The recently introduced monolithic silica columns were tested for their use in micellar liquid chromatography. Micellar methods are utilized in high-throughput quantitative structure-retention relationships to estimate an indicator of the membrane permeability of drugs, namely, the octanol-water partition coefficient, log P. The monolithic column's ability to function at higher flow rates might be useful to speed up these chromatographic methods estimating the log P. Therefore, the elution behavior of diverse basic pharmaceutical substances was determined on a classical particle-based and a monolithic column, both with and without a micellar medium in the mobile phase. Utilizing among others principal component analysis, the extent to which these methods differ in retention characteristics was examined in the context of high-throughput determination of log P. Results indicate that combining monolithic columns with micellar media leads to faster log P and possibly even better permeability predictions.  相似文献

There are two separate, and presumably opposing, processes that affect colony odor in the desert ant Cataglyphis niger: (1) biosynthesis and turnover of these chemicals by individual ants, and (2) homogenization of colony odor through exchange of cues. The first increases signal variability; the latter decreases it. The impact of these factors was tested by splitting colonies and monitoring the profile changes occurring in the postpharyngeal glands (PPG) and cuticular hydrocarbons.From each of two polygynous nests four daughter colonies were formed, three monogynous and one queenless. Thereafter, 10 ants from each were randomly selected each month, for three successive months, for analyses of their PPG and cuticular hydrocarbons. From two colonies we also obtained ants from a known matriline. Over time, there was a shift in hydrocarbon profiles of both the PPG and cuticular washes in each of the tested colonies. Moreover, by subjecting selected hydrocarbon constituents to a discriminant analyses based on their relative proportions, all of the daughter colonies (queenright and queenless) were distinguishable from each other and from their respective mother colonies. In each of the queenright daughter colonies, the queen profile was indiscriminable from that of the workers and often was in the center of the group. Full sisters were clearly distinguishable from their nestmates, emphasizing the genetic versus environmental processes that govern colony odor. The effect of time was always superior to the separation effect in contributing to odor segregation. Comparison of the Mahalanobis distances indicated that the shift in hydrocarbon seems to proceed along parallel lines rather than in divergence. However, there was no overt aggression between ants that originated from the different subgroups in dyadic encounters. It appears that in this species a three-month separation period is not sufficient to change the hydrocarbon profile beyond the recognition threshold.  相似文献

Human serum albumin (HSA) binds numerous molecules, among which are suitably designed MRI contrast agents. The rotational tumbling of the protein is thus one of the parameters likely to affect the in vivo relaxivity of these agents. Literature unveils discrepancies about the value of the rotational correlation time (τR) of HSA. In the present work, the τR of this protein has been determined by studying the deuterium relaxation rate of small molecules known for their strong binding to HSA (warfarin and 4-hydroxycoumarin). Values of approx. 20–22 ns are obtained at 310 K in a 4% HSA solution and are in good agreement with the theoretical predictions.  相似文献

The Failure Mode and Effect Analysis (FMEA) is a useful instrument born in the aerospace industry and widely used to improve a process or product's efficiency. Over the years, this instrument has been adopted in increasingly different contexts, such as HealthCare. This paper proposes an approach aimed at improving the defects typical of the classic FMEA in the design phase, that is, in a scenario full of uncertainties and with little information available, using a new hybrid Multiple-criteria decision-making (MCDM) method in order to obtain a priority index more performant than Risk Priority Number (RPN). In the proposed method, the three assessment criteria have a different weighting in the index's final computation, differently from the classical RPN. These weights are obtained with a scientific technique, thus avoiding that excessive subjectivity influences the final result. A more efficient priority index is obtained through a new hybrid approach that solves some classical RPN gaps. A case study concerning an endoscope Ear Nose Throat Entropy (ENT) prototype is examined to illustrate the proposed method. FMEA analysis in HealthCare is increasingly used for its flexibility and reliability. This study focuses on using new techniques to eliminate certain defects or exploit some qualities better. The use of a robust and elastic innovative MCDM method to calculate a new priority index and a scientific technique to obtain the weight of the selection are the interesting insights proposed in this paper.  相似文献

Surface properties of nanoparticles to be used for radioimmunotherapy need to be optimized to allow antibody conjugation while ensuring biocompatibility. We aimed to investigate cell adhesion and proliferation onto different coatings to be used for nanoparticles. C, CH x or SiO x coatings deposited onto glass coverslips by magnetron deposition as well as nitrogen functionalized materials synthetized using different reactive sputtering conditions and PPAA (plasma polymerized allylamine) coating, were compared. Amine functionalization did increase hydrophilicity in all the materials tested. Biocompatibility was assessed by measuring cell viability, morphology, attachment, spreading, and pro-inflammatory cytokine secretion. The results show that C and CN x were the most biocompatible substrates while SiO x and SiO x N y were the most toxic materials. PPAA coatings displayed unexpectedly an intermediate biocompatibility. A correlation could be observed between wettability and cell proliferation except for C coated surface, indicating that more complex processes than hydrophilicity alone are taking place that affect cell functions.  相似文献

Gabapentin affects the glutamate and gamma amino butyric acid (GABA) neurotransmitters through which it may facilitate smoking abstinence. To obtain preliminary estimates of efficacy of gabapentin for smoking cessation, we conducted a single-arm, open-label study of gabapentin, 1,800-mg/day administered in three equal divided doses for 8 weeks. A total of 50 adult smokers were enrolled. All participants received a brief behavioral intervention at each medication visit. A total of 37 participants completed all follow-up assessments. At end-of-treatment the biochemically confirmed point-prevalence and prolonged smoking abstinence rates were 28% (95% CI=16%-42%) and 24% (95% CI=13%-38%), respectively. At 6 months, the biochemically confirmed point-prevalence and prolonged smoking abstinence rates were 20% (95% CI=10%-34%) and 16% (95% CI=7%-29%), respectively. Among subjects who continued to smoke and completed the follow-up assessments, the reported number of cigarettes smoked per day (mean+/-standard deviation) was significantly less than at baseline: -10.0+/-8.2 (p<.001). Adverse effects were minor and well tolerated. Our results suggest that gabapentin may increase smoking abstinence. An adequately powered randomized clinical trial assessing different doses of this drug against a placebo would be the reasonable next step.  相似文献
