Characterisation of the microbiota of Atlantic mackerel (Scomber scombrus)   总被引:1,自引:0,他引:1  
In this study the microbiota of Atlantic mackerel (Scomber scombrus) collected by a commercial purse seiner was examined. Fish were collected directly from the purse seine and from the Refrigerated Sea Water (RSW) transport tank after loading. The culturable microbiota and Specific Spoilage Bacteria (SSB) were quantified on Iron Agar Lyngby (IAL) and identified using commercially available Biochemical API® kits on pure cultured isolates. These kits showed to be sub-optimal in characterising the isolates, since only half of the strains were identified. The same isolates were also identified by a nucleic acid based PCR-DGGE approach, and only half of the sequences gave the same results as the API®. Characterisation by PCR-DGGE was also performed on bacterial DNA from IAL plates (bulk cell samples) and on samples where the bacterial DNA was extracted directly from fish material without any cultivation (direct DNA samples). The microbiota of Atlantic mackerel was dominated by members of the Gram-negative genera as Psychrobacter sp., Proteus sp., Photobacterium sp., Vibrio sp., Shewanella sp., Synechococcus sp., Oceanisphaerae sp., Bizonia sp., Pseudoalteromonas sp., and members of Flavobacteriaceae. Gram-positive bacteria in the genera Vagococcus sp., Bacillus sp., Mycobacterium sp., Staphylococcus sp., Mycoplasma sp. and Clostridia sp. were also found. Examination by PCR-DGGE and sequencing of the bulk cell pellet after cultivation on IAL, gave a higher number of taxa as compared to extraction and examination of bacterial DNA from fish materials without prior cultivation. This shows the benefit of combining both culture dependent and culture independent methods, when studying the microbiota of marine fish. Several Vibrio spp. were found only in gut samples collected from the purse seine, but in all samples including the skin and the gills collected from the RSW tank, indicating microbial contamination by faecal bacteria from the fish under these transport conditions.  相似文献

A quality index method (QIM) was developed for farmed Atlantic halibut and together with instrumental, chemical, sensory, and bacteriological analysis, quality changes of halibut stored on ice for 26 d was evaluated. Two groups of fish were fed diets that differed only in the source of lipid, where 1 diet contained only marine oil sources and the other a 50/50 mixture of marine and soybean oil. Fish were slaughtered after 1 y and then stored on ice for 26 d. The fish were sampled on day 1, day 2, and every 2nd day after that. Dietary lipid sources had no effect on freshness, (ATP) degradation (K‐value), texture, color, or liquid‐holding capacity. The QIM scores increased with storage time, in particular the appearance and eyes parameters. The QIM is a good freshness indicator for halibut. The K‐value was strongly correlated with storage time (r= 0.99), while total bacterial counts increased after 7 to 8 d of ice storage. The texture, liquid‐holding capacity, and color were significantly affected by storage time during the early period of storage, probably due to rigor stiffness and rigor resolution. The texture, liquid‐holding capacity, and color did not change significantly from approximately day 8 of storage until the end of the experiment at day 26.  相似文献

The oil and gas (O&G) industry on the Norwegian continental shelf (NCS) leads the world in terms of the number of subsea O&G installations. Ensuring the dependability of these assets is critical. Non-intrusive inspection, maintenance and repair (IMR) services are therefore needed to reduce risks. These services are planned and executed using a mono-hull offshore vessel complete with remotely operated vehicles (ROVs), a module handling system and an active heave compensated crane. Vessel time is shared between competing jobs, using a prioritized forward-looking schedule. Extension in planned job duration may have an impact on O&G production, service costs and health, safety, and environmental (HSE) risks. This paper maps factors influencing the job schedule efficiency. The influence factors are identified through reviews of literature as well as interviews with experts in one of the large IMR subsea service providers active on the Norwegian Continental Shelf. The findings show that the most obvious factors are weather disruption and water depth. Other factors include job complexity, job uncertainty, IMR equipment availability, as well as the mix of job complexity.  相似文献

The present article describes the use of broad-range molecular analyses to characterise the microbial population of farmed Atlantic cod (Gadus morhua) packaged for the retail market. Cod was filleted post rigor, packaged in air or in modified atmosphere (MA) (50% CO(2):50% N(2) or 50% CO(2):50% O(2)) and stored at 0 degrees C for 11 days. To determine the community profiles of the samples the variable V3-region of the bacterial 16S rRNA gene were amplified by PCR, before the PCR products were separated by denaturing gradient gel electrophoresis (DGGE). From sequence analyses Pseudomonas spp. were found to be the predominant bacteria in oxygen enriched atmospheres, whereas the spoilage bacteria Photobacterium sp., Shewanella putrefaciens and Pseudomonas spp. dominated in CO(2):N(2) and air packaged samples. Additional microbial analyses by cultivation methods observed highest bacterial numbers in air stored samples, and both MA mixtures gave growth inhibition when measuring aerobic plate count, psychrotrophic bacteria and H(2)S-producing bacteria. The results show that PCR-DGGE can be applied to examine bacterial diversity and population shifts among different MA-packaged products.  相似文献

Recent years have shown a tremendous increase in consumer demands for healthy, natural, high-quality convenience foods, especially within the fish and seafood sector. Traditional processing technologies such as drying or extensive heating can cause deterioration of nutrients and sensory quality uncompilable with these demands. This has led to development of many novel processing technologies, which include several mild technologies. The present review highlights the potential of mild thermal, and nonthermal physical, and chemical technologies, either used alone or in combination, to obtain safe seafood products with good shelf life and preference among consumers. Moreover, applications and limitations are discussed to provide a clear view of the potential for future development and applications. Some of the reviewed technologies, or combinations thereof, have shown great potential for non-seafood products, yet data are missing for fish and seafood in general. The present paper visualizes these knowledge gaps and the potential for new technology developments in the seafood sector. Among identified gaps, the combination of mild heating (e.g., sous vide or microwave) with more novel technologies such as pulsed electric field, pulsed light, soluble gas stabilization, cold plasma, or Ohmic heat must be highlighted. In New Orleans, LA prior to hurricane Katrina 20-30% of inner-city children had elevated blood Pb levels > or =10 microg/ dL and 10 census tracts had a median surface soil level of Pb >1000 mg/kg (2.5 times the U.S. standard). This project tests the feasibility of transporting and grading contaminated properties (n = 25) with 15 cm (6 in.) of clean Mississippi River alluvium from the Bonnet Carré Spillway (BCS) (median soil Pb content 4.7 mg/kg; range 1.7-22.8). The initial median surface soil Pb was 1051 mg/kg (maximum 19 627). After 680 metric tons (750 tons) of clean soil cover was emplaced on 6424 m2 (69 153 ft2), the median surface soil Pb decreased to 6 mg/kg (range 3-18). Interior entrance wipe samples were collected at 10 homes before and after soil treatment and showed a decreasing trend of Pb (p value = 0.048) from a median of 52 microg/ft2 to a median of 36 microg/ft2 (25th and 75th percentiles are 22 and 142 microg/ft2 and 12 and 61 microg/ft2, respectively). Average direct costs for properties with homes were $3,377 ($1.95 per square foot), with a range of $1,910-7,020, vs $2,622 ($0.61 per square foot), with a range of $2,400-3,040 for vacant lots. Approximately 40% (86,000) of properties in New Orleans are in areas of >400 mg Pb/kg soil and estimated direct costs for treatment are between $225.5 and $290.4 million. Annual costs of Pb poisoning in New Orleans are estimated at approximately $76 million in health, education, and societal harm. Urban accumulation of Pb is an international problem; for example, the new Government of Norway established a policy precedence for an isolated soil cleanup program at daycare centers, school playgrounds, and parks to protect children. New Orleans requires a community-wide soil cleanup program because of the extent and quantity of accumulated soil Pb. The post-Katrina benefits of reducing soil Pb are expected to outweigh the foreseeable costs of Pb poisoning to children returning to New Orleans.  相似文献

The stress–strain behaviour of the aluminium alloy 7075 in T651 temper is characterized by tension and compression tests. The material was delivered as rolled plates of thickness 20 mm. Quasi-static tension tests are carried out in three in-plane directions to characterize the plastic anisotropy of the material, while the quasi-static compression tests are done in the through-thickness direction. Dynamic tensile tests are performed in a split Hopkinson tension bar to evaluate the strain-rate sensitivity of the material. Notched tensile tests are conducted to study the influence of stress triaxiality on the ductility of the material. Based on the material tests, a thermoelastic–thermoviscoplastic constitutive model and a ductile fracture criterion are determined for AA7075-T651. Plate impact tests using 20 mm diameter, 197 g mass hardened steel projectiles with blunt and ogival nose shapes are carried out in a compressed gas-gun to reveal the alloy's resistance to ballistic impact, and both the ballistic limit velocities and the initial versus residual velocity curves are obtained. It is found that the alloy is rather brittle during impact, and severe fragmentation and delamination of the target in the impact zone are detected. All impact tests are analysed using the explicit solver of the non-linear finite element code LS-DYNA. Simulations are run with both axisymmetric and solid elements. The failure modes are seen to be reasonably well captured in the simulations, while some deviations occur between the numerical and experimental ballistic limit velocities. The latter is ascribed to the observed fragmentation and delamination of the target which are difficult to model accurately in the finite element simulations.  相似文献
