature dependence in the marine atmosphere of several alternative brominated flame retardants (BFRs), Dechlorane Plus (DP) and polybrominated diphenyl ethers (PBDEs) were investigated during a sampling cruise from the East Indian Archipelago toward the Indian Ocean and further to the Southern Ocean. Elevated concentrations were observed over the East Indian Archipelago, especially of the non-PBDE BFR hexabromobenzene (HBB) with concentrations up to 26 pg m(-3) which were found to be related to continental air masses from the East Indian Archipelago. Other alternative BFRs- pentabromotoulene (PBT), pentabromobenzene (PBBz), and 2,3-dibromopropyl-2,4,6-tribromophenyl ether (DPTE)-were elevated, too, with concentrations up to 2.8, 4.3, and 2.3 pg m(-3), respectively. DP was detected from 0.26 to 11 pg m(-3) and bis-(2-ethylhexyl)-tetrabromophthalate (TBPH) ranged from not detected (nd) to 2.8 pg m(-3), respectively. PBDEs ranged from nd to 6.6 pg m(-3) (Σ(10)PBDEs) with the highest individual concentrations for BDE-209. The approach of Clausius-Clapeyron (CC) plots indicates that HBB is dominated by long-range atmospheric transport at lower temperatures over the Indian and Southern Ocean, while volatilization processes and additional atmospheric emissions dominate at higher temperatures. In contrast, BDE-28 and -47 are dominated by long-range transport without fresh emissions over the entire cruise transect and temperature range, indicating limited fresh emissions of the meanwhile classic PBDEs.  相似文献

Endometriosis is a gynaecological disease defined by the growth of endometrium-like tissue outside the uterus. The disease is present in approximately 5–10% of women of reproductive age and causes pelvic pain and infertility. The pathophysiology is not completely understood, but retrograde menstruation and deficiency in natural killer (NK) cells that clear endometriotic cells in the peritoneal cavity play an important role. Nowadays, hormonal therapy and surgery to remove endometriosis lesions are used as treatment. However, these therapies do not work for all patients, and hormonal therapy prevents patients from getting pregnant. Therefore, new treatment strategies should be developed. Since the cytotoxicity of NK cells is decreased in endometriosis, we performed a literature search into the possibility of NK cell therapy. Available treatment options include the inhibition of receptor–ligand interaction for KIR2DL1, NKG2A, LILRB1/2, and PD-1/PD-L1; inhibition of TGF-β; stimulation of NK cells with IL-2; and mycobacterial treatment with BCG. In preclinical work, these therapies show promising results but unfortunately have side effects, which have not specifically been studied in endometriosis patients. Before NK cell treatment can be used in the clinic, more research is needed.  相似文献
