A standard procedure for the enumeration of heterotrophic bacteria, utilizing heterotrophic plate count medium (m-HPC), has been modified to allow bacterial ammonifier populations also to be counted. Nessler's reagent was included as an outside indicator to ascertain what portion of the population is capable of ammonification. Typically the ammonifier population varied among aquatic habitats, ranging from 0.08 to 37.0% of the heterotrophic population in river water samples, 0.2 to 10.6% for irrigation water samples, and 1.4 to 3.0% for pond and lake water samples.  相似文献

Motor neuron disease (MND) comprises a group of fatal neurodegenerative diseases with no effective cure. As progressive motor neuron cell death is one of pathological characteristics of MND, molecules which protect these cells are attractive therapeutic targets. Accumulating evidence indicates that EphA4 activation is involved in MND pathogenesis, and inhibition of EphA4 improves functional outcomes. However, the underlying mechanism of EphA4’s function in MND is unclear. In this review, we first present results to demonstrate that EphA4 signalling acts directly on motor neurons to cause cell death. We then review the three most likely mechanisms underlying this effect.  相似文献

Optical microelectromechanical system pressure sensors based on the principle of Fabry-Perot interferometry have been developed and fabricated using the technique of silicon-to-silicon anodic bonding. The pressure sensor is then integrated onto an optical fiber by a novel technique of anodic bonding without use of any adhesives. In this anodic bonding technique we use ultrathin silicon of thickness 10 microm to bond the optical fiber to the sensor head. The ultrathin silicon plays the role of a stress-reducing layer, which helps the bonding of an optical fiber to silicon having conventional wafer thickness. The pressure-sensing membrane is formed by 8 microm thick ultrathin silicon acting as a membrane, thus eliminating the need for bulk silicon etching. The pressure sensor integrated onto an optical fiber is tested for static response, and experimental results indicate degradation in the fringe visibility of the Fabry-Perot interferometer. This effect was mainly due to divergent light rays from the fiber degrading the fringe visibility. This effect is demonstrated in brief by an analytical model.  相似文献

We introduce a new chemical vapor deposition (CVD) process that can be used to selectively deposit materials of many different types. The technique makes use of the plasmon resonance in nanoscale metal structures to produce the local heating necessary to initiate deposition when illuminated by a focused low-power laser. We demonstrate the technique, which we refer to as plasmon-assisted CVD (PACVD), by patterning the spatial deposition of PbO and TiO(2) on glass substrates coated with a dispersion of 23 nm gold particles. The morphology of both oxide deposits is consistent with local laser-induced heating of the gold particles by more than 150 degrees C. We show that temperature changes of this magnitude are consistent with our analysis of the heat-loss mechanisms. The technique is general and can be used to spatially control the deposition of virtually any material for which a CVD process exists.  相似文献

Intervention technologies including ionizing radiation, antimicrobials, and modified atmospheres (MA) can be used to inhibit the growth of or inactivate foodborne pathogens on complex ready-to-eat foods such as sandwiches. However, the effect of these technologies when used in combination (the hurdle concept) on the survival of foodborne pathogens is unknown. The ability of ionizing radiation to inactivate Escherichia coli O157:H7, Salmonella, Listeria monocytogenes, and Staphylococcus aureus inoculated onto a frankfurter on a roll product containing the antimicrobials sodium diacetate and potassium lactate in the presence of an MA (100% N2, 50% N2 plus 50% CO2, or 100% CO2) was investigated. The radiation resistances (D10-values) of the foodborne pathogens were 0.43 to 0.47 kGy for E. coli O157:H7, 0.61 to 0.71 kGy for Salmonella, 0.53 to 0.57 for L. monocytogenes, and 0.56 to 0.60 for S. aureus. The MA had no effect on the radiation resistance of the pathogens. During a 2-week storage period under mild temperature abuse (10 degrees C), none of the pathogens were able to proliferate on the frankfurter on a roll product, regardless of the MA used. However, application of sublethal doses of ionizing radiation resulted in increased mortality of the gram-positive pathogens L. monocytogenes and S. aureus during the storage period regardless of the MA. Although the pathogens were unable to proliferate on the frankfurter on a roll product during the storage period, application of a postpackaging intervention step was needed to actually inactivate the foodborne pathogens. Ionizing radiation used in combination with sodium diacetate and potassium lactate resulted in additional mortality of L. monocytogenes and S. aureus, independent of the MA, during the 2-week storage period.  相似文献

Zinc-based glass polyalkenoate cements have been synthesised and their potential use in orthopaedic applications investigated. Zinc ions were released from the materials in a rapid burst over the first 24 h after synthesis, with the release rate falling below detectable levels after 7 days. Cement-implanted bone samples were prepared and the released zinc was shown, using energy dispersive X-ray analysis, to penetrate from the cement into the adjacent bone by up to 40 μm. Finally, the cements exhibited antibacterial activity against Streptococcus mutans and Actinomyces viscosus that reflected the pattern of zinc release, with the inhibition of growth greatest shortly after cement synthesis and little or no inhibition measureable after 30 days.  相似文献

Structural components that operate at high temperatures (for example, turbine blades) rely on thermally grown oxide (TGO), commonly alumina, for corrosion protection. Strains that develop in TGOs during operation can reduce the protectiveness of the TGO. However, the occurrence of growth strains in TGOs, and mechanisms that cause them, are poorly understood. It is accepted that compressive strains can develop as oxygen and metal atoms meet to form new growth within constrained oxide. More controversial is the experimental finding that large tensile stresses, close to 1 GPa, develop during isothermal growth conditions in alumina TGO formed on a FeCrAlY alloy. Using a novel technique based on synchrotron radiation, we have confirmed these previous results, and show that the tensile strain develops as the early oxide, (Fe,Cr,Al)(2)O(3), converts to alpha-Al2O3 during the growth process. This allows us to model the strain behaviour by including creep and this diffusion-controlled phase change.  相似文献

Being able to control the behaviour of osteoblast-like cells on a surface may provide a genuine insight into the material surface characteristics and help in creating a successful coating/cell interface. The possibility of creating a micro-environment that can induce proliferation, differentiation and mineralisation of bone cells in vitro, by successfully combining both chemistry and topography of a micro-fabricated substrate is an area that requires a multi-disciplinary approach. Utilising sputter deposition, a process that lends itself to high processability, in conjunction with photolithography allowing for the creation of highly repeatable etched surfaces, we aim to provide a successful combination of chemistry and topography. Correlating the substrate conditions with resultant osteoblast biological function and activity can ultimately be used with a view to modulating the behavior of osteoblast-like cells in vitro.  相似文献
