True constant strain rate tensile tests of polypropylene up to 2% initial strain were performed to investigate the strain rate dependency of the initial stress–strain gradient dσ/dε with the aid of “slack grip.” The experimental results fairly agree with even the simplest mechanical model analysis with three parameters, showing the conspicuous strain rate dependency on the initial stress–strain gradient dσ/dε. The limiting cases corresponding to zero strain rate and strain rate of infinite magnitude, both predicted by the analysis, were experimentally verified.  相似文献

A general and simple implementation of simultaneous multiparametric sensing in a single microchip is presented by using a capillary-assembled microchip (CAs-CHIP) integrated with the plural different reagent-release capillaries (RRCs), acting as various biochemical sensors. A novel "drop-and-sip" technique of fluid handling is performed with a microliter droplet of a model sample solution containing proteases (trypsin, chymotrypsin, thrombin, elastase) and divalent cations (Ca2+, Zn2+, Mg2+) that passes through the microchannel with the aid of a micropipette as a vacuum pump, concurrently filling each RRC via capillary force. To avert the evaporation of the nanoliter sample volume in each capillary, PDMS oil is dropped on the outlet hole of the CAs-CHIP exploiting the capillary force that results in spontaneous sealing of all the RRCs. In addition, this high-speed sample introduction alleviates the possibility of protein adsorption and capillary cross-contamination, allowing a reliable and multianalyte determination of a sample containing many different proteases and divalent cations by using the fluorescence image analysis. Presented results suggested the possible application of this microchip in the field of drug discovery and systems biology.  相似文献

Two distinct marine organisms, diatoms and sponges, deposit dissolved silicates to construct highly architectural and species-specific body supports. Several factors such as proteins, long-chain polyamines (LCPAs), or polypeptides modified with LCPAs are known to be involved in this process. The LCPAs contained in the silica walls of diatoms are thought to play pivotal roles in the silica deposition. In sponges, however, a protein called silicatein and several other proteins have been reported to be the factors involved in the silica deposition. However, no other factors involved in this process have been reported. We have identified the LCPAs from the marine sponge Axinyssa aculeata and present here some evidence that sponge-derived LCPAs can deposit silica and that the LCPA derivatives are associated with spicules. The results indicate a common chemistry between sponges and diatoms, the two major players in the biological circulation of silicon in the marine environment. A wide variety of organisms are known to utilize silica in their biological processes. Polyamines or other functional molecules might be involved, in combination with proteins, in their biosilicification process.  相似文献

In order to develop life assessment techniques for aged components made of modified 9Cr–1Mo steel, specimens were artificially deteriorated by aging, creep and fatigue tests at elevated temperatures, and associated changes in the microstructure and mechanical properties were examined. It was observed that aging resulted in formation of Laves phase causing a decrease in toughness. The creep damage in base metal could be correlated with decrease in hardness, while creep damage in weldments could be correlated with the area fraction and density of creep voids. Creep rupture in weldments occurred in the fine-grained heat affected zone by the formation and growth of creep voids. The fatigue damage in base metal correlated to the maximum length of a crack among micro-cracks initiated during fatigue cycles.  相似文献

The mechanism of intracellular calcium ions (Ca(2+)) increase in chicken myoblasts was studied using histological, immunohistochemical, immunoblotting and Ca(2+) imaging techniques. Mononuclear myoblasts at embryonic day 12 (E12) contained myofibrils in the peripheral cytoplasm, and the sarcoplasmic reticulum was observed in the cytoplasm. Several Ca(2+)-related receptors, namely acetylcholine (ACh) receptors, dihydropyridine receptors (DHPRs) and ryanodine receptors (RyRs), were detected in the tissue as early as E12. Western blotting analyses detected one band corresponding to RyR subtype 3 (RyR3) at E12 and two bands corresponding to RyR1 and RyR3 after E13. Ca(2+) imaging of mononuclear myoblasts in vitro revealed an intense Ca(2+)-increase response to ACh stimulation, and this effect was abolished after EGTA addition to the culture medium. Nifedipine treatment also led to a lack of Ca(2+) increase in response to ACh stimulation, while ryanodine treatment led to a weak Ca(2+)-increase response. On the other hand, multinuclear myoblasts showed a Ca(2+)-increase response to ACh stimulation in the presence of not only EGTA but also nifedipine, although ryanodine treatment led to a lack of Ca(2+) increase. These results suggest that the mechanism of Ca(2+) increase in mononuclear myoblasts involves extracellular Ca(2+) entry through DHPRs, which is amplified by Ca(2+) release from the cytoplasmic Ca(2+) store, while multinuclear myoblasts mainly depend on Ca(2+) release from the cytoplasmic Ca(2+) store.  相似文献

Layer-by-layer (LBL) self-assembled multilayer films of gold nanoparticles (AuNPs) on a silicon wafer were demonstrated to be promising substrates for surface-assisted laser desorption/ionization (SALDI) mass spectrometry (MS) of peptides and environmental pollutants for the first time. LBL multilayer films, (AuNPs/PAHC)n, consisting of alternating layers of ammonium citrate capped AuNPs and poly(allylamine hydrochloride) (PAHC) were prepared on a silicon surface. Silicon plates with aggregated AuNPs were more suitable than those with dispersed AuNPs for the SALDI-MS of peptides. The number of particle layers had a significant effect on the laser desorption/ionization of angiotensin I; the peak intensity of the peptide (molecular ion amount) increased with an increase in the number of layers of AuNPs. As a result, the (AuNPs/PAHC)5 multilayer films increased the sensitivity of the angiotensin I to subfemtomoles and raised the useful analyte mass range, thus making it possible to detect small proteins (a 12 kDa cytochrome c). The signal enhancement when using (AuNPs/PAHC)5 may be due to (i) the high absorption of the UV laser light at 337 nm by the AuNP layers, (ii) the low thermal conductivity due to the AuNPs being covered with a thin monolayer of PAHC, and (iii) the increase in the surface roughness (approximately 100 nm) with the number of AuNP layers. Thus, laser-induced rapid high heating of AuNPs for effective desorption/ionization of peptides is possible. In addition, it was found that (AuNPs/PAHC)5 could be used to extract environmental pollutants (pyrene and dimethyldistearylammonium chloride) from very dilute aqueous solutions with concentrations less than 10(-10) mg/mL, and the analytes trapped in the LBL film could be identified by introducing the film directly into the SALDI mass spectrometer without needing to elute the analytes out of the film.  相似文献

Compatibility with Digital Versatile Disc and Compact Disc has been demonstrated for two options of high-density optical disk systems using a blue laser diode. Option 1 (HD DVD) employs a 0.65 numerical aperture objective lens and a 0.6 mm thick protective layer disk, while Option 2 (Blu-ray Disc) employs a 0.85 numerical aperture objective lens and a 0.1 mm thick protective layer disk. An optical head has three laser diodes whose wavelengths are 405 nm, 660 nm, and 785 nm. A spherical aberration caused by the difference in the protective layer thickness of the disk is corrected by an objective lens magnification change, while the numerical aperture of the objective lens is controlled by a wavelength-selective aperture. Experimental results have shown that Option 1 is more preferable, but Option 2 is also acceptable from the viewpoint of the compatibility.  相似文献

Low-resistivity indium tin oxide [ITO] film was successfully deposited on oxygen plasma-treated polyethylene terephthalate [PET] surfaces at room temperature. X-ray diffraction [XRD] measurements demonstrated that the film deposited on the PET surface that had not been treated with oxygen plasma had an amorphous structure. In contrast, after the low-power oxygen plasma treatment of the PET surface, the ITO film deposited on the PET surface had a poly-crystalline structure due to interactions between electric dipoles on the PET surface and electric dipoles in the ITO film. The minimum resistivity of the poly-crystalline ITO was about 3.6 times lower than that of the amorphous ITO film. In addition, we found that the resistivity of ITO film is proportional to the intensity of the (400) line in the film's XRD spectra.  相似文献

The purpose of this study is to predict the turbulent scalar flux at a free surface subject to a fully developed turbulent flow based on a hydrodynamic analysis of turbulence in the region close to the free surface. The effect of the Reynolds number on turbulent scalar transfer mechanisms is extensively examined. A direct numerical simulation technique is applied to achieve the purpose. The surface‐renewal approximation is used to correlate the free‐surface hydrodynamics and scalar transport at the free surface. Two types of characteristic time scales have been examined for predicting turbulent scalar flux. One is the time scale derived from the characteristic length and velocity scale at the free surface. The other is the reciprocal of the root‐mean‐square surface divergence. The results of this study show that scalar transport at the free surface can be predicted successfully using these time scales based on the concept of the surface‐renewal approximation. © 2012 American Institute of Chemical Engineers AIChE J, 2012  相似文献
