Insulin-like growth factor I (IGF-I) and IGF-II, acting under the regulatory control of growth hormone, are the principal mediators of vertebrate growth. We have previously demonstrated that like humans, but unlike rodents, rainbow trout maintain high hepatic IGF-II messenger RNA levels into adulthood. Here we describe a rainbow trout IGF-II gene with a proximal promoter that contains two CCAAT/enhancer-binding protein (C/EBP) binding sites (TCBS1 and TCBS2). Nuclear proteins corresponding in size to rat C/EBPalpha and C/EBPbeta were detected on Western immunoblots of growth-hormone-treated and mock-treated trout liver extracts. Electrophoretic mobility shift assay of these nuclear extracts further suggests the presence of C/EBPs in trout liver and confirms the ability of TCBS1 to form a complex with trout liver nuclear proteins that is identical in mobility and specificity to that formed by a mammalian consensus CBS construct. In both Western blot and mobility assay results, the growth-hormone-treated trout livers appeared to have a greater accumulation of C/EBP, suggesting a molecular mechanism by which growth hormone can influence the level of serum IGF-II.  相似文献

In situ hybridization histochemistry with somatostatin sst1-sst5 receptor messenger RNA-selective oligoprobes and quantitative receptor autoradiographic binding studies using [125I]Tyr3-octreotide, [Leu2,D-Trp22,125I-Tyr25]somatostatin-28 and [125I]CGP 23996 ([125I]c[Asn-Lys-Asn-Phe-Phe-Trp-Lys-Thr-Tyr-Thr-Ser]) were performed to determine the level of expression of somatostatin receptor messenger RNA and receptor binding sites in the hippocampal formation, limbic system and cerebral cortex of adult rats electrically kindled in the dorsal hippocampus. In control rats (implanted with electrodes but not electrically stimulated), the somatostatin-1 receptor-selective [125I]Tyr3-octreotide and the non-subtype-selective [Leu3,D-Trp22,125I-Tyr25]somatostatin-28 preferentially labelled the strata oriens and radiatum of the CA1 subfield of the hippocampus, the molecular layer of the dentate gyrus, the subiculum and presubiculum of the hippocampal formation, the inner layer of the frontal cortex, and the lateral and basolateral nuclei of the amygdala. The non-subtype-selective radioligand [125I]CGP 23996 (in 5 mM Mg2+ buffer) preferentially labelled the strata oriens and radiatum of the CA1 subfield of the hippocampus, the subiculum and the basolateral nucleus of the amygdala. Under conditions where primarily somatostatin-2 receptors were labelled, [125I]CGP 23996 (in 120 mM Na+ buffer) showed strong binding in the strata oriens and radiatum of the CA1 subfield of the hippocampus and the frontal cortex, whereas the dentate gyrus, subiculum and amygdala showed only weak signals. During and after kindling, no significant differences were observed between the ipsi- and contralateral sides of the hippocampus. A significant decrease (about 40%) of somatostatin receptor binding sites was observed in the molecular layer of the dentate gyrus with all radioligands (except [125I]CGP 23996 in Na+ buffer, which did not label this area) at stage 2 (pre-convulsive stage) and one week, but not one month, after stage 5 (generalized motor seizures). In contrast to somatostatin receptor binding, no alterations of the messenger RNA levels for sst1-sst5 receptors were found either at stage 2 or at stage 5. Similarly, no changes in receptor binding or messenger RNA levels were observed in the brain of rats which experienced a single afterdischarge. The present study shows a significant and selective decrease of somatostatin-1 receptor binding sites in the dentate gyrus of kindled rats. This is part of the plastic changes induced by kindling and may contribute to the increased sensitivity for the induction of generalized seizures during kindling.  相似文献

The water absorption characteristics of pineapple leaf fiber (PALF)/glass fiber (GF) hybrid polyester(PER) composites, and chemically modified PALF/polyester composites were evaluated by immersion in distilled water at 28, 60, and 90°C. The diffusion properties of the intimately mixed (IM) and the layered hybrid composite GPG (Glass skin and PALF core) of different PALF/GF ratio at the three temperatures were compared in order to identify the environmental ageing mechanism at different temperatures. The effect of temperature on the kinetics and thermodynamics of diffusion were also examined. The water uptake of both IM and GPG hybrid composites was decreased with increase in glass fiber content; the lowest water uptake was observed for 0.46 Vf GF hybrid composite. Among the chemically modified composites, vinyl tri 2‐methoxy ethoxy silane treated composites showed the lowest water uptake. Finally, parameters like diffusion, sorption, and permeability coefficients were determined. It was observed that equilibrium water uptake is dependent on the nature of the composite and temperature. The rind of rambutan, which is normally discarded was found to contain extremely high antioxidant activity when assessed using several methods. Although having a yield of only 18%, the ethanolic rambutan rind extract had a total phenolic content of 762 ± 10 mg GAE/g extract, which is comparable to that of a commercial preparation of grape seed extract. Comparing the extract’s pro-oxidant capabilities with vitamin C, α-tocopherol, grape seed and green tea, the rind had the lowest pro-oxidant capacity. In addition, the extract at 100 μg/ml was seen to limit oxidant-induced cell death (DPPH at 50 μM) by apoptosis to an extent similar to that of grape seed. The extracts were not cytotoxic to normal mouse fibroblast cells or splenocytes while the powderised rind was seen to have heavy metals contents far below the permissible levels for nutraceuticals. Our study for the first time reveals the high phenolic content, low pro-oxidant capacity and strong antioxidant activity of the extract from rind of Nephelium lappaceum. This extract, either alone or in combination with other active principles, can be used in cosmetic, nutraceutical and pharmaceutical applications.  相似文献

BACKGROUND: β‐Glucan is a bioactive component of cereal grains that has many potential uses and health‐promoting benefits. Recent research has focused on improving the nutritional value of food by increasing human exposure to β‐glucan. This study looks at the development of a farm‐level baseline model (including scenario analysis) to evaluate the impact of pre‐ and postharvest stages (including genotypic factors, environmental conditions, agronomic factors and storage) on β‐glucan levels in barley. Monte Carlo simulation techniques were employed to model various stages in pre‐ and postharvest processes and to simulate the factors influencing the level of β‐glucan content in both hulled barley (HB) and hull‐less barley (HLB) genotypes. RESULTS: The baseline model found that the mean simulated level of β‐glucan was 40.99 and 56.77 g kg?1 for HB and HLB genotypes respectively. A sensitivity analysis highlighted that genotype was the most important parameter in determining the final β‐glucan content (correlation coefficients of 0.66 and 0.78 for HB and HLB respectively), more so than any of the agronomic factors analysed. The scenario analysis highlighted the importance of harvest date (scenario 2) and storage conditions (scenario 3), with a potential 32.6 and 32.7% decrease in β‐glucan (compared with the baseline model) if harvesting is carried out early during physiological maturity (i.e. at growth stage 92) and a potential 20.1 and 19.5% increase in β‐glucan for HB and HLB respectively if storage time is minimised. CONCLUSION: This study predicted the influence of genotypic, pre‐ and postharvest operations on β‐glucan content and thus allows strategies to be identified to influence β‐glucan content in barley products.
