Casein kinase I (CKI) is a family of serine/threonine protein kinases found in all eukaryotes examined to date. Here, the rat CKI isoforms alpha and alpha L were cloned and expressed in both eukaryotic and prokaryotic systems. Characterization of the genomic DNA flanking the exon unique to CKI alpha L demonstrated that CKI alpha and CKI alpha L arise by the alternative splicing of a common pre-mRNA molecule. To the best of our knowledge, the alpha L isoform is the only known active serine/threonine kinase to contain an insert within its catalytic domain. Tissue distribution of each splicing isoform was examined by RT-PCR, immunoprecipitation, and Western blotting. Both isoforms were expressed in all tissues tested but at different levels. Bacterially expressed CKI alpha isoforms were active and therefore biochemically characterized. CKI alpha and CKI alpha L proteins were demonstrated to have casein kinase I catalytic properties. More importantly, the recombinant isoform proteins exhibited differences in binding and activity toward common CKI substrates. These observations demonstrate that the alpha L insert within the kinase domain modulates substrate kinetics. These kinetic differences suggest that CKI alpha and CKI alpha L may perform different biological roles.  相似文献

The development of a solution method and computer program for the evaluation of multiarea reliability is reported. An interconnected power system is modelled as a capacitated network with probabilistic arc capacities. The proposed solution method consists of an analytic state space decomposition phase and a Monte-Carlo simulation phase. An optimization problem is solved to minimize the total computational time for the two phases. The solution of the optimal mix problem determines the termination of the decomposition phase and the size of sample for the Monte-Carlo phase. A new reliability index, the inadequate transfer capability, is introduced. This measure indicates the relative effectiveness of either increasing existing capacities or opening new interconnections between two areas. The proposed method has been incorporated into a computationally efficient production grade software package, called Remain (Reliability Evaluation of Multiarea Interconnections). The application of Remain to a seven-area example for planning-system enhancement is given. Computational-times data is also presented.  相似文献

Bacteremia in a community hospital: spectrum and mortality   总被引:1,自引:0,他引:1  
The problem of bacteremia was studied at our 500-bed community hospital during 1974-1975. All patients with positive blood cultures for clinically significant organisms were analyzed with respect to age, sex, host factors, type, dose, and time of institution of therapy with antibiotics, and mortality. Of 142 episodes of bacteremia, outcome according to underlying host factors showed that 12/15 died in the rapidly, fatal group, 12/24 died in the ultimately fatal group, and in the nonfatal group 25/103 died. Mortality for Gram-positive bacteremia was 37% (7/54) for Gram-negative bacteremia in the nonfatal group. In addition to underlying host factors, inappropriate antibiotic therapy also contributed to higher mortality in patients with gram-negative bacteremia.  相似文献

Arguably the most important decision to be made when developing an object recognition algorithm is selecting the scene measurements or features on which to base the algorithm. In appearance-based object recognition, the features are chosen to be the pixel intensity values in an image of the object. These pixel intensities correspond directly to the radiance of light emitted from the object along certain rays in space. The set of all such radiance values over all possible rays is known as the plenoptic function or light-field. In this paper, we develop a theory of appearance-based object recognition from light-fields. This theory leads directly to an algorithm for face recognition across pose that uses as many images of the face as are available, from one upwards. All of the pixels, whichever image they come from, are treated equally and used to estimate the (eigen) light-field of the object. The eigen light-field is then used as the set of features on which to base recognition, analogously to how the pixel intensities are used in appearance-based face and object recognition.  相似文献

Good performance of optical coatings depends on the appropriate combination of optical and mechanical properties. Therefore, successful applications require good understanding of the relationship between optical microstructural and mechanical characteristics and film stability. In addition, there is a lack of standard mechanical tests that allow one to compare film properties measured in different laboratories. We give an overview of the methodology of mechanical measurements suitable for optical coatings; this includes depth-sensing indentation, scratch resistance, friction, abrasion and wear testing, and stress and adhesion evaluation. We used the techniques mentioned above in the same laboratory to systematically compare the mechanical behavior of frequently used high- and low-index materials, namely, TiO2, Ta2O5, and SiO2, prepared by different complementary techniques. They include ion-beam-assisted deposition by electron-beam evaporation, magnetron sputtering, dual-ion-beam sputtering, plasma-enhanced chemical-vapor deposition, and filtered cathodic arc deposition. The mechanical properties are correlated with the film microstructure that is inherently related to energetic conditions during film growth.  相似文献
