We have shown that autoxidized polyphenolic nutraceuticals oxidize H2S to polysulfides and thiosulfate and this may convey their cytoprotective effects. Polyphenol reactivity is largely attributed to the B ring, which is usually a form of hydroxyquinone (HQ). Here, we examine the effects of HQs on sulfur metabolism using H2S- and polysulfide-specific fluorophores (AzMC and SSP4, respectively) and thiosulfate sensitive silver nanoparticles (AgNP). In buffer, 1,4-dihydroxybenzene (1,4-DB), 1,4-benzoquinone (1,4-BQ), pyrogallol (PG) and gallic acid (GA) oxidized H2S to polysulfides and thiosulfate, whereas 1,2-DB, 1,3-DB, 1,2-dihydroxy,3,4-benzoquinone and shikimic acid did not. In addition, 1,4-DB, 1,4-BQ, PG and GA also increased polysulfide production in HEK293 cells. In buffer, H2S oxidation by 1,4-DB was oxygen-dependent, partially inhibited by tempol and trolox, and absorbance spectra were consistent with redox cycling between HQ autoxidation and H2S-mediated reduction. Neither 1,2-DB, 1,3-DB, 1,4-DB nor 1,4-BQ reduced polysulfides to H2S in either 21% or 0% oxygen. Epinephrine and norepinephrine also oxidized H2S to polysulfides and thiosulfate; dopamine and tyrosine were ineffective. Polyphenones were also examined, but only 2,5-dihydroxy- and 2,3,4-trihydroxybenzophenones oxidized H2S. These results show that H2S is readily oxidized by specific hydroxyquinones and quinones, most likely through the formation of a semiquinone radical intermediate derived from either reaction of oxygen with the reduced quinones, or from direct reaction between H2S and quinones. We propose that polysulfide production by these reactions contributes to the health-promoting benefits of polyphenolic nutraceuticals. 相似文献

Alkyl lysophospholipids have been shown to be cytooxic to a number of neoplastic tissues. One, ET-18-OCH3, has been used to selectively purge leukemic cells from mixtures with normal marrow progenitor cells,in vitro andin vivo. We have measured the 50% inhibitory (IC50) effect of a series of ether, lipids (EL) on leukemic cells (HL60, K562, Daudi, KG-1, KG-1a) and normal marrow progenitor
cells. Cells were incubated with varying concentrations of EL for 4 hr and assayed for viability, [3H]thymidine incorporation and clonogenicity in semi-solid media. The effect on protein kinase C (PKC) activity was assayed
for each compound. Compounds tested included three glycerophosphocholine analogs-ET-18-OCH3, ET-16-NHCOCH3, and BM 41.440. In addition, a lipoidal amine, CP 46665, an ethyleneglycolphospholipid, AEPL, and four single chain alkylphosphocholine
analogs, HePC2, HePC3, HePC4 and HePC6 were also tested. During the period of incubation, the cells remained viable (>70%) as judged by trypan blue dye exclusion.
The glycerophosphocholines were the most active and showed the highest therapeutic index. The lipoidal amine was active, but
toxic to normal marrow progenitor cells. The ethyleneglycolphospholipid was active against HL60, but not against the other
cell lines. The single chain alkylphosphocholine analogs were less active. All of the compounds inhibited PKC activity; however,
the glycerophosphocholines were the most inhibitory.

A bacterium identified as Arthrobacter sp. was grown on dibenzothiophene (DBT) sulfone as a sole source of sulfur, producing sulfite and sulfate. Sulfur in DBT sulfone (1.0 mM) was nearly quantitatively converted to sulfate by the organism. The organism could also use DBT sulfone as a sole source of carbon and energy. There was evidence for transient accumulation of benzoic acid in the culture medium after growth of the cells slowed. The DBT sulfoxide analogue 9-fluorenone was converted by resting cells to a product identified as 1,10-dihydroxy-1,10-dihydrofluoren-9-one, suggesting that DBT sulfone may be metabolized via an angular hydroxylation resulting in carbon- sulfur bond cleavage. In this paper, we study the phase transition behavior emerging from the interactions among multiple agents in the presence of noise. We propose a simple discrete-time model in which a group of non-mobile agents form either a fixed connected graph or a random graph process, and each agent, taking bipolar value either +1 or -1, updates its value according to its previous value and the noisy measurements of the values of the agents connected to it. We present proofs for the occurrence of the following phase transition behavior: At a noise level higher than some threshold, the system generates symmetric behavior (vapor or melt of magnetization) or disagreement; whereas at a noise level lower than the threshold, the system exhibits spontaneous symmetry breaking (solid or magnetization) or consensus. The threshold is found analytically. The phase transition occurs for any dimension. Finally, we demonstrate the phase transition behavior and all analytic results using simulations. This result may be found useful in the study of the collective behavior of complex systems under communication constraints. 相似文献

Thermodynamic assessments were made to optimize thermodynamic models and parameter fits to selected experimental and first principles hypothetical predicted phase data within the Na–Al–Ti–H system. This enabled thermodynamic modeling of Ti solubility within the sodium alanates: NaAlH4 and Na3AlH6, and the relative stability of Ti-bearing phases. The modeling provides insights into the role of Ti originating from Ti-based activating agents commonly referred to as ‘catalysts’ in promoting reversibility of the Na–Al–H dehydrogenation and rehydrogenation reactions under moderate temperature and pressure conditions relevant to H storage applications. Preliminary assessments were made to evaluate H solubility in bcc-Ti and hcp-Ti, and stability of the hydride δ-TiH2. To model possible Ti dissolution in NaAlH4 and α-Na3AlH6, sub-lattice models were applied. A repulsive interaction is predicted by first principles calculations when Ti is dissolved in NaAlH4 or α-Na3AlH6, which becomes stronger with increasing temperature. Although Ti is virtually insoluble in NaAlH4 or α-Na3AlH6, a small addition of TiCl3 will induce a thermodynamic driving force for formation of TiH2 and/or TiAl3. The addition of pure Ti shows a weaker effect than TiCl3 and leads to formation of TiH2 only. Based on a combined interpretation of present thermodynamic modeling and prior experimental observations, the TiAl3 and TiH2 phases are ascribed to have a catalytic effect, not a thermodynamic destabilization effect, on the reversibility of the dehydrogenation/rehydrogenation reactions in the Na–Al–H system. 相似文献

The d.c. conductivity, , and low-frequency relative dielectric constant, k, of Portland cement paste were monitored, using impedance spectroscopy, during cooling from room temperature down to -50 °C. Dramatic decreases in the values of and k, as great as two orders of magnitude, occurred at the initial freezing point of the aqueous phase in the macropores and larger capillary pores. This result provides strong experimental support for the dielectric amplification mechanism, proposed in Part II of this series, to explain the high measured low-frequency relative dielectric constant of hydrating Portland cement paste. Only gradual changes in the electrical properties were observed below this sudden drop, as the temperature continued to decrease. The values of and k of frozen cement paste, at a constant temperature of -40 °C, were dominated by properties of calcium-silicate-hydrate (C-S-H) and so increased with the degree of hydration of the paste, indicating a C-S-H gel percolation threshold at a volume fraction of approximately 15%–20%, in good agreement with previous predictions. Good agreement was found between experimental results and digital-image-based model computations of at -40 °C. Freeze-thaw cycling caused a drop in the dielectric constant of paste in the unfrozen state, indicating that measurements of k could be useful for monitoring microstructural changes during freeze-thaw cycling and other processes that gradually damage parts of the cement paste microstructure. 相似文献

Transposons of gypsy group are assigned to LTR-containing retrotransposons present in the genomes of invertebrates, fungi, and plants. In this work, a theoretical analysis of the potential products of ORFs of these retrotranposons was conducted. Alignments were obtained and trees of similarity were constructed for domains of the POL region. On the basis of the obtained data, two hypothetically monophyletic subgroups of transposons were distinguished within the framework of the gypsy group, settling the genomes of taxonomically related organisms (the subgroup of "true" gypsy of insects and the subgroup of gypsy-like transposons of plants and fungi). A number of peculiarities of the topology of these trees hypothetically indicate cases of genetic conversion and recombination of domains accompanying the evolution of this group. The amino acid substitution fixation rate was evaluated on the basis of comparison of sequences of the protein products of ORFs. Estimates of the time of divergence of subgroups of gypsy-group transposons are significantly less than estimates of the times of divergence of their host species. One explanation for this discrepancy might be the hypothesis of settlement by transposons of the genomes of isolated host species. 相似文献

PURPOSE: The goal of this research was to discover new biological indicators in urine which could be used for short-term prognosis of local Bacillus Calmette-Guerin (BCG) therapy outcome in patients with superficial bladder cancer. PATIENTS AND METHODS: We measured and statistically evaluated soluble immunological molecules in urine from bladder cancer patients (n = 34) receiving BCG intravesically. Urine was collected following each of 6 weekly treatments, processed and assayed. The data base included measurements of interleukin-1 (IL-2, IL-4, IL-6, IL-10, IL-12, soluble intercellular adhesion molecule-1 (sICAM-1), tumour necrosis factor-alpha (TNF alpha), soluble CD14 (sCD14), interferon-gamma (IFN gamma), GM-CSF, volume of urine and its pH. The clinical response was evaluated by urine histology and random quadrant biopsy 3 months after the start of therapy. Patients were divided into 2 groups, with good and poor therapeutic effect. The initial complete response rate was 62% (21/34). The data base was analyzed using traditional multivariate statistical methods and a pattern recognition method which deals with combinatorial-statistical analysis (statistically weighted syndromes (SWS) method) of the gradated features. The SWS method is capable of identifying robust patterns in small "fuzzy" sets with high dimensional objects and some missing values. RESULTS: Only one parameter gave significant differences at p < 0.05, GM-CSF at instillation 6. Repeated measurement analysis of variance, backward stepwise multiple logistic regression and linear discriminant analysis failed to show any significance. However, significant differences in the structure of correlation between features in the groups with and without therapeutic effect were observed and four highly informative variables (the masses of sICAM-1, TNF alpha, sCD14 and pH) relating to 5th-6th installations were selected by SWS. These features provided accurate individual prediction of therapeutic outcome for all our patients. Cross-validation analysis and computer simulation showed the statistically significant stability of the prediction. CONCLUSION: We have selected a set of urinary variables that could be considered as a perspective combination of indicators (syndromes) of outcome of pre-operation BCG therapy of patients with superficial bladder cancer. A larger patient database will provide testing and evaluation of the biological and clinical significance of selected features. The computational syndrome-disease approach should be applicable for the solution of decision-making problems for management of cancer. 相似文献
