High-speed GC with time-of-flight (TOF) MS detection is used for the characterization and analysis of oils rendered from the peel of five diverse species of orange including bergemot orange, bitter orange, tangerine, mandarin orange, and sweet orange. With a user-defined signal-to-noise threshold of 100, 44 peaks were found and 36 compounds identified in the various oils. Some major constituent components show large concentration ranges over the five species. A 14-m-long, 0. 18-mm-i.d. column ensemble consisting of 7.0-m lengths of a trifluoropropylmethyl polysiloxane and a 5% phenyl dimethyl polysiloxane column was temperature-programmed at 50 degrees C/min starting at the time of injection to achieve analysis times under 140 s. The TOFMS was operated with a spectral acquisition rate of 25 spectra/s, and automated peak finding software successfully found all of the components, with the exception of one severely overlapping peak pair in bitter-orange oil. Of the 44 peaks, 25 were identified by use of a TOFMS library created for this study; another 11 were identified with a commercial terpene library, and 8 were not identified. A quantitative comparison (percent of total peak area) is presented for 16 components, which comprise 98.8-99.5% of the total peak area for the five orange species. Stop-flow operation of the column ensemble is used to enhance selectivity for targeted component pairs to facilitate single-channel detection for QA/ QC analysis of characterized samples and to enhance column selectivity for TOFMS characterization in cases in which peak overlap is so severe that only a single peak is observed.  相似文献

This paper reports the first development of high-performance, silicon-glass micro-gas chromatography (/spl mu/GC) columns having integrated heaters and temperature sensors for temperature programming, and integrated pressure sensors for flow control. These 3-m long, 150-/spl mu/m wide and 250-/spl mu/m deep columns, integrated on a 3.3 cm square die, were fabricated using a silicon-on-glass dissolved wafer process. Demonstrating the contributions to heat dissipation from conduction, convection, and radiation with and without packaging, it is shown that using a 7.5-mm high atmospheric pressure package reduces power consumption to about 650 mW at 100/spl deg/C, while vacuum packaging reduces the steady-state power requirements to less than 100 mW. Under vacuum conditions, 600 mW is needed for a temperature-programming rate of 40/spl deg/C/min. The 2300 ppm//spl deg/C TCR of the temperature sensors and the 50 fF/kPa sensitivity of the pressure sensors satisfy the requirements needed to achieve reproducible separations in a /spl mu/GC system. Using these columns, highly resolved 20-component separations were obtained with analysis times that are a factor of two faster than isothermal responses.  相似文献

The development of lightweight, portable GC instrumentation is handicapped by the need for compressed carrier gas to drive the separation. The use of air as carrier gas eliminates the need for compressed gas tanks. If a vacuum pump is used to pull carrier gas and injected samples through the column, atmospheric pressure air can be used as carrier gas. Vacuum outlet operation also improves performance for high-speed separations by reducing detector dead time and by shifting optimal carrier gas velocity to higher values. Under vacuum outlet conditions using atmospheric pressure air as carrier gas, a 6-m-long, 0.25-mm-i.d. capillary column can generate approximately 12,500 theoretical plates, and a 12-m-long column can generate approximately 44,000 plates but with a 3-4-fold increase in separation time. The principal issues in column selection for high-speed GC with air as a carrier gas are efficiency and stability. Several bonded and nonbonded stationary phases were evaluated for use with air as carrier gas in the analysis of volatile organic compounds of interest in airmonitoring applications. These include dimethylpolysiloxane, 50% phenyl-50% methyl polysiloxane, 50% cycanopropylphenyl-50% methyl polysiloxane, trifluoropropyl polysiloxane, poly(ethylene glycol), and dicyanoallyl polysiloxane (nonbonded). The dimethyl polysiloxane and the trifluoropropyl polysiloxane columns showed good efficiency and no significant deterioration after 5 days of continuous operation with air as carrier gas. The 50% phenyl-50% methyl polysiloxane and the 50% cycanopropylphenyl-50% methyl polysiloxane columns showed poorer efficiency, and the poly(ethylene glycol) and dicyanoallyl polysiloxane columns showed excessive deterioration in air.  相似文献

In this paper we present an exact calculation of the tunnelling current for two different one-dimensional barrier potentials. The purpose is twofold: First, we demonstrate the feasibility of applying the new method of C. Noguera as a direct analytical tool for solving tunnelling problems in the case of independent electrons. In this approach, the tunnelling current is expressed in terms of the logarithmic derivative of the Green's function which must be evaluated at the cutting surface. This can be done analytically in the one-dimensional case. Secondly, we compare these results with the corresponding tunnelling current obtained using the Bardeen perturbation method, valid only in the large barrier limit. In particular, we investigate the use of the Bardeen approach in describing tunnelling spectroscopy. The dynamic conductance dI/dV, as a function of bias and barrier width, is compared in both cases.  相似文献

BACKGROUND: Tacrolimus (FK506) may represent a major advance in the management of allograft rejection after solid organ transplantation. In August 1994 a European heart transplantation pilot study was initiated to assess the efficacy and safety of tacrolimus when administered exclusively through an oral route. METHODS: Eighty-two heart transplant recipients were randomized to treatment (2:1 ratio) with either tacrolimus- (n=54) or cyclosporine-based therapy (n=28). RESULTS: No significant differences were evident between the two treatment groups in either rejection or survival rates at 1 year. Kaplan-Meier estimates of the freedom from rejection were 26.3% and 18.5%, respectively, for the tacrolimus and cyclosporine treatment groups (p=.444). Survival rates were 79.6% and 92.9% (p=.125). At 3 of the 5 centers, patients received antithymocyte globulin during the immediate postoperative period and fared better than those who did not (with acute rejection-free rates of 49.2% and 26.7% for tacrolimus and cyclosporine, respectively [p=.080], as opposed to 7.1% and 8.3% [p=.965]; patient survival rates of 84.6% and 93.3% [p=.382] vs 75.0% and 92.3% [p=.243]). The overall rates of infection, impaired renal function (31.5% vs 21.4%), and glucose intolerance (7.0% vs 4.3%) did not differ significantly between the tacrolimus and cyclosporine treatment groups. Tacrolimus seemed to possess an advantage with regard to a reduced requirement for antihypertensive therapy (59.5% vs 87.5%, p=.025). CONCLUSIONS: Immunosuppression with oral tacrolimus provides a viable alternative to treatment with cyclosporine, particularly when administered in conjunction with antibody therapy. Further studies are warranted to optimize the administration of tacrolimus in this indication.  相似文献

PURPOSE: To test the capabilities of vacuum core biopsy (VCB) in the diagnosis of mammographically indeterminate lesions. MATERIALS AND METHODS: 120 patients (131 lesions) were examined using VCB with 14G or 11G vacuum core. RESULTS: VCB was mostly performed because of indeterminate microcalcifications (67 cases) or soft tissue densities/architectural distortion (64 cases). 112 benign changes, 14 DCIS and 5 invasive carcinomas were found. Excellent accuracy was achieved (presently 100%), since complete excision of small lesions/areas (< or = 1 cm) or partial excision of larger lesions was possible. No relevant hematomas or infections occurred. Patients tolerated the painless procedure very well. DISCUSSION: This report confirms our previous experiences. This method promises to replace diagnostic open biopsy of indeterminate or suspicious nonpalpable lesions.  相似文献

The determination of diagnostic features in recorded heart sounds was investigated with Carpentier-Edwards (CE) bioprosthetic valves. Morphological features, extracted using the Choi-Williams distribution, achieved between 96 and 61% correct classification. The time-scale wavelet-transform feature set achieved 100% correct classification with native valve populations, and 87% with the CE replacements.  相似文献

Previously, we showed that truncated soluble forms of herpes simplex virus (HSV) glycoprotein D (gDt) bound directly to a truncated soluble form of the herpesvirus entry mediator (HveAt, formerly HVEMt), a cellular receptor for HSV. The purpose of the present study was to determine the affinity of gDt for HveAt by surface plasmon resonance and to compare and contrast the kinetics of an expanded panel of gDt variants in binding to HveAt in an effort to better understand the mechanism of receptor binding and virus entry. Both HveAt and gDt are dimers in solution and interact with a 2:1 stoichiometry. With HveAt, gD1(306t) (from the KOS strain of HSV-1) had a dissociation constant (KD) of 3.2 x 10(-6) M and gD2(306t) had a KD of 1.5 x 10(-6) M. The interaction between gDt and HveAt fits a 1:1 Langmuir binding model, i.e., two dimers of HveAt may act as one binding unit to interact with one dimer of gDt as the second binding unit. A gD variant lacking all signals for N-linked oligosaccharides had an affinity for HveAt similar to that of gD1(306t). A variant lacking the bond from cysteine 1 to cysteine 5 had an affinity for HveAt that did not differ from that of the wild type. However, variants with double cysteine mutations that eliminated either of the other two disulfide bonds showed decreased affinity for HveAt. This result suggests that two of the three disulfide bonds of gD are important for receptor binding. Four nonfunctional gDt variants, each representing one functional domain of gD, were also studied. Mutations in functional regions I and II drastically decreased the affinity of gDt for HveAt. Surprisingly, a variant with an insertion in functional region III had a wild-type level of affinity for HveAt, suggesting that this domain may function in virus entry at a step other than receptor binding. A variant with a deletion in functional region IV [gD1(Delta290-299t)] exhibited a 100-fold enhancement in affinity for HveAt (KD = 3.3 x 10(-8) M) due mainly to a 40-fold increase in its kinetic on rate. This agrees with the results of other studies showing the enhanced ability of gD1(Delta290-299t) to block infection. Interestingly, all the variants with decreased affinities for HveAt exhibited decreased kinetic on rates but only minor changes in their kinetic off rates. The results suggest that once the complex between gDt and HveAt forms, its stability is unaffected by a variety of changes in gD.  相似文献
