NMR studies have been carried out to determine the nature of the orientational ordering of N 2 in dilute solid N 2Ar mixtures. The orientational ordering for cubic lattices (x(N 2)50%) was compared to that observed for the hexagonal lattice structures for 57 < x(N 2) < 75%. The NMR line shapes for the cubic structures were found to be sharply defined and could only be explained in terms of a narrow distribution of order parameters. This is in contrast to the quadrupolar glass state with a very broad order parameter distribution observed for the hexagonal structures.  相似文献

Juvenile Eisenia fetida (Savigny) were exposed for 20 weeks to an uncontaminated soil and to soils contaminated with cadmium, copper, lead, and zinc collected from seven sites at different distances from a smelting works at Avonmouth, southwest England. The survival, growth (= weight after 5 weeks exposure), time to sexual maturation (= percentages of adults present after 8 weeks), and reproduction (= number of cocoons produced by the worms) were compared with soil metal concentrations. Of the parameters measured, growth and sexual maturation time had the lowest EC50 values. The effects of metal-contaminated soils could be attributed both to the direct toxicity of the metals and to changes in the "scope for growth" of the exposed worms. A comparison of the results with those of an earlier toxicity test conducted with adult worms indicated that juveniles are more sensitive to metals than adults. Significant toxic effects on the growth and sexual maturation times of juveniles were detected in soils from sites for which no significant effects on the cocoon production of adults could be detected. The greater sensitivity of juvenile worms indicates the importance of considering effects on a variety of life history stages when conducting a risk assessment of the effects of pollutants in soils. Although E. fetida does not occur naturally in soils at Avonmouth, the present study provides evidence to support the suggestion that pollution from the smelter is responsible for the absence of worms within 2 km of the factory. Results presented in this paper, and from previous studies, suggest the observed absence is due to the effects of zinc on the growth and maturation of juveniles and the cocoon production rate of adult worms.  相似文献

The objective of the research described was to devise an efficient procedure to cryopreserve in vitro-matured bovine oocytes, using in vitro fertilization (IVF) and development of resultant zygotes into blastocysts as criteria of oocyte survival. Oocytes at metaphase II were found to be extremely sensitive to chilling. Cooling them to O degrees C for as little as 5 sec significantly decreased their capability to cleave and develop further after IVF; after 80 sec at 0 degrees C, only approximately 10% of chilled oocytes developed into blastocysts. Oocytes were also adversely affected by brief exposures to 4 M and 5.5 M ethylene glycol (EG) solutions supplemented with sucrose; after being suspended in either of these EG solutions in plastic straws and plunged directly into liquid nitrogen (LN2), few of the oocytes were fertilized and developed. To "outrace" chilling injury, oocytes contained in < 1 microliter of EG solution were placed onto electron microscope grids and plunged directly into N2 slush or LN2. After such ultra-rapidly cooled oocytes were warmed, 30% of them cleaved after IVF, and half of these developed into blastocysts-- survival rates equivalent to those for oocytes that had been exposed to EG without any cooling. This method offers promise as a novel way to cryopreserve bovine oocytes.  相似文献

Atomic force microscopy (AFM) provides a unique opportunity to study live individual bacteria at the nanometer scale. In addition to providing accurate morphological information, AFM can be exploited to investigate membrane protein localization and molecular interactions on the surface of living cells. A prerequisite for these studies is the development of robust procedures for sample preparation. While such procedures are established for intact bacteria, they are only beginning to emerge for bacterial spheroplasts. Spheroplasts are useful research models for studying mechanosensitive ion channels, membrane transport, lipopolysaccharide translocation, solute uptake, and the effects of antimicrobial agents on membranes. Furthermore, given the similarities between spheroplasts and cell wall-deficient (CWD) forms of pathogenic bacteria, spheroplast research could be relevant in biomedical research. In this paper, a new technique for immobilizing spheroplasts on mica pretreated with aminopropyltriethoxysilane (APTES) and glutaraldehyde is described. Using this mounting technique, the indentation and cell elasticity of glutaraldehyde-fixed and untreated spheroplasts of E. coli in liquid were measured. These values are compared to those of intact E. coli. Untreated spheroplasts were found to be much softer than the intact cells and the silicon nitride cantilevers used in this study.  相似文献

A new spectrum-based model for describing the behavior of time-dependent materials is presented. In this paper, unlike most prior modeling techniques, the time-dependent response of viscoelastic materials is not expressed through the use of series. Instead, certain criteria have been imposed to select a spectrum function that has the potential of describing a wide range of material behavior. Another consequence of choosing the spectrum function of the type used in this paper is to have a few closed form analytic solutions in the theory of linear viscoelasticity. The Laplace transform technique is used to obtain the necessary formulae for viscoelastic Lame' functions, relaxation and bulk moduli, creep bulk and shear compliance, as well as Poisson's ratio. By using the Elastic–Viscoelastic Correspondence Principle (EVCP), material constants appearing in the proposed model are obtained by comparing the experimental data with the solution of the integral equation for a simple tensile test. The resulting viscoelastic functions describe the material properties which can then be used to express the behavior of a material in other loading configurations. The model's potential is demonstrated and its limitations are discussed.  相似文献

Self-piercing riveting as an alternative joining method to spot-welding has attracted considerable interest from the automotive industry and has been widely used in aluminium intensive vehicles. Pressing and stamping are important processes in automotive production and result in additional straining on the vehicle body sheet material. It is therefore important to have knowledge of the effect of sheet pre-straining on the quality of the self-piercing riveted joints and on the mechanical behaviour of the riveted aluminium alloy sheets. This paper reports the influence of sheet pre-straining on the static and fatigue behaviour of self-piercing riveted aluminium alloy sheet. Wrought aluminium alloy sheet, NG5754 with a nominal thickness value of 2 mm was used to obtain pre-strained NG5754 sheets with pre-straining levels of 3%, 5% and 10%. Pairs of pre-strained NG5754 sheets were joined to create single-riveted lap joints which subsequently underwent lap-shear and fatigue testing. Microscopic inspection showed that the joint quality was satisfactory despite the increasing sheet straining levels. The results showed that by increasing the pre-straining level up to 10%, the shear and fatigue strength also increased. The rate of increase of the static and fatigue strength differed as the pre-straining levels varied.  相似文献

Infant rats exhibit sensitive-period odor learning characterized by olfactory bulb neural changes and odor preference acquisitions critical for survival. This sensitive period is coincident with low endogenous corticosterone (CORT) levels and stress hyporesponsivity. The authors hypothesized that low corticosterone levels modulate sensitive-period learning. They assessed the effects of manipulating CORT levels by increasing and removing CORT during (Postnatal Day 8) and after (Postnatal Day 12) the sensitive period. Results show that (a) exogenous CORT prematurely ends sensitive-period odor-shock-induced preferences; (b) adrenalectomy developmentally extends the sensitive period as indicated by odor-shock-induced odor-preference learning in older pups, whereas CORT replacement can reinstate fear learning; and (c) CORT manipulation modulates olfactory bulb correlates of sensitive-period odor learning in a manner consistent with behavior. National traumatic events can produce extremely vivid memories. Using a questionnaire administered during telephone interviews, the authors investigated emotional responses to, and memory for. the September 11, 2001, terrorist attacks in patients with Alzheimer's disease (AD), patients with mild cognitive impairment (MCI), and healthy older adults in the initial weeks following the event and again 3-4 months later. There were several notable findings. First, patients with AD showed less memory than patients with MCI and older adults. Second, patients with AD, but not patients with MCI or older adults, appeared to retain more memory for personal versus factual information. Third, patients with AD and older adults did not differ in the intensity of their reported emotional responses to the attacks, whereas patients with MCI reported relatively less intense emotional responses.
