Ligands with affinity to specific sequences of DNA base pairs. X. Synthesis and binding of netropsin analogs, containing a chelating copper ion peptide, with DNA

An analogue of netropsin has been synthesized consisting of two N-propylpyrrolcarboxamide units linked covalently to a copper-chelating tripeptide Gly-Gly-L-His by means of two and three glycine residues. Binding to DNA and synthetic polynucleotides of netropsin analogue containing three glycine residues between Gly-Gly-L-His tripeptide and the N-end of netropsin analogue (His-Nt) has been studied. It is shown that this netropsin analogue chelates a copper ion with 1:1 stoichiometry, similar to a free Gly-Gly-L-His peptide. It is found that this netropsin analogue occupies 3 to 4 base pairs upon binding to poly(dA).poly(dT) and poly[d(AT)].poly[d(AT)] polymers, irrespective of whether it binds in Cu(2+)-ligated or unligated forms. Binding constants and binding site sizes have been calculated for netropsin analogue complexes with DNA, poly(dA).poly(dT) and poly[d(AT)].poly[d(AT)] polymers at the [Cu2+]/[His-Nt] ratio equal to 0 and 1.0. In the three-component system including His-Nt and Cu(2+)-His-Nt, cooperative effects are recognized which can be explained by heterodimer generation on interaction of His-Nt and Cu(2+)-His-Nt at adjacent binding sites.     

The Bi2O3-SrO phase diagram

The phase relations in the Bi₂O₃-SrO system have been examined in air at temperatures above 740 °C. Intermediate phases were found to agree with previous work, but the homogeneity ranges and high-temperature phase relationships were in some cases at variance. A modified phase diagram is proposed.

68 3-yr-olds received a standard appearance–reality task along with either a trick task, in which the appearance question was placed in the context of a deceptive game, or a reduced information processing task, in which a dual object (e.g., a sponge-rock) was presented along with an object that matched the dual object's identity (a sponge) and one that matched the dual object's appearance (a rock). Children were more likely to pass either the trick or reduced information processing task and fail the standard than the reverse. Thus, 3-yr-olds can grasp the distinction between appearance and reality (a) when their goal is to trick someone, which may prime them to think about the other's mental state, and (b) when they do not need to hold conflicting object identities in mind at the same time. The early diagnosis of the severity of blast trauma to the brain and auditory system]

This paper presents the information on the early period of an explosive head trauma and trauma to the auditory system. Different types of hearing impairment, the staging of the pathological process were determined along with various immunological and biochemical changes occurring in this group of patients. The results of the study call for necessity of the early ENT observation of all patients in whom an explosive trauma is suspected.     

An analysis of the coupling of an injection laser diode to a planar LiNbO3waveguide

Calculated butt coupling efficiency between a GaAlAs double heterostructure laser and a single-mode LiNbO3Ti-diffused optical waveguide is presented in this paper as a function of the transverse displacement, the angular alignment, the longitudinal separation between the laser and the waveguide end surfaces, the radiation field of the laser diode and its dependence on the laser diode structure, and the mode profile of the waveguide and its dependence on experimental diffusion parameters. The maximum efficiency is less than 40 percent because of the mismatch between the waveguide mode and the laser radiation, caused primarily by the experimental limitations in their structures. The effect of the reflection of the waveguide end surface on the laser oscillation has also been estimated.     

Evaluation of humoral immune responses in cattle grazing endophyte-infected or endophyte-free fescue

Anecdotal reports suggest cattle with fescue toxicosis may not respond to vaccination and thus, experience increased incidence of Bovine Respiratory Disease Complex (BRDC) when shipped to feedlots. Fescue toxicosis causes hypoprolactemia in cattle. Hypoprolactemia decreases humoral immune responses in mice. Therefore, a study was conducted to compare the magnitude of primary and secondary humoral immune responses against specific antigens in cattle grazing endophyte-infected or endophyte-free fescue. Angus steers were blocked by weight and allocated into four groups. Two groups grazed endophyte-infected (EI) fescue and the other two groups grazed endophyte-free (EF) fescue. All steers were injected IM on d 0 and 21 with lysozyme without adjuvant and concanavalin. A (Con A) with sheep red blood cells (SRBC) in incomplete adjuvant of Freund. Steers were bled on days 0, 21 and 35 post-vaccination. Average daily gains (ADG), alkaline phosphatase (ALP) activity, cholesterol concentrations, rectal temperatures, and serum prolactin concentrations were measured to confirm fescue toxicosis in steers grazing EI fescue. Antibodies to Con A and SRBC were determined by ELISA and hemagglutination assay, respectively. The ADG were decreased for the EI group during the first month. Rectal temperature were elevated and serum prolactin concentrations were decreased in the EI group. Cholesterol and ALP concentrations also were decreased in the EI group. Primary and secondary immune responses against Con A tended to be increased and were increased against SRBC in the EI group. Antibodies against lysozyme were not induced in either group. In conclusion, cattle grazing EI fescue mounted similar humoral immune responses to vaccination, despite hypoprolactemia, as cattle grazing EF fescue. Increases in bovine respiratory disease in cattle maintained on EI fescue probably is not associated with lack of humoral immune response to vaccination protocols as a result of fescue toxicosis.     

Cuprates intermediate between one and two dimensions

The key elements in all known cuprate superconductors are lightly doped CuO_i-planes. Recently a new homologous series of compounds Sr_n–1Cu_n+1O_2n have been reported in which the planes contain a parallel array of line defects which form a trellis lattice with ladder-segments of the square lattice weakly coupled through triangular line defects. The width of the ladder segments is determined by the parameter n and varies from single chains to arbitrarily wide ladders. The magnetic properties of undoped compounds will be dominated by the properties of the ladders. Heisenberg s = 1/2 ladders can have a spin liquid groundstate with a spin gap if the number of rungs is odd so that a short range RVB groundstate is predicted for such trellis lattices. Using a t-J model to describe the doped material leads to the prediction of a d-wave RVB superconducting groundstate with a large spin gap.     

An experimental study of the possibilities of using pentoxifylline for the prevention of cancer at different sites

Three models of cancerogenesis were used to test the anti-cancerogenic effects of pentoxiphylline. In female rats, breast adenocarcinoma was induced by intramammary injections of N-methyl-N-nitrosourea (MNU) or colonic and rectal adenocarcinomas by intrarectal instillations of MNU. In female mice, squamous cell carcinomas of the cervix uteri and vagina were induced by intravaginal applications of 7,12-dimethyl-benz(a) anthracene (DMBA). Pentoxifylline was given with drinking water at a concentration of 500 mg/l long at the stage of carcinogenesis promotion/progression. Pentoxifylline exerted a strong inhibitory effect on the development of mammary tumors and a moderate inhibitory effect on the development of colonic and rectal tumors induced by MNU in rats. However, the drug did not affect the development of cervical and vaginal tumors caused by DMBA in mice.     

Perfection of methodical approaches to designing vaccines against tick-borne encephalitis

Nine female patients with a urethral diverticulum were admitted to our center over a six period: The clinical symptomatology was not specific. The diagnosis has been performed by voiding and retrograde cysto-uretrogram. The surgical treatment consisted in the excision of the diverticula, with the patient in the supine position. No perioperative morbidity was observed.
