The mechanisms underlying the transport of leptin into the brain are still largely unclear. While the leptin receptor has been implicated in the transport process, recent evidence has suggested an additional role of LRP2 (megalin). To evaluate the function of LRP2 for leptin transport across the blood-brain barrier (BBB), we developed a novel leptin-luciferase fusion protein (pLG), which stimulated leptin signaling and was transported in an in vitro BBB model based on porcine endothelial cells. The LRP inhibitor RAP did not affect leptin transport, arguing against a role of LRP2. In line with this, the selective deletion of LRP2 in brain endothelial cells and epithelial cells of the choroid plexus did not influence bodyweight, body composition, food intake, or energy expenditure of mice. These findings suggest that LRP2 at the BBB is not involved in the transport of leptin into the brain, nor in the development of obesity as has previously been described.  相似文献

Different samples of xTeO2.(25-y)B2O3.zV2O5.yYb2O3 (or TBVY) new glass material were synthesized by the classical melt-quenching method. Structural, optical, physical, and thermal analyses of the synthesized glasses were performed in addition to Monte Carlo simulation to test radiation shielding properties. The results showed that increasing ratios of Yb2O3 (y = 0.0, 0.5, 1.0, and 1.5 mol%) produced monotonic density values of the synthesized glasses ranging from 4.70058 g cm?3 to 5.01038 g cm?3. XRD and FTIR analyses were used to confirm the glass structure of all samples. Optical transmittance and absorption parameters varied almost monotonically with increasing ratios of Yb2O3 indicating the ability to predict and control these properties using Yb2O3 additive. Furthermore, simulated radiation interaction parameters, such as attenuation coefficients and half-value layer, exhibited well-behaved dependence on the concentration ratio of the Yb2O3 additive. This approach to glass material synthesis demonstrate the useful synergetic effect of combining structural, optical, and radiation characteristics.  相似文献

Recruitment of European eels (Anguilla anguilla) has declined to the extent that they have been added to the IUCN Red List of Threatened Species. Therefore, it is critical to ensure that eels complete their outward river migration in order to contribute to the available spawning stock. We conducted a 4‐year (2007–2011) telemetry study to understand the migratory behaviour and potential impact of environmental factors on the eel during this critical life stage. Out of 399 female eels tagged with acoustic transmitters, only 28% demonstrated clear downstream migratory behaviour. Fifty‐five percent were detected exhibiting no downstream migration behaviour and 17% were not detected at any monitoring station. Movement patterns of downstream‐migrating (silver) eels were characterized by nocturnal activity and seasonal migration, with distinct peaks in autumn and spring. Migration was often discontinuous and exhibited phases of active locomotion and expanded stopovers. The most important determinants of movement activity were water temperature, cumulative precipitation and moonlight, although the significance varied by season and location in the river basin. Our results evidence a discontinuous, stepwise migration over an extended period. Furthermore, our findings indicate that migration success depends on holding duration prior to tagging and environmental predictors with varying importance depending on the season, as well as the locations of capture, tagging and release. ABSTRACT:  The changes in skin and fillet color of anesthetized and exhausted Atlantic salmon were determined immediately after killing, during rigor mortis, and after ice storage for 7 d. Skin color (CIE L *, a *, b *, and related values) was determined by a Minolta Chroma Meter. Roche Salmo Fan™ Lineal and Roche Color Card values were determined by a computer vision method and a sensory panel. Before color assessment, the stress levels of the 2 fish groups were characterized in terms of white muscle parameters (pH, rigor mortis, and core temperature). The results showed that perimortem handling stress initially significantly affected several color parameters of skin and fillets. Significant transient fillet color changes also occurred in the prerigor phase and during the development of rigor mortis. Our results suggested that fillet color was affected by postmortem glycolysis (pH drop, particularly in anesthetized fillets), then by onset and development of rigor mortis. The color change patterns during storage were different for the 2 groups of fish. The computer vision method was considered suitable for automated (online) quality control and grading of salmonid fillets according to color.  相似文献

ABSTRACT

Five‐level combinations of African breadfruit, corn and soybean, in the ratios 40:5:55; 55:5:40; 70:5:25; 85:5:10 and 100:0:0%, respectively, were hydrated to 15, 18, 21, 24 and 27% and extruded into snacks at screw speeds of 100, 120, 140, 160 and 180 rpm. Physical characteristics of snacks from blends containing 15 and 18% moisture ranged from thin‐smooth to thin‐fine‐smooth pellets. Those containing 21 to 27% moisture were either thin‐smooth or thick‐smooth, fine‐smooth or rough strands. Feed moisture and feed composition were the most significant process variables influencing physical and sensory characteristics. The optimum process variable combination that had maximum influence on physical and sensory characteristics of snacks was the 70:5:25 feed ratio with 21% moisture and extruded at 140 rpm. This resulted in an overall acceptability score of 8.20 on a 9‐point hedonic scale.

PRACTICAL APPLICATIONS

African breadfruit is widely grown in the high rainforest parts of Nigeria and other African countries, where the seeds are traditionally consumed as porridge meal when cooked with ingredients or as snacks when roasted. There is therefore a need to adopt more efficient extrusion technologies to rapidly and efficiently transform African breadfruit and its blends with corn and soybean into acceptable snacks. Optimizing process variable conditions using response surface analysis to evaluate product physical characteristics and acceptability was the thrust of this study. It is expected to give direction toward a more scientific approach to scaling up operations in African breadfruit seed processing and utilization.

The contents of the 2 antihypertensive peptides Val-Pro-Pro (VPP) and Ile-Pro-Pro (IPP) were determined in 101 samples from 10 different Swiss cheese varieties using HPLC with subsequent triple mass spectrometry. In the category of extra hard and hard cheeses, the Protected Denomination of Origin cheeses Berner Alpkäse and Berner Hobelkäse, L’Etivaz à rebibes, Le Gruyère, Sbrinz, Emmentaler (organic and conventional) and in the category of semihard cheeses, the varieties Tilsiter, Appenzeller fat and full fat, Tête de Moine, and Vacherin fribourgeois were screened in the study. The average concentration of the sum of VPP and IPP in the screened cheese varieties varied to a large extent, and substantial variations were obtained for individual samples within the cheese varieties. The lowest average concentration of the 2 tri-petides was found in L’Etivaz à rebibes (n = 3) at 19.1 mg/kg, whereas Appenzeller fat (n = 4) contained the greatest concentration at 182.2 mg/kg. In individual samples, the total concentration of VPP and IPP varied between 1.6 and 424.5 mg/kg. With the exception of a 10-yr-old cheese, VPP was always present at greater concentrations than IPP. Milk pretreatment, cultures, scalding conditions, and ripening time were identified as the key factors influencing the concentration of these 2 naturally occurring bioactive peptides in cheese. The results of the present study show that various traditional cheese varieties contain, on average, similar concentrations of the 2 antihypertensive peptides to the recently developed fermented milk products with blood pressure-lowering property. This may serve as a basis for the development of a functional cheese with blood pressure-lowering property.  相似文献

In the field of food safety, meta-analysis can be used to combine results of prevalence studies of pathogens at critical stages within the food processing chain so that policy makers can access reliable and concise information on the effectiveness of interventions for controlling and preventing foodborne illnesses in humans. The objective of this work was to demonstrate the applicability of a parametric approach of meta-analysis to the specific case of determining the overall effect of chilling on Salmonella prevalence on pig carcasses. A meta-analysis was performed on each of two parameters measuring effect size for binary outcomes (relative risk and risk difference). Both meta-analyses confirmed that the chilling operation has a significant beneficial effect (P < 0.001) on the reduction of Salmonella prevalence on pig carcasses. Because risk difference is a parameter sensitive to the differences across studies in carcass swab areas and Salmonella detection methods, its meta-analysis highly reflected this heterogeneity (P < 0.001). However, parameterization of relative risk, not being biased by the above sources of variability, did not give rise to heterogeneity among studies and produced a fixed-effects meta-analysis solution, which is deemed more suitable for compilations based on a small number of individual studies (n = 9). Because of the systematic approach of meta-analysis (i.e., individual studies are weighed according to precision) and its reliance for actual data, the output distribution of the relative risk effect size (approximately eN(-0.868,0.166)) merits consideration for inclusion in the chilling stage of quantitative risk assessments modeling the prevalence of this pathogen along the pork production chain.  相似文献

Dielectric heating of soil using radio waves (RW) can be applied to support various remediation techniques, namely biodegradation and soil vapor extraction, under in situ, on site or ex situ conditions. To improve the spatial resolution of energy dissipation, the design of rod electrodes was modified with an air gap around the electrode allowing thermal treatment focused to the desired soil volume. A combination of low- and high-frequency electrical energy was successfully applied to homogeneously heat the capillary fringe, the boundary region of saturated and unsaturated zones. The energetic efficiency of the method was evaluated showing that an efficient transformation of RW energy to heat in the target volume can be achieved. By comparing biodegradation and soil respiration under conventional and electric (low-frequency resistive and dielectric RW) heating, the compatibility of the electric heating methods with bioremediation processes could be proven. Therefore, RW-supported microbial degradation of pollutants is a real option for accelerated soil remediation.  相似文献
