SARS-CoV-2 variants raise concern because of their high transmissibility and their ability to evade neutralizing antibodies elicited by prior infection or by vaccination. Here, we compared the neutralizing abilities of sera from 70 unvaccinated COVID-19 patients infected before the emergence of variants of concern (VOCs) and of 16 vaccine breakthrough infection (BTI) cases infected with Gamma or Delta against the ancestral B.1 strain, the Gamma, Delta and Omicron BA.1 VOCs using live virus. We further determined antibody levels against the Nucleocapsid (N) and full Spike proteins, the receptor-binding domain (RBD) and the N-terminal domain (NTD) of the Spike protein. Convalescent sera featured considerable variability in the neutralization of B.1 and in the cross-neutralization of different strains. Their neutralizing capacity moderately correlated with antibody levels against the Spike protein and the RBD. All but one convalescent serum failed to neutralize Omicron BA.1. Overall, convalescent sera from patients with moderate disease had higher antibody levels and displayed a higher neutralizing ability against all strains than patients with mild or severe forms of the disease. The sera from BTI cases fell into one of two categories: half the sera had a high neutralizing activity against the ancestral B.1 strain as well as against the infecting strain, while the other half had no or a very low neutralizing activity against all strains. Although antibody levels against the spike protein and the RBD were lower in BTI sera than in unvaccinated convalescent sera, most neutralizing sera also retained partial neutralizing activity against Omicron BA.1, suggestive of a better cross-neutralization and higher affinity of vaccine-elicited antibodies over virus-induced antibodies. Accordingly, the IC50: antibody level ratios were comparable for BTI and convalescent sera, but remained lower in the neutralizing convalescent sera from patients with moderate disease than in BTI sera. The neutralizing activity of BTI sera was strongly correlated with antibodies against the Spike protein and the RBD. Together, these findings highlight qualitative differences in antibody responses elicited by infection in vaccinated and unvaccinated individuals. They further indicate that breakthrough infection with a pre-Omicron variant boosts immunity and induces cross-neutralizing antibodies against different strains, including Omicron BA.1.  相似文献

The field of single-cell analysis has advanced rapidly in the last decade and is providing new insights into the characterization of intercellular genetic heterogeneity and complexity, especially in human cancer. In this regard, analyzing single circulating tumor cells (CTCs) is becoming particularly attractive due to the easy access to CTCs from simple blood samples called “liquid biopsies”. Analysis of multiple single CTCs has the potential to allow the identification and characterization of cancer heterogeneity to guide best therapy and predict therapeutic response. However, single-CTC analysis is restricted by the low amounts of DNA in a single cell genome. Whole genome amplification (WGA) techniques have emerged as a key step, enabling single-cell downstream molecular analysis. Here, we provide an overview of recent advances in WGA and their applications in the genetic analysis of single CTCs, along with prospective views towards clinical applications. First, we focus on the technical challenges of isolating and recovering single CTCs and then explore different WGA methodologies and recent developments which have been utilized to amplify single cell genomes for further downstream analysis. Lastly, we list a portfolio of CTC studies which employ WGA and single-cell analysis for genetic heterogeneity and biomarker detection.  相似文献

This paper presents an in-depth and processual case study of a major infrastructural innovation project involving diverse private and public-sector organizations. The case study shows how organizing developed recursively in response to diverging temporal requirements, induced by the temporal institutional complexity facing the project. We introduce the idea of temporal conditioning to demonstrate how large-scale temporary organizations dynamically cope with conflicting temporal institutional requirements by making use of three strategies: (1) temporal avoidance, (2) temporal splitting, and (3) temporal matching. With its focus on the emergence of the project, this paper adds to our understanding of the dynamics of organizing in temporary and institutionally pluralistic settings – settings that put greater pressures on our ability to deal with conflicting institutional requirements pertaining to time and timing. Accordingly, we offer a new perspective on the dynamics of large-scale projects and how they respond to a particular kind of institutional complexity.  相似文献

This article draws on material gathered from Swedish tweets about the Roma population, in order to map different discourses. Based on this material, a legal analysis was made focusing on how the legal protection under the European Convention on Human Rights (ECHR) and Swedish law for such types of expression may vary depending on the wider discursive context. This article concludes that the legal protection for hateful expressions against for instance Roma, will vary depending on the discursive context. When the expression is a direct part of a political discussion, the protection of freedom of expression will be higher. However, emphasis must also be placed on the aim, value, and accuracy of the statement, even in a political context. This will increase the possibilities to legally intervene against speech that may perhaps be triggered by an ongoing political debate, but is hateful and without value to that debate.  相似文献

In this paper, a study of the relationship between the impact performance of rotationally molded polyethylenes and their dynamic mechanical properties is carried out. A wide range of conventional linear low density polyethylene powders and met‐allocene catalyzed linear low density polyethylene powders were rotationally molded and tested. Instrumented falling weight impact tests were carried out over a temperature range of ?60°C to 20°C. Dynamic Mechanical Thermal Analysis (DMTA) tests were also carried out between ?100°C and 90°C, at a frequency of 100 Hz. Comparisons between the impact performance of metallocene catalyzed LLDPEs and Ziegler‐Natta LLDPEs are made. The transitions evident in the DMTA results are related to changes in impact performance with temperature. The beta transition is found to fall in the transition region between high impact performance at low temperatures and lower impact performance at ambient temperatures.  相似文献

A range-finding scanner can collect information about the shape of an (unknown) polygonal room in which it is placed. Suppose that a set of scanners returns not only a set of points, but also additional information, such as the normal to the plane when a scan beam detects a wall. We consider the problem of reconstructing the floor plan of a room from different types of scan data. In particular, we present algorithmic and hardness results for reconstructing two-dimensional polygons from point-wall pairs, point-normal pairs, and visibility polygons. The polygons may have restrictions on topology (e.g., to be simply connected) or geometry (e.g., to be orthogonal). We show that this reconstruction problem is NP-hard under most models, but that some restrictive assumptions do allow polynomial-time reconstruction algorithms.  相似文献

The present study investigated the effects of azuki bean (Vigna angularis) extract (VAE) on the progress of atopic dermatitis (AD)-like skin lesions in NC/Nga mice induced by 1-chloro-2,4-dinitrobenzene. The efficacy of VAE in NC/Nga mice was determined by measuring gross and histological skin lesions, serum IgE levels, eosinophil ratio in peripheral leucocytes, and mRNA expression levels of interleukin (IL)-4, tumour necrosis factor (TNF)-α and interferon (IFN)-γ in splenocytes. Continuous ingestion of VAE inhibited the development of the AD-like skin lesions in a dose-dependent manner. In the VAE-treated mice, the numbers of mast cells in the skin, eosinophil ratio in peripheral leucocytes, relative mRNA expression of inflammatory cytokines in the spleen, and serum IgE levels were significantly reduced. Results suggest that VAE can inhibit the development of AD-like skin lesions in NC/Nga mice by regulating immune mediators and cells, and may be an effective alternative therapy for AD.  相似文献
