Optics clustered to output unique solutions (OCTOPUS) is a microscopy platform that combines single molecule and ensemble imaging methodologies. A novel aspect of OCTOPUS is its laser excitation system, which consists of a central core of interlocked continuous wave and pulsed laser sources, launched into optical fibres and linked via laser combiners. Fibres are plugged into wall-mounted patch panels that reach microscopy end-stations in adjacent rooms. This allows multiple tailor-made combinations of laser colours and time characteristics to be shared by different end-stations minimising the need for laser duplications. This setup brings significant benefits in terms of cost effectiveness, ease of operation, and user safety. The modular nature of OCTOPUS also facilitates the addition of new techniques as required, allowing the use of existing lasers in new microscopes while retaining the ability to run the established parts of the facility. To date, techniques interlinked are multi-photon/multicolour confocal fluorescence lifetime imaging for several modalities of fluorescence resonance energy transfer (FRET) and time-resolved anisotropy, total internal reflection fluorescence, single molecule imaging of single pair FRET, single molecule fluorescence polarisation, particle tracking, and optical tweezers. Here, we use a well-studied system, the epidermal growth factor receptor network, to illustrate how OCTOPUS can aid in the investigation of complex biological phenomena.  相似文献

This article provides an introduction to the usage of software engineering approaches and tools for the design of user experience (UE). This approach was developed for three main reasons: (a) to provide UE design teams a rigorous, methodical, encompassing method that can be applied throughout the entire product development cycle; (b) to provide designers a way to work with abstract concepts to establish the conceptual design of the system before progressing to the realization of the design to the appropriate platform; and (c) to communicate the intended design to the implementation team in a manner that is concise, complete, and familiar. The approach is the application of Unified Modeling Language concepts to describe users and the UE design rather than the technical architecture. It is believed that designing the UE design separately from the technical architecture will help ensure teams deliver a usable and delightful UE. The methods in this article have been applied to a handful of real-world projects in the last year and a half. Thus far, the application of software engineering approaches and tools have been found to be very powerful aids that have enabled the creation of highly useful Web experiences as well as software applications.

Our intent is to address the full gamut of issues involved in creating a total user experience (UE)-from discovery of users needs through high-level and low-level design, ensuring fidelity during development, and deployment, with a full assessment of how well the design meets users' needs. We believe that through a concerted use of such a rigorous approach we can greatly improve not only the UE of products we create, but also the experience of designing and developing them. We are not the only team taking this direction in UE design. It has become clear to many in our field that a more rigorous, thorough, and accurate approach to UE design is not only possible-it is necessary, for many of the same reasons that software engineers long ago embraced such practices. As we are evolving our approaches in other phases, this article focuses on the discovery and design phase activities.  相似文献

The 'end use' of water is a breakdown of the total household water usage such as water used for toilets, showers, washing machines, taps, lawn watering, etc. Understanding end uses of water will enable water planners, water authorities and household owners determine where water is used/wasted, how much and how often. This paper describes the end uses of water from a number of single-family homes in Greater Melbourne, Australia. The study involves the analysis of water consumption data recorded at 5-s intervals from logged households collected by Yarra Valley Water in Melbourne in 2004. The study determines how much water is used for outdoor and indoor purposes in a single-family home in Melbourne and compares the water usage during winter and summer. Hourly patterns of major end uses of water are also developed. The aim of this study is to improve the understanding of the end uses of water and to assist where to focus water conservation efforts that would be most effective financially and environmentally, and be acceptable to everyone.  相似文献

The Moderate-Resolution Imaging Spectroradiometer (MODIS) sensors on NASA's Terra and Aqua satellites image most of the Earth multiple times each day, providing useful data on fires that cannot be practically acquired using other means. Unfortunately, current fire products from MODIS and other sensors leave large uncertainties in measurements of fire sizes and temperatures, which strongly influence how fires spread, the amount and chemistry of their gas and aerosol emissions, and their impacts on ecosystems. In this study, we use multiple endmember spectral mixture analysis (MESMA) to retrieve subpixel fire sizes and temperatures from MODIS, possibly overcoming some limitations of existing methods for characterizing fire intensities such as estimating the fire radiative power (FRP). MESMA is evaluated using data from the Advanced Spaceborne Thermal Emission and Reflection Radiometer (ASTER) to assess the performance of FRP and MESMA retrievals of fire properties from a simultaneously acquired MODIS image, for a complex of fires in Ukraine from August 21, 2002. The MESMA retrievals of fire size described in this paper show a slightly stronger correlation than FRP does to fire pixel counts from the coincident ASTER image. Prior to this work, few studies, if any, had used MESMA for retrieving fire properties from a broad-band sensor like MODIS, or compared MESMA to higher-resolution fire data or other measures of fire properties like FRP. In the future, MESMA retrievals could be useful for fire spread modeling and forecasting, reducing hazards that fires pose to property and health, and enhancing scientific understanding of fires and their effects on ecosystems and atmospheric composition.  相似文献

In this article, we discuss experiences and lessons learned from deploying four mobile sensing applications on off-the-shelf mobile phones within a recreational framework called MobSens that contains elements of health, social, and environmental sensing at both individual and community levels. We describe the main components of our applications, which facilitate logging and external communications. We also outline the challenges faced when building and testing these applications and describe our strategies for overcoming them.  相似文献

Visual localization systems that are practical for autonomous vehicles in outdoor industrial applications must perform reliably in a wide range of conditions. Changing outdoor conditions cause difficulty by drastically altering the information available in the camera images. To confront the problem, we have developed a visual localization system that uses a surveyed three‐dimensional (3D)‐edge map of permanent structures in the environment. The map has the invariant properties necessary to achieve long‐term robust operation. Previous 3D‐edge map localization systems usually maintain a single pose hypothesis, making it difficult to initialize without an accurate prior pose estimate and also making them susceptible to misalignment with unmapped edges detected in the camera image. A multihypothesis particle filter is employed here to perform the initialization procedure with significant uncertainty in the vehicle's initial pose. A novel observation function for the particle filter is developed and evaluated against two existing functions. The new function is shown to further improve the abilities of the particle filter to converge given a very coarse estimate of the vehicle's initial pose. An intelligent exposure control algorithm is also developed that improves the quality of the pertinent information in the image. Results gathered over an entire sunny day and also during rainy weather illustrate that the localization system can operate in a wide range of outdoor conditions. The conclusion is that an invariant map, a robust multihypothesis localization algorithm, and an intelligent exposure control algorithm all combine to enable reliable visual localization through challenging outdoor conditions. © 2009 Wiley Periodicals, Inc.  相似文献

Despite the high number of total hip arthroplasty (THA) procedures performed each year, there is no common consensus on the best surgical approach. Gait is known to improve following THA although it does not return to what is typically quantified as normal, and surgical approach is believed to be a contributing factor. The current study evaluates postoperative hip function and provides an objective assessment following two common surgical approaches: the McFarland-Osborne direct lateral and the southern posterior. Faced with the common problem of providing an objective comparison from the wealth of data collected using motion analysis techniques, the current study investigates the application of an objective classification tool to provide information on the effectiveness of each surgery and to differentiate between the characteristics of hip function following the two approaches. Seven inputs for the classifier were determined through statistical analysis of the biomechanical data. The posterior approach group exhibited greater characteristics of non-pathological gait and displayed a greater range of functional ability as compared with the lateral approach cohort. The classification tool has proved to be successful in characterizing non-pathological and THA function but was insufficient in distinguishing between the two surgical cohorts.  相似文献

Eight anticonvulsant drugs-including clonazepam, diazepam and phenobarbital-were tested for their effects on GABA-stimulated chloride uptake in rat cerebral cortical microsacs (unfiltered synaptoneurosomes). "Mid" and "high" therapeutic concentrations were screened, and, if significant enhancement was found, full concentration-response tests were done. In the initial screens, enhancement of GABA-stimulated uptake was found only with phenobarbital, clonazepam and diazepam. In subsequent concentration-response tests, the effects of phenobarbital were found to occur throughout the range of normal, anticonvulsant concentrations, whereas the effects of clonazepam and diazepam were observed only above the concentrations normally used for the chronic control of seizures or anxiety. These data suggest that phenobarbital's anticonvulsant effects are mediated via the GABAA receptor complex, but that the low-dose effects of the benzodiazepines may be mediated via some other mechanism.  相似文献

Faults and geological barriers can drastically affect the flow patterns in porous media. Such fractures can be modelled as interfaces that interact with the surrounding matrix. We propose a new technique for the estimation of the location and hydrogeological properties of a small number of large fractures in a porous medium from given distributed pressure or flow data. At each iteration, the algorithm builds a short list of candidates by comparing fracture indicators. These indicators quantify at the first order the decrease of a data misfit function; they are cheap to compute. Then, the best candidate is picked up by minimization of the objective function for each candidate. Optimally driven by the fit to the data, the approach has the great advantage of not requiring remeshing, nor shape derivation. The stability of the algorithm is shown on a series of numerical examples representative of typical situations.  相似文献
