One of the most important steps in the process of viral infection is a fusion between cell membrane and virus, which is mediated by the viral envelope glycoprotein. The study of activity of the glycoprotein in the post-fusion state is important for understanding the progression of infection. Here we present a first real-time kinetic study of the activity of gp41 (the viral envelope glycoprotein of human immunodeficiency virus—HIV) and its two mutants in the post-fusion state with nanometer resolution by atomic force microscopy (AFM). Tracking the changes in the phosphatidylcholine (PC) and phosphatidylcholine–phosphatidylserine (PC:PS) membrane integrity over one hour by a set of AFM images revealed differences in the interaction of the three types of protein with zwitterionic and negatively charged membranes. A quantitative analysis of the slow kinetics of hole formation in the negatively charged lipid bilayer is presented. Specifically, analysis of the rate of roughness change for the three types of proteins suggests that they exhibit different types of kinetic behavior.  相似文献

n‐Si/CnH2n + 1/Hg junctions (n = 12, 14, 16 and 18) can be prepared with sufficient quality to assure that the transport characteristics are not anymore dominated by defects in the molecular monolayers. With such organic monolayers we can, using electron, UV and X‐ray irradiation, alter the charge transport through the molecular junctions on n‐ as well as on p‐type Si. Remarkably, the quality of the self‐assembled molecular monolayers following irradiation remains sufficiently high to provide the same very good protection of Si from oxidation in ambient atmosphere as provided by the pristine films. Combining spectroscopic (UV photoemission spectroscopy (UPS), X‐ray photoelectron spectroscopy (XPS), Auger, near edge‐X‐ray absorption fine structure (NEXAFS)) and electrical transport measurements, we show that irradiation induces defects in the alkyl films, most likely C?C bonds and C? C crosslinks, and that the density of defects can be controlled by irradiation dose. These altered intra‐ and intermolecular bonds introduce new electronic states in the highest occupied molecular orbital (HOMO)–lowest unoccupied molecular orbital (LUMO) gap of the alkyl chains and, in the process, dope the organic film. We demonstrate an enhancement of 1–2 orders of magnitude in current. This change is clearly distinguishable from the previous observed difference between transport through high quality and defective monolayers. A detailed analysis of the electrical transport at different temperatures shows that the dopants modify the transport mechanism from tunnelling to hopping. This study suggests a way to extend significantly the use of monolayers in molecular electronics.  相似文献

In this paper, we define a new problem that has not been addressed in the past: the trade-off between energy efficiency and throughput for multicast services in 802.16e or similar mobile networks. In such networks, the mobile host can reduce its energy consumption by entering the sleep mode when it is not supposed to receive or transmit information. For unicast applications, the tradeoff between delay and energy efficiency has been extensively researched. However, for mobile hosts running multicast (usually push- based) applications, it is much more difficult to determine when data should be transmitted by the base station and when each host should enter the sleep mode. In order to maximize the channel throughput while limiting the energy consumption, a group of hosts needing similar data items should be active during the same time intervals. We define this as an optimization problem and present several algorithms for it. We show that the most efficient solution is the one that employs cross-layer optimization by dividing the hosts into groups according to the quality of their downlink physical (PHY) channels.  相似文献

Population-based data suggest that individuals who consume large dietary amounts of n-3 polyunsaturated fatty acids (PUFA) have lower odds of peripheral artery disease (PAD); however, clinical studies examining n-3 PUFA levels in patients with PAD are sparse. The objective of this study is to compare erythrocyte membrane fatty acid (FA) content between patients with PAD and controls. We conducted a cross-sectional study of 179 vascular surgery outpatients (controls, 34; PAD, 145). A blood sample was drawn and the erythrocyte FA content was assayed using capillary gas chromatography. We calculated the ratio of the n-3 PUFA eicosapentaenoic acid (EPA) to the n-6 PUFA arachidonic acid (ARA) as well as the omega-3 index (O3I), a measure of erythrocyte content of the n-3 PUFA, EPA, and docosahexaenoic acid (DHA), expressed as a percentage of total erythrocyte FA. Compared with controls, patients with PAD smoked more and were more likely to have hypertension and hyperlipidemia (p < 0.05). Patients with PAD had a lower mean O3I (5.0 ± 1.7% vs 6.0 ± 1.6%, p < 0.001) and EPA:ARA ratio (0.04 ± 0.02 vs 0.05 ± 0.05, p < 0.001), but greater mean total saturated fats (39.5 ± 2.5% vs 38.5 ± 2.6%, p = 0.01). After adjusting for several patient characteristics, comorbidities, and medications, an absolute decrease of 1% in the O3I was associated with 39% greater odds of PAD (odds ratio [OR] 1.39, 95% confidence interval [CI] 1.03–1.86, and p = 0.03). PAD was associated with a deficiency of erythrocyte n-3 PUFA, a lower EPA:ARA ratio, and greater mean total saturated fats. These alterations in FA content may be involved in the pathogenesis or development of poor outcomes in PAD.  相似文献

The potential for mineral scaling that may limit the generation of new potable water resources by reverse osmosis (RO), from inland brackish water of high gypsum scaling propensity, was experimentally explored via flux decline measurements and real-time RO membrane surface imaging. Antagonistic gypsum and calcium carbonate scaling kinetics were demonstrated for high-sulfate brackish water desalting. RO scaling studies with brackish water from the California San Joaquin Valley (approximately 10 000 mg/L total dissolved solids) revealed that membrane gypsum scaling was increasingly retarded with rising bicarbonate concentrations. Crystal growth rate, fractional membrane scale coverage, and flux decline decreased by up to about 63, 78, and 73%, respectively, as the bicarbonate concentration increased, at the membrane surface, from < 0.01 to 7.81 mM, for a gypsum saturation index of 2. Inhibition of gypsum crystal growth was attributed to bicarbonate adsorption onto the crystal surfaces, and CaCO3 scaling was undetected even up to a calcite saturation index of approximately 16. Given the suppression of gypsum scaling by bicarbonate, it is essential to considerthis effect in the conventional practice of pH adjustment to suppress CaCO3 scaling. The present results suggest that antagonistic and synergistic mineral crystallization kinetics effects are important for optimizing scale-control strategies (e.g., acid and antiscalants addition to the RO feed).  相似文献

Although concentrated animal feeding operations constantly generate physiologically active steroidal hormones, little is known of their environmental fate. Estrogen and testosterone concentrations in groundwater and their distribution in sediments below a dairy-farm wastewater lagoon were therefore determined and compared to a reference site located upgradient of the farm. Forward simulations of flow as well as estrogen and testosterone transport were conducted based on data from the sediment profile obtained during drilling of a monitoring well belowthe dairy-farm waste lagoon. Testosterone and estrogen were detected in sediments to depths of 45 and 32 m, respectively. Groundwater samples were directly impacted by the dairy farm, as evidenced by elevated concentrations of nitrate, chloride, testosterone, and estrogen as compared to the reference site. Modeling potential transport of hormones in the vadose zone via advection, dispersion, and sorption could not explain the depths at which estrogen and testosterone were found, suggesting that other transport mechanisms influence hormone transport under field conditions. These mechanisms may involve interactions between hormones and manure as well as preferential flow paths, leading to enhanced transport rates. These types of interactions should be further investigated to understand the processes regulating hormone transport in the subsurface environment and parametrized to forecast long-term fate and transport of steroidal hormones.  相似文献

Transfer of chemicals from contaminated surfaces such as foliage, floors, and furniture is a potentially significant source of both occupational exposure and children's residential exposure. Increased understanding of relevant factors influencing transfers from contaminated surfaces to skin and resulting dermal-loading will reduce uncertainty in exposure assessment. In a previously reported study, a fluorescence imaging system was developed, tested, and used to measure transfer of riboflavin residues from surfaces to hands. Parameters evaluated included surface type, surface loading, contact motion, pressure, duration, and skin condition. Results of the initial study indicated that contact duration and pressure were not significant for the range of values tested, but that there are potentially significant differences in transfer efficiencies of different compounds. In the study reported here, experimental methods were refined and additional transfer data were collected. A second fluorescent tracer, Uvitex OB, with very different physicochemical properties than riboflavin, was also evaluated to better characterize the range of transfers that may be expected for a variety of compounds. Fluorescent tracers were applied individually to surfaces and transfers to skin were measured after repeated hand contacts with the surface. Additional trials were conducted to compare transfer of tracers and co-applied pesticide residues. Results of this study indicate that dermal loadings of both tracers increase through the seventh brief contact. Dermal loading of Uvitex tends to increase at a higher rate than dermal loadings of riboflavin. Measurement of co-applied tracer and pesticide suggest results for these two tracers may provide reasonable bounding estimates of pesticide transfer.  相似文献

Upholstered furniture is often manufactured with polyurethane foam (PUF) containing flame retardants (FRs) to prevent the risk of a fire and/or to meet flammability regulations, however, exposure to certain FRs and other chemicals have been linked to adverse health effects. This study developed a new methodology for evaluating volatile organic compound (VOC) and FR exposures to users of upholstered furniture by simulating use of a chair in a controlled exposure chamber and assessing the health significance of measured chemical exposure. Chairs with different fire-resistant technologies were evaluated for VOC and FR exposures via inhalation, ingestion, and dermal contact exposure routes. Data show that VOC exposure levels are lower than threshold levels defined by the US and global indoor air criteria. Brominated FRs were not detected from the studied chairs. The organophosphate FRs added to PUF were released into the surrounding air (0.4 ng/m3) and as dust (16 ng/m2). Exposure modeling showed that adults are exposed to FRs released from upholstered furniture mostly by dermal contact and children are exposed via dermal and ingestion exposure. Children are most susceptible to FR exposure/dose (2 times higher average daily dose than adults) due to their frequent hand to mouth contact.  相似文献

When major incident emergencies occur in the UK (e.g., a terrorist attack), senior representatives convene from local emergency services, civil resource organizations, health agencies, and government. These groups make decisions that minimize the societal and economic impacts of major incidents. Their adherence to UK doctrine was assessed using video footage of 18 groups responding to a major incident in a simulation suite, and six groups responding to a large‐scale exercise. There were marked between‐group differences in the sequencing of decision‐making activities, and limited consideration of alternative courses of action. These results provide an impetus for future policy, guidance, and training to address (a) between‐group inconsistency in decision processes and (b) the lack of consideration of alternative courses of action.  相似文献
