The granule-bound starch synthase I (GBSSI) encoded by the waxy gene is responsible for amylose synthesis in the endosperm of wheat grains. In the present study, a novel Wx-B1 null mutant line, M3-415, was identified from an ethyl methanesulfonate-mutagenized population of Chinese tetraploid wheat landrace Jianyangailanmai (LM47). The gene sequence indicated that the mutated Wx-B1 encoded a complete protein; this protein was incompatible with the protein profile obtained using sodium dodecyl sulfate–polyacrylamide gel electrophoresis, which showed the lack of Wx-B1 protein in the mutant line. The prediction of the protein structure showed an amino acid substitution (G470D) at the edge of the ADPG binding pocket, which might affect the binding of Wx-B1 to starch granules. Site-directed mutagenesis was further performed to artificially change the amino acid at the sequence position 469 from alanine (A) to threonine (T) (A469T) downstream of the mutated site in M3-415. Our results indicated that a single amino acid mutation in Wx-B1 reduces its activity by impairing its starch-binding capacity. The present study is the first to report the novel mechanism underlying Wx-1 deletion in wheat; moreover, it provided new insights into the inactivation of the waxy gene and revealed that fine regulation of wheat amylose content is possible by modifying the GBSSI activity. 相似文献

CFD results are presented for a curved aircap used for coating interior surfaces such as engine cylinder bores. The device
analyzed is similar to the Metco diamond jet rotating wire torch, but wire feed is not simulated. The feed gases are injected
through an axisymmetric nozzle into the curved aircap. Argon is injected through the center of the nozzle. Premixed propylene
and oxygen are introduced from an annulus in the nozzle, while cooling air is injected between the nozzle and the interior

To improve the physicomechanical properties of jute yarn, grafting with 1,6‐hexanediol diacrylate (HDDA) monomer was performed by a UV radiation technique. A series of HDDA solutions of various concentrations in methanol were prepared. A small quantity of photoinitiator (Darocur‐1664) was also added to HDDA solutions. To optimize the conditions for grafting, the effects of monomer concentration, soaking time, and radiation doses were studied by varying the number of soaking times along with variation of monomer concentrations and UV radiation intensities. The extent of polymer loading and the mechanical properties like tensile strength (TS), elongation at break (Eb), and tensile modulus of both treated and untreated jute were investigated. The highest tensile strength, polymer loading, and modulus were achieved with 5% HDDA concentration, 5 min soaking time, and the 4th pass of UV radiation. This set of conditions was selected as optimum and produced enhanced tensile strength (67%), modulus (108%), and polymer loading (11%) over those of virgin fiber. To further improve the mechanical properties the jute yarns were pretreated with alkali (5% NaOH) solution and after that the alkali‐treated yarn were treated under UV radiation of various intensities. The pretreated samples were grafted with optimized monomer concentration (5% HDDA). Increased properties of alkali + UV‐pretreated and grafted samples such as polymer loading (12%), tensile strength (103%), elongation at break (46%), and modulus (114%) were achieved over those of virgin jute yarn. © 2004 Wiley Periodicals, Inc. A theoretical investigation has been conducted for the prediction of the critical height at the onset of gas entrainment during single discharge from a stratified, two‐phase region, through a branch installed on an inclined flat wall. The predicted critical height at the onset of gas entrainment was proven to be a function of Froude number (Fr) and density ratio of the interface fluids. Three different experimental data sets at wall inclination angles of zero, 45 and 90 degrees (i.e. side, inclined and bottom branches) were used for comparisons. This paper deals with influence of chitosan nanoparticles (CNPs) loaded by tetracycline, as a drug, on the physico-mechanical and antibacterial properties as well as drug release behavior of poly(vinyl alcohol), PVA, hydrogels prepared by electron beam irradiation. The formation of spherical chitosan particles in nanoscale size prepared by an ionic gelation method was confirmed by FTIR and UV spectroscopy, and scanning electron microscopy analyses. The drug release kinetic studies from drug loaded chitosan nanoparticles (DLCNPs) at pH = 7.4 revealed a linear and steady release behavior over long period of time. The theoretical analysis of the swelling kinetic data, using Peppas’s model showed that the swelling kinetic is governed by Fickian diffusion for all the prepared hydrogels, however, the water diffusion coefficient, and therefore, the swelling content were lower for the hydrogels loaded with DLCNPs as compared to the ones with the neat drug. In agreement with these results, the hydrogels containing DLCNPs exhibited a more controlled drug release behavior with significantly stronger antibacterial activity. The tensile mechanical properties of the hydrogels not affected by the DLCNPs were found to be suitable for wound dressing applications. 相似文献

A series of new non-ionic amphiphiles based on bis-galactobenzimidazolones have been synthesized by grafting alkyl bis-benzimidazolone units as hydrophobic tails on hydroxypropyloxygalacto-pyranose moieties as hydrophilic heads. Their surface and self-aggregation properties in water were evaluated. The compounds show very low critical micellar concentrations (CMCs) that decrease with increasing chain length; values for the minimal area per molecule at the interface (A min) follow the same trend. The synthesized compounds also form hexagonal liquid crystals in water for a certain range of hydrophobic tail lengths. On the other hand, the new amphiphiles show characteristic UV?CVis absorption and fluorescence emission bands associated with the benzimidazolone moiety. The fluorescence emission is quenched with a certain degree of selectivity by cations, due to their strong affinity towards the benzimidazolone group, which shows ion complexation properties. Hence, the reported new amphiphiles are candidates as self-assembling chemosensors. The quenching efficiency and also ion sensing sensitivity is higher in the monomeric state as compared to the micellar state. The fluorescence emission intensity is higher for compounds with a shorter alkyl chain. 相似文献

Azo-bis(isobutyroamidoxime) was synthesized and used as functionalized initiator to prepare a liquid isoprene bearing amidoxime end groups via radical polymerization. The polymer was characterized by FT-IR, Gel Permeation Chromatography and NMR. In particular, the structure of the polymer was investigated using 1D and 2D NMR techniques. The distribution of the different isoprene units (1,4-trans, 1,4-cis, 1,2 and 3,4), as well as the structure of the amidoxime end-groups was determined. It was found that the structure of the end groups was governed by the steric hindrance of the initiator. Rice straw (Rs)/polypropylene (PP) composites were prepared in the different ratio of 5 : 95, 10 : 90, 15 : 85, 20 : 80, 25 : 75, and 30 : 70 (Rs wt % : PP wt %) by an injection molding process. This work investigated the tensile strength (TS), bending strength (BS), and impact strength (IS) of the composites. From the results, it is observed that Rs20 : PP80 mixture composite showed better performance with mechanical properties (TS = 26.2 MPa, BS = 58 N/mm2, and IS = 1.7 KJ/mm2) among the composites prepared. Two hybrid composites were also fabricated using 20% Rs, 10% seaweed with 70% PP and 20% Rs, 30% seaweed with 70% PP. In between the two hybrid composites, superior mechanical behavior showed by the hybrid composite in ratio of Rs20 : Sw10 : PP70 with enhanced results such as TS = 28 MPa, BS = 68 N/mm2, and IS = 2.5 KJ/mm2.
