A new numerical implementation is developed to calculate the diffraction efficiency of relief diffraction gratings. In the new formulation, vectors containing the expansion coefficients of electric and magnetic fields on boundaries of the grating layer are expressed by additional constants. An S-matrix algorithm has been systematically described in detail and adapted to a simple matrix form. This implementation is suitable for the study of optical characteristics of periodic structures by using modern object-oriented programming languages and different standard mathematical software. The modeling program has been developed on the basis of this numerical implementation and tested by comparison with other commercially available programs and experimental data. The castor bean plant (Ricinus communis) is used in large quantities for oil production and is also a common ornamental garden plant. However, the beans contain 1-3% of the highly toxic protein ricin, a type II ribosome-inactivating protein that is covered by the Chemical Weapons Convention, and there have been a number of reports concerning the use, or alleged use, of the toxin in terrorist and criminal activities. In the study reported here, we investigated the potential utility of organic solvent-assisted trypsin digestion of crude extracts containing the closely related toxins ricin or abrin to prepare samples for peptide analysis by liquid chromatography combined with electrospray ionization quadrupole time-of-flight tandem mass spectrometry. Diagnostic tryptic fragments of the toxins were detected and unambiguously identified by this procedure. The sample preparation protocol substantially reduces the sample preparation time, from overnight to an hour, and thus greatly reduces the total time required for analyses, to less than 2 h. Furthermore, the reported procedure leaves the disulfide bonds in the protein intact. This is highly relevant in the context of the Chemical Weapons Convention, since the disulfide bond connecting the two chains of ricin indicates the presence of an intact toxin and provides additional forensic evidence for the analytical results. 相似文献

Plant-based platforms have been successfully applied for the last two decades for the efficient production of pharmaceutical proteins. The number of commercialized products biomanufactured in plants is, however, rather discouraging. Cytokines are small glycosylated polypeptides used in the treatment of cancer, immune disorders and various other related diseases. Because the clinical use of cytokines is limited by high production costs they are good candidates for plant-made pharmaceuticals. Pressure effect on the rheological behaviour of powder-polymer compounds intended for powder injection moulding was determined. A single-piston capillary rheometer modified by addition of a second chamber with a restricting needle valve generating backpressure increasing the pressure in the melted material during the flow through the die was employed to determine pressure sensitivity coefficients. The results obtained for three compounds varying in the characteristics of powders used confirmed that compounds at the loading level close to the maximum packing are more sensitive to pressure than polymer binder. It is shown that pressure sensitivity coefficients of these materials are strongly dependent on powder characteristics — particle size and particle size distribution. The highest coefficient (32.9 GPa−1) was found for compound containing broad particle size distributed powder having a perceptible portion of small particles. 相似文献

Various pellet heating systems are marketed in Sweden, some of them in combination with a solar heating system. Several types of pellet heating units are available and can be used for a combined system. This article compares four typical combined solar and pellet heating systems. System 1 and 2 with a pellet stove, system 3 with a store integrated pellet burner and system 4 with a pellet boiler. The often lower efficiency of pellet heaters compared to oil or gas heaters increases the final energy demand. Consequently, heat losses of the various systems have been studied. The systems have been modeled in TRNSYS and simulated with parameters identified from measurements. For almost all systems the flue gas losses are the main heat losses except for system 3 where store heat losses prevail. Relevant are also the heat losses of the burner and the boiler to the ambient. Significant leakage losses are noticed for system 3 and 4. For buildings with an open internal design system 1 is the most efficient solution. Other buildings should preferably apply system 2 or 3. The right choice of the system depends also on whether the heater is placed inside or outside of the heated area. Unlike the expectations and results from other studies, the operation of the pellet heaters with modulating combustion power is not necessarily improving the performance. A large potential for system optimisation exists for all studied systems, which when applied could alter the relative merits of the different system types. 相似文献

In this study, we analyzed the high temperature tribological behavior of AlCrTiN coatings deposited on WC substrates by low cathodic arc technique. The coatings chemical composition, Al 31 at.%, Cr 16 at.%, Ti 7 at.% and N 46 at.%, and the bonding state were evaluated by X-ray photoelectron spectroscopy. The mechanical properties of the coatings were studied by scratch-test and nanohardness depth sensing indentation. The morphology of the coatings surface, ball scars, wear tracks and wear debris as well as the oxidized samples was examined by scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX). The structure was analyzed using X-ray diffraction (XRD). Wear testing was carried out using a high temperature tribometer (pin-on-disc) with alumina balls as counterparts. The evaluation of the friction coefficient with the number of cycles (sliding distance) was assessed at different temperatures and the wear rates of the coatings and balls were determined; the maximum testing temperature was 800 °C. The coating showed an excellent thermal stability and wear resistance. The friction reached a maximum at 500 °C and then decreased, whereas the wear rate was negligible up to 600 °C and increased significantly at higher temperatures. 相似文献

Baseline correction is an important pre-processing technique used to separate true spectroscopic signals from interference effects or remove background effects, stains or traces of compounds, e.g. in 2D gel electrophoresis. In some cases parts of the spectra or images need correction using rigid baselines (limited curvature) while other parts need more flexible baselines (more curvature). Often one has to make a compromise that is not optimal over the whole spectral range, or focus on one part and let the rest be treated sub-optimally. A customizing wrapper is proposed that rescales the spectrum abscissa and therefore makes the baseline correction algorithm behave right in all parts of the spectra. Improvements are demonstrated both visually and through regression using recently reported Raman spectra on melted fat from pork adipose tissue. 相似文献

In this study, the toxic effects of verapamil (VRP) were studied on juvenile rainbow trout, Oncorhynchus mykiss, by chronic semi-static bioassay. Fish were exposed to sublethal concentrations of VRP (0.5, 27 and 270 μg/L) for 0, 21 and 42 d. Multiple biomarkers were measured, including morphological indices, hematological parameters and antioxidant responses of different tissues (brain, gill, liver, muscle and intestine). Based on the results, there was no significant change in all parameters measured in fish exposed to VRP at environmental related concentration, but VRP-induced stress in fish exposed to higher concentrations reflected the significant changes of physiological and biochemical responses. Through principal component analysis and integrated biomarker response assessment, effects induced by VRP-stress in each test group were distinguished. Additionally, all parameters measured in this study displayed various dependent patterns to VRP concentrations and exposure time using two-way ANOVA statistic analysis. In short, the multiple responses in fish indicated that VRP induced physiological stress and could be used as potential biomarkers for monitoring residual VRP in aquatic environment; but molecular and genetic mechanisms of these physiological responses in fish are not clear and need to be further studied. 相似文献

We report on the development of a new environmental simulation chamber coupled with an in situ continuous wave cavity ring-down spectrometer operating in the near IR (~1.5 μm). The first application reported in this paper dealt with the chemical mechanism of UV photolysis of methyl nitrite (CH(3)ONO) in air. HONO has been detected for the first time and shown to be formed in the OH + CH(3)ONO reaction. A dense spectrum of cis-HONO absorption lines has been observed near 1.5 μm, in agreement with a previous study (Guilmot et al.). CH(2)O has been measured as primary product with good sensitivity and time resolution. In contrast to Zhao et al., we did not detect any NO(2) absorption features in this wavelength range. Calibration experiments provided very low NO(2) absorption cross sections in this region (~10(-25) cm(2)), leading to conclude that NO(2) cannot be observed in this wavelength range in the presence of equal amounts of CH(2)O. 相似文献
