Combination therapy with intravesical bacillus Calmette-Guérin (BCG) plus IFN-alpha for superficial bladder cancer has been demonstrated to be more effective than either single agent alone in animal studies and of suggested greater efficacy in clinical studies. However, the mechanism by which IFN-alpha enhances BCG-mediated antitumor activity is poorly understood. Using PBMCs from bladder cancer patients, IFN-alpha was found to substantially enhance the efficacy of BCG to induce IFN-gamma production. Among 34 patients tested, 80% showed >4-fold increase. This effect of IFN-alpha was observed in both initial and memory responses to BCG. In addition, IFN-alpha up-regulated BCG-induced IL-12 and TNF-alpha and down-regulated BCG-induced IL-10. Neutralizing endogenous IL-10 or adding exogenous IL-12 provided further synergy for IFN-gamma production. In clinical practice, intravesical IFN-alpha 2B (50 million units (MU)/dose) was observed to accelerate urinary IFN-gamma production to low-dose BCG (one-tenth or one-third of a full dose) in patients treated with combination therapy compared with BCG alone. These results suggest that IFN-alpha is a potent BCG enhancer that polarizes the BCG-induced immune response toward the cellular immune pathway by promoting Th1 cytokine expression and reducing Th2 cytokine expression. This study provides an immunological basis for future rational use of IFN-alpha in conjunction with intravesical BCG for bladder cancer immunotherapy.  相似文献

This study investigated the effects of organic and colloidal fouling on the removal of a representative micropollutant sulphamethoxazole by two commercially available NF membranes. Alginate, bovine serum albumin and colloidal silica were selected as model foulants to simulate hydrophilic and hydrophobic organic fractions, and colloidal matter that are often found in treated effluent and surface water. Membrane fouling was related to the membrane and foulant characteristics and subsequently the separation behaviour of the micropollutant sulphamethoxazole under different solution pH. On the basis of these results, it was confirmed that membrane fouling is strongly dependent on both the foulant and membrane characteristics. The complex relationship among retention mechanisms, fouling mechanisms and the effects of fouling on retention was systematically delineated. Of the three model foulants selected for this study, colloidal fouling resulted in the most significant reduction in retention of sulphamethoxazole as well as inorganic salts, while flux decline as a result of colloidal fouling was quite moderate. Reduction in retention caused by fouling was attributed to a phenomenon known as cake-enhance concentration polarisation, which was a predominant mechanism of colloidal fouling. In addition, the reported results suggested that the effect of fouling on retention is also membrane pore size dependent.  相似文献

We have developed a new detection method for measuring distributions of energy depositions from particles characterized by low linear energy transfers (LETs). In particular, we have developed a detection system to measure energy depositions produced by electrons and photons on nanometer scales. The detector is based upon the operational principles of the superheated drop detector (SDD). SDDs consist of tiny droplets of superheated liquid suspended within a gel-like emulsion. The SDDs in this study are fabricated using Freon-115 and a glycerol-based gel as the superheated liquid and host medium. This SDD configuration is operated as a threshold temperature-dependent detector for measuring nanoscopic distributions of energy depositions from low-LET particles. Measured results are compared to the calculated distributions of energy depositions along an electron track. A new electron track code, ESLOW3.1, is used to perform the computational study. Measurements show good agreement with computational results in the energy deposition range of 40-200 eV.  相似文献

Multi-Camera Human Activity Monitoring   总被引:1,自引:0,他引:1  
With the proliferation of security cameras, the approach taken to monitoring and placement of these cameras is critical. This paper presents original work in the area of multiple camera human activity monitoring. First, a system is presented that tracks pedestrians across a scene of interest and recognizes a set of human activities. Next, a framework is developed for the placement of multiple cameras to observe a scene. This framework was originally used in a limited X, Y, pan formulation but is extended to include height (Z) and tilt. Finally, an active dual-camera system for task recognition at multiple resolutions is developed and tested. All of these systems are tested under real-world conditions, and are shown to produce usable results. A kinematic method is developed to determine the shakedown limits of elastic-perfectly plastic steel-reinforced concrete beams under quasi-static and dynamic cyclic loads. This procedure, like the respective kinematic approach of plastic limit analysis, is visual and easy to use in engineering applications. The load parameters (amplitudes, frequency) versus yield moment diagrams, constructed from possible collapse modes, should assist in choosing the reinforcement scheme and amounts of reinforcement to meet the load bearing requirements for the structures.  相似文献
