Application of the technique of specular neutron reflection to the study of adsorbed layer structure is illustrated for the case of β-casein at the air—water interface in the presence and absence of nonionic water—soluble surfactant C12E6. Guinier analysis of reflectivity data for a 5 × 10−3 wt% solution of pure β-casein in air-contrast-matched water (8 vol% D2O) at pH 7.0 gives a time-independent adsorbed amount of 2.05 ± 0.10 mg/m2 and an adsorbed layer thickness of 1.65 ± 0.07 nm; these values are found to increase quite substantially as the pH is reduced towards the protein's isoelectric point. In the presence of surfactant the loss of reflectivity is a direct measure of protein displacement from the interface because the hydrogenated surfactant is almost contrast matched to the aqueous phase. At a C12E6 concentration in the range 2–2.5 × 10−4 wt% (surfactant: protein molar ratio, R ≈ 2.2), there is roughly half the protein lost from the interface but little change in adsorbed layer thickness as inferred from the slopes of the Guinier plots. Protein is effectively completely removed from the surface for R 10. These results are in semi-quantitative agreement with complementary competitive adsorption data for β-casein in soya oil-in-water emulsions. 相似文献

Five good and four poor chymosincoagulating individual cow milk samples were analyzed for casein composition using hydroxyapatite chromatography and polyacrylamide gel electrophoresis to establish possible relationships between casein fractions and differences in coagulation properties.Samples exhibited wide variation in casein composition. Poor chymosincoagulating milk had higher content of γ- and degraded caseins and lower κ- and β-caseins than the good-coagulating milk. One milk sample that did not coagulate 30 min after chymosin addition had low αS-casein concentration and an additional major casein fragment (not identified). A substantial peak representing unidentified minor caseins was apparent in a poorcoagulating milk sample, which coagulated early but whose coagulum did not become firm in 30 min. Oil-in-water emulsions stabilized with nonionic emulsifiers change to water-in-oil emulsions as the temperature rises when the hydrophilic and lipophilic properties of the mixed emulsifier are just balanced. Preparation above the phase inversion temperature followed by rapid cooling yields emulsions that exhibit very fine droplet size and extreme long-term stability. Cosmetic emulsions were prepared by this phase inversion temperature (PIT) method using typical raw materials such as polar oils, e.g. decyl oleate, 2-octyl dodecanol or isopropyl myristate, and nonionic emulsifiers, e.g. ceteareth-12 or polyoxyethylene eicosyl/docosyl ether combined with cetostearyl alcohol as a co-emulsifier. The phase inversion temperature was measured as a function of the oil polarity and the concentration of mixed emulsifier. The relationship between phase inversion temperature, droplet size and emulsion stability was investigated. In addition, self-bodying agents such as cetostearyl alcohol or monoglycerides were added to these thin, fine disperse emulsions to adjust the consistency. Using gas chromatography/mass spectrometry (GC/MS), we investigated the formation of disinfection byproducts (DBPs) from high bromide waters (2 mg/L) treated with chlorine or chlorine dioxide used in combination with chlorine and chloramines. This study represents the first comprehensive investigation of DBPs formed by chlorine dioxide under high bromide conditions. Drinking water from full-scale treatment plants in Israel was studied, along with source water (Sea of Galilee) treated under carefully controlled laboratory conditions. Select DBPs (trihalomethanes, haloacetic acids, aldehydes, chlorite, chlorate, and bromate) were quantified. Many of the DBPs identified have not been previously reported, and several of the identifications were confirmed through the analysis of authentic standards. Elevated bromide levels in the source water caused a significant shift in speciation to bromine-containing DBPs; bromoform and dibromoacetic acid were the dominant DBPs observed, with very few chlorine-containing compounds found. Iodo-trihalomethanes were also identified, as well as a number of new brominated carboxylic acids and 2,3,5-tribromopyrrole, which represents the first time a halogenated pyrrole has been reported as a DBP. Most of the bromine-containing DBPs were formed during pre-chlorination at the initial reservoir, and were not formed by chlorine dioxide itself. An exception wasthe iodo-THMs, which appeared to be formed by a combination of chlorine dioxide with chloramines or chlorine (either added deliberately or as an impurity in the chlorine dioxide). A separate laboratory study was also conducted to quantitatively determine the contribution of fulvic acids and humic acids (from isolated natural organic matter in the Sea of Galilee) as precursor material to several of the DBPs identified. Results showed that fulvic acid plays a greater role in the formation of THMs, haloacetic acids, and aldehydes, but 2,3,5-tribromopyrrole was produced primarily from humic acid. Because this was the first time a halopyrrole has been identified as a DBP, 2,3,5-tribromopyrrole was tested for mammalian cell cytotoxicity and genotoxicity. In comparison to other DBPs, 2,3,5-tribromopyrrole was 8x, 4.5x, and 16x more cytotoxic than dibromoacetic acid, 3-chloro-4-(dichloromethyl)-5-hydroxy-2-[5H]-furanone [MX], and potassium bromate, respectively. 2,3,5-Tribromopyrrole also induced acute genomic damage, with a genotoxic potency (299 microM) similar to that of MX. 相似文献

Variations in texture were determined for 10 day aged raw and cooked Longissimus thoracis (LT) muscle from 436 bulls of 15 European cattle breeds slaughtered at an age of 13–16 months. Variations in texture were related to differences in pH 24 h post-mortem, sarcomere length, collagen characteristics and lipid content. The shear force of cooked meat samples varied from 43.8 to 67.4 N/cm2. Simmental, Highland and Marchigiana cattle had the highest shear force values and Avileña-Negra Ibérica, Charolais, Casina and Pirenaica cattle had the lowest values. Cooked meat toughness showed a weak negative correlation to lipid content (P < 0.001) but no correlation to collagen characteristics. Raw meat texture measured by compression correlated positively (P < 0.001) with total and insoluble collagen. In conclusion, collagen characteristics showed correlation to raw meat texture but not to cooked meat toughness of LT muscle in European young bulls. 相似文献

Eighty pigs, male and female littermate pairs, were fed a control or a test diet from 25 to 95 kg live weight. The diets, as fed, contained 15.5 g/kg linoleic acid (18:2) and 1.9 g/kg α-linolenic acid (18:3) (control) or 10 g/kg linoleic acid and 4 g/kg α-linolenic acid (test). The test diet, with added linseed, was, therefore, high in the main n-3 polyunsaturated fatty acid (PUFA) 18:3 and low in the main n-6 PUFA 18:2. Making this relatively small change led to a 56% increase in the content of 18:3 in muscle and major increases in the contents of the beneficial longer chain PUFAs EPA (20:5n-3) (100% increase) and DHA (22:6n-3) (35% increase) which are synthesised from 18:3n-3. Levels of EPA and DHA in pigmeat adipose tissue were also increased by the test diet. In liver, the test diet resulted in an 18:3 level 4× higher than in muscle, with 10× more EPA and 20× more DHA. Sausages, analysed after 6 months frozen storage also had high n-3 PUFA levels, due to the contribution of these fatty acids from both muscle and adipose tissue. From a health perspective these results confirm the potential of pigmeat to supply valuable n-3 PUFA to the human diet. The test diet produced a PUFA:saturated FA ratio in muscle of 0.4, close to the minimum recommended value for the diet as a whole and an n-6:n-3 ratio of 5, a significant improvement on the current average for pigmeat (7). It is estimated that the test diet would provide 12 g of long chain n-3 PUFA to the human diet per annum at current pigmeat consumption levels in the UK, about a third of that from oily fish. 相似文献

An increase in the intake of the n-3 series polyunsaturated fatty acids (PUFA) is recommended by nutritionists for the human diet and beef is a significant source of these fatty acids. Enhancing the n-3 PUFA content of beef is important in view of the generally saturated nature of fatty acids in ruminant meats and the potentially negative effect this can have on human health. This study examined the effects of breed and diet on the fatty acid composition of beef M. longissimus. Ninety-six steers were used, 48 Aberdeen Angus cross (AA) and 48 Holstein-Friesian (HF). At 6months of age, 3 groups were identified, to be slaughtered at 14, 19 and 24months, respectively. Each group consisted of eight steers of each breed fed on a concentrate or a grass silage diet, rich in n-6 and n-3 PUFA, respectively. The intake of the concentrate diet was restricted so that steers of each breed grew at a similar rate on each diet. The early maturing AA produced heavier, fatter carcasses with better conformation. Animals fed grass silage had higher carcass fatness and conformation scores and higher levels of neutral lipid and total lipid in muscle than those fed concentrate. When all animals were pooled, a decline in PUFA% as total muscle lipid increased was evident. Feeding a grass silage diet rich in α-linolenic acid (18:3n-3) increased levels of this fatty acid in muscle neutral lipid by a factor of about 3.0 compared with the concentrate diet, as well as enhancing the synthesis of the n-3 series long-chain C20-22 PUFA in the phospholipid fraction, including docosahexaenoic acid (DHA, 22:6n-3). In contrast, both levels and proportions of linoleic acid (18:2n-6) and the n-6 series C20-22 PUFA were higher in animals fed the concentrate diet. The proportions of 18:1trans and conjugated linoleic acid (CLA) in muscle neutral lipid were higher in animals fed concentrate compared with silage in all 3 groups. This was partly due to increased consumption of 18:2n-6. The ratio of PUFA to saturated fatty acids (P:S) in muscle was reduced by feeding grass silage, partly as the result of increased fat deposition. However, the increase in levels of n-3 series fatty acids with silage-feeding resulted in beneficially low n-6:n-3 ratios in muscle in all age groups (approximately 1.2 compared with 12.0 in the concentrate diet). Subtle breed differences in PUFA amounts and proportions were noted. Holstein-Friesians had higher proportions of PUFA and higher P:S ratios compared with AA, partly due to a higher proportion of phospholipid in total lipid. In phospholipid itself, HF in the 19 and 24months groups had higher proportions of most n-3 PUFA. In all age groups the ratio of DHA to its precursor, 18:3n-3 was higher in HF. 相似文献

A collaborative industry-government research program was carried out at the National Research Council Canada (NRC) to develop new sound-transmission-class and fire-resistance ratings for gypsum-board protected walls. Forintek Canada Corp. and the Canadian Wood Council participated in the program on behalf of Canada's wood industry. As a result of that NRC research program, sound-transmission-class (STC) and fire-resistance (FR) ratings were formulated for approximately 140 gypsum-protected wood-frame walls. Fire-resistance ratings for the walls range from 30 minutes to 2 hours. Sound-transmission-class ratings range from 30 to 65. Sound-transmission-class and fire-resistance ratings for approximately 90 of the wall designs appear in the 1995 edition of the National Building Code of Canada (NBCC). A new assay termed the dome disappearance method for classical swine fever virus (CSFV) using FS-L3 cells with serum-free culture medium was developed. The CSFV live vaccine GPE- strain grows well and shows a slight cytopathic effect (CPE) in FS-L3 cells. This CPE results in the disappearance of the unique fluid-filled multicellular domes on a single monolayer of FS-L3 cells. By using this phenomenon, dome disappearance, as a marker of infection, it was possible to determine the titers of CSFV and its neutralizing antibody. The virus titer determined by this method shows a good correlation with that determined by immunochemical and interference methods. Furthermore, the amount of neutralizing antibody measured by this method also correlated with that measured by the Exaltation of Newcastle Disease Virus (END) neutralizing method. The dome disappearance method developed in this experiment is a simple and safe procedure and has the great advantage that bovine serum, which may contain antibody against bovine viral diarrhea virus, is not necessary for the cultivation of FS-L3 cells. 相似文献
