ABSTRACT: Set yogurts were prepared from fortified milk subjected to the following processes: (a) Thermal process (85C,°C,35 min), (b) High hydrostatic pressure process (193 or 676 MPa for 5 or 30 min), and (c) Nonprocessed milk (control). Yogurts from milk treated with 676 MPa for 30 min exhibited similar yield stress and water-holding capacity (WHC) to yogurts from heated milk. Transmission Electron Microscopy (TEM) micrographs exhibited small round casein micelle aggregates without appendages in yogurts from heated milk. Yogurts from milk treated with 193 MPa and untreated milk exhibited low yield stress, low WHC, and large clusters of coalesced micelles. Mechanisms for gel formation are discussed.  相似文献

The objectives of this study were to determine the effects on milk yield, milk composition, ruminal fermentation and total tract nutrient utilization of feeding roasted whole sunflower seed to dairy cows. Three diets were formulated: a control diet with no sunflower seed (NSF), a raw sunflower seed diet (USF) and a roasted sunflower seed diet (RSF). The level of sunflower seed in USF and RSF was 78 g kg?1 of dry matter (DM). The effects of dietary treatments on yield and composition of milk were determined using nine Holstein cows in three 3 × 3 Latin squares. Three ruminally fistulated cows were used to determine the effects of dietary treatments on ruminal fermentation and total tract nutrient digestibilities. Cows fed sunflower seed diets consumed 8% less (P < 0.05) DM but produced similar amounts of milk as cows fed NSF. However, milk fat content (30.7 vs 33.5 g kg?1) and yield (1.33 vs 1.47 kg day?1) were lower (P < 0.05) for cows fed USF and RSF than for those fed NSF. Supplemental sunflower seed had no effect on concentrations and yields of other milk components. The concentrations of short‐chain (C4:0 to C12:0) and medium‐chain (C14:0 to C16:0) fatty acids were, respectively, 27% and 29% lower (P < 0.05) while those of long‐chain fatty acids (C18:0 to C18:3) were 51% higher (P < 0.05) in the milk of cows fed USF and RSF than for cows fed NSF. Ruminal pH, ammonia N and total volatile fatty acids were not affected by dietary treatments. Feeding sunflower seed (USF or RSF) reduced (P < 0.05) the concentration of acetate and increased (P < 0.05) the concentration of propionate. Total tract nutrient digestibilities were not affected by sunflower seed supplementation or by heat treatment. Supplementing dairy cow diets with unheated or roasted sunflower seed improved the efficiency of milk production and increased concentrations of long‐chain and polyunsaturated fatty acids. Feeding sunflower seed at up to 78 g kg?1 of diet DM had no adverse effects on nutrient utilization. Roasting had no additional benefits on milk yield or milk fatty acid composition. Essential oils obtained by simultaneous distillation–extraction (SDE) from leaves, petioles and roots of three types of parsley (turnip‐rooted, plain leaf and curly leaf type), sown on three different dates, were analysed by GC‐MS (gas chromatography–mass spectrometry) analysis. Parsley plants were found to produce mainly β‐phellandrene, 1,3,8‐p‐menthatriene, α‐,p‐dimethylstyrene, myristicin, β‐myrcene and apiole. In some cases α‐ and β‐pinene were also found, whereas β‐elemene was detected, especially in the curly leaf type. The growth stage, plant tissue and date of sowing, as well as the climate conditions, all had a significant effect on the essential oil composition by altering the ratio of the above substances. 本实验在体外应用脂多糖(lipopolysaccharides,LPS)诱导小鼠腹腔巨噬细胞建立炎症模型的基础上,探讨沙葱总黄酮水洗组分的体外抗炎活性。应用CCK-8法检测0、50、100、200、400、800 μg/mL沙葱总黄酮水洗组分对小鼠腹腔巨噬细胞增殖活力的影响;将细胞分为空白对照组、LPS应激模型组及不同质量浓度沙葱总黄酮水洗组分预处理组,采用Griess法及酶联免疫吸附测定法分别测定各处理组细胞上清液中NO浓度和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素(interleukin,IL)-1β、IL-6、IL-10的质量浓度,反转录实时荧光定量聚合酶链式反应法检测各处理组细胞中诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)、TNF-α、IL-1β、IL-6、IL-10、髓样分化蛋白(myeloid differential protein,MyD)88、核因子 κB(nuclear factor κB,NF-κB)mRNA的表达水平。结果显示:与对照组相比,沙葱总黄酮水洗组分在50~800 μg/mL时对小鼠腹腔巨噬细胞无明显细胞毒性作用。与LPS应激模型组相比,沙葱总黄酮水洗组分能够极显著抑制促炎介质NO、TNF-α、IL-1β、IL-6质量浓度及其mRNA的表达(P<0.01或P<0.001);高度显著提高抗炎细胞因子IL-10质量浓度及其mRNA的表达(P<0.001),且呈剂量依赖效应;极显著降低MyD88、NF-κB mRNA的表达水平(P<0.01或P<0.001)。由此得出,沙葱总黄酮水洗组分对LPS诱导的小鼠腹腔巨噬细胞具有抗炎作用,其抗炎活性可能是通过抑制促炎性介质NO、TNF-α、IL-1β、IL-6的分泌并提高抗炎性细胞因子IL-10的质量浓度实现的,其作用机制可能与NF-κB信号通路有关。  相似文献

We compared total mercury concentrations ([Hg]) among 6 forage fish species in 25 central Canadian lakes and related [Hg] to adjusted-delta15N (an index of trophic position), delta13C, growth rate, and a suite of environmental variables. Growth rates were also compared among species and related to environmental variables. We found that rainbow smelt (Osmerus mordax), a recent invader in many of these lakes, had intermediate [Hg] and growth rates relative to other species. Forage fish growth rates differed significantly among species and were related to latitude (inverse relationship) and lake conductivity (positive relationship). Mercury concentrations also differed significantly among species and the strongest predictors were growth rate and lake conductivity; [Hg] was significantly and negatively related to both. Adjusted-delta15N explained very little variation in [Hg] and was significant only when the analysis was restricted to biotic variables. These results indicate that biomagnification may not be observed at fine scales of trophic differentiation and that rainbow smelt are unlikely to cause post-invasion [Hg] increases in most predatory fish species.  相似文献

以三孢布拉氏霉为菌种,对其发酵制备的番茄红素菌体粉的经口急性毒性、遗传毒性及短期毒性等进行了研究。结果表明:番茄红素菌体粉经口最大耐受量(MTD)大于18g·kg-1,毒性分级为无毒级;Ames实验、小鼠骨髓细胞染色体畸变实验、小鼠精子畸形实验均表明番茄红素菌体粉无明显遗传毒性;30d短期喂养各组小鼠无异常症状,无死亡,其对小鼠的体重增长、血常规、血生化、脏器系数均无显著的影响。揭示了番茄红素菌体粉有较好的安全性,为番茄红素菌体粉直接应用成保健食品等提供理论依据。  相似文献

Abstract: Anthocyanins, natural plant pigments in the flavonoid group, are responsible for the red color and some of the nutraceutical benefits of raspberries. This study explores anthocyanin degradation in frozen and freeze‐dried raspberries during storage in relation to glass transition temperatures. Frozen raspberries were stored at ?80, ?35, and ?20 °C, while freeze‐dried raspberries were stored at selected water activity (aw) values ranging from 0.05 to 0.75 at room temperature (23 °C) for more than a year. The characteristic glass transition temperatures (Tg) of raspberries with high water content and glass transition temperature (Tg) of raspberries with small water content were determined using a differential scanning calorimeter. The pH differential method was used to determine the quantity of anthocyanins in frozen and freeze‐dried raspberries at selected time intervals. The total anthocyanins in raspberries fluctuated during 378 d of storage at ?20 and ?35, and ?80 °C. Anthocyanin degradation in freeze‐dried raspberries ranged from 27% to 32% and 78% to 89% at aw values of 0.05 to 0.07 and 0.11 to 0.43, respectively, after 1 y. Anthocyanins were not detectable in freeze‐dried raspberries stored at aw values of 0.53 to 0.75 after 270 d. First order and Weibull equations were used to fit the anthocyanin degradation in freeze‐dried raspberries. The 1st‐order rate constant (k) of anthocyanin degradation ranged from 0.003 to 0.023 days?1 at the selected water activities. Significant anthocyanin degradation occurred in both the glassy and rubbery states of freeze‐dried raspberries during long‐term storage. However, the rate of anthocyanin degradation in freeze‐dried raspberries stored in the glassy state was significantly smaller than the rate of anthocyanin degradation in the rubbery state.  相似文献

The flow mechanism of particulate polymers during processing is reviewed. The extrusion of starch-g-poly(methyl acrylate) polymers is a good example of this type of processing, since graft copolymers retain their granular appearance and do not melt, even at high temperature, due to the rigid starch matrix. The formation of a continuous plastic by extrusion is accomplished by the sintering of heat-deformed granules in the high-pressure zone of the extruder die. Photomicrographs confirm this mode of formation. Plots of flow rate through the extruder die versus pressure in the die are presented. Tensile strengths and percent elongation of extruded specimens are not greatly affected by variations in flow rate. The equilibrium moisture content (about 5 percent) of starch-g-poly(methyl acrylate) plasticizes the starch component and thus reduces the operating pressure in the die. Tensile strengths, however, are unaffected by the presence of equilibrium moisture. Final traces of residual acid (resulting from initiator solution) are difficult to remove from starch-g-poly(methyl acrylate), and prolonged heating of the graft copolymer in the presence of this acid results in polymer degradation.  相似文献

D-Glucal and a series of substituted derivatives have been tested as substrates, inhibitors and inactivators of the Agrobacterium faecalis beta-glucosidase in order to probe structure/function relationships in this enzyme. D-Glucal is shown to be a substrate (kcat = 2.3 min-1, Km = 0.85 mM) undergoing hydration with stereospecific protonation from the alpha-face to yield 2-deoxy-beta-D-glucose. 1-Methyl-D-glucal surprisingly serves as only a poor substrate (kcat = 0.056 min-1, Km = 57 mM), also undergoing protonation from the alpha-face. 2-Fluoro-D-glucal, however is completely inert, as a result of inductive destabilisation of the oxocarbenium ion-like transition state for protonation, and functions only as a relatively weak (Ki = 24 mM) inhibitor. Similar behaviour was seen with almond beta-glucosidase and yeast alpha-glucosidase and for the interaction of 2-fluoro-D-galactal with Escherichia coli beta-galactosidase. A series of of alpha, beta-unsaturated glucal derivatives was also synthesised and tested as potential substrates, inhibitors or inactivators of A. faecalis beta-glucosidase. Of these only 1-nitro-D-glucal functioned as a time dependent, irreversible inactivator (ki = 0.011 min-1, Ki = 5.5 mM), presumably acting as a Michael acceptor. Electrospray mass spectrometric analysis revealed multiple labeling of the enzyme by this inactivator, lessening its usefulness as an affinity label. Less reactive Michael acceptor glycals which might have been more specific (1-cyano-, 2-cyano-, 1-carboxylic acid, 1-carboxylic acid methyl ester) unfortunately did not function as inactivators or substrates, only as relatively weak reversible inhibitors (Ki = 3-96 mM).  相似文献
