Optimal conditions for in vitro cultivation of mouse embryos were sought. The embryos at 8--9.5 days of development were explanted into the culture in rotating tubes. A number of nutritional media were studied: the human umbilical blood serum; the rat blood serum, as well as its mixture (1 : 1) with Eagle's medium. It was demonstrated that the optimal medium for cultivation of 8-day-old embryos was the mixture of the rat blood serum with Eagle's medium, and for 8.5--9.5-day-old embryos--the rat blood serum, as well. Comparing in vitro and in vivo embryonic development, it was evident that in the cultivated embryos the parameters characterizing their growth decreased significantly. Possible causes of this phenomenon are discussed.  相似文献

Behavioral thresholds were measured in four cats by training them to respond behaviorally to acoustic auditory stimuli using food as a reward in an operant reinforcement paradigm. Following training, the subjects were implanted unilaterally with either a scaled-UCSF electrode containing four contacts or an electrode array containing eight intracochlear contacts and one extracochlear contact under temporalis muscle. Behavioral thresholds for electrical stimuli were obtained as a function of sinusoidal frequency, duration, and electrode configuration. Threshold functions for both electrode types and all animals had minima between 48 and 125 Hz and, in general, were relatively flat below this minima; functions increased at 3-6 dB/octave from 96 Hz to 1 kHz. Threshold varied predictably as a function of electrode configuration, with thresholds decreasing as much as 20 dB as electrode spacing was widened from a radial bipolar (200-microns separation) to a monopolar configuration (apical stimulating and temporalis return). With long-duration stimuli, increasing the electrode separation systematically increased the slope of the threshold-versus-frequency contours in all animals. Irrespective of electrode type or configuration, charge/phase thresholds for single-cycle sinusoids were relatively flat for stimulus periods up to 1-5 ms, approximating a constant charge/phase determination of threshold. At phase durations greater than 5 ms, charge thresholds increased at a rate slightly above 6 dB/octave (constant peak current), which was suggestive of neural accommodation. Thresholds for the cat share many features with those reported for implanted humans and monkeys.  相似文献

The importance of ProB28 and LysB29 on the self-associationof insulin was established by systematically truncating theC terminus of the B chain. The relationship between structureand association was further explored by making numerous aminoacid replacements at B28 and B29 Association was studied bycircular dichroism, size-exclusion chromatography and ultracentrifugation.Our results show that the location of a prolyl residue at B28is critical for high-affinity self-association. Removal of ProB28in a series of C-terminal truncated insulins, or amino acidreplacement of Pro28, greatly reduced association. The largestdisruption to association was achieved by replacing LysB29 withPro and varying the amino acid at B28 Several of the analogswere predominantly monomers in solutions up to 3 mg/ml. Theseamino acid substitutions decreased association by primarilydisrupting the formation of dimers. Such amino acid substitutionsalso substantially reduced the Zn-induced insulin hexamer formation.The formation of monomeric insulins through amino acid replacementswas accompanied by conformational changes that may be the causefor decreased association. It is demonstrated that self-associationof insulin can be drastically altered by substitution of oneor two key amino acids.  相似文献

Clinico-statistical evaluation of reparative process in 332 gastric ulcer and 294 duodenal ulcer patients revealed factors of objective value to its course. In order of significance the factors rank as follows: area of ulcer, localization, combination with certain mucosal erosions, total fasting acidity, free fasting hydrochloric acid, basal gastric secretion, cardial function, duodenogastric reflux, sex, age, number of cigarettes smoked daily. When studying treatment effects on regeneration in ulcer patients in is necessary to take into consideration basal clinico-endoscopic parameters.  相似文献

Our objective in this study was a comparative evaluation of efficiency of modern drug alternatives to treat peptic ulcer, based on new algorithms of diagnosis and treatment. The results obtained suggest to us that in a major proportion of cases uncomplicated duodenal ulcers might be efficiently managed in an outpatient-clinic setting even with a two-to-four week monotherapy. The best results of treatment are recordable in those groups of patients having received inhibitors of proton pump.  相似文献

OBJECTIVE: To report a case of respiratory failure, possibly due to anaphylaxis or asthma exacerbation, following the administration of metoclopramide. CASE SUMMARY: A 32-year-old white woman with a history of severe asthma and short-bowel syndrome was admitted for Hickman catheter line sepsis. Two doses of oral metoclopramide 10 mg in solution were administered for nausea and vomiting. Transient dyspnea followed the first dose of metoclopramide, but respiratory failure requiring intubation followed the second dose. DISCUSSION: Respiratory failure has been reported with metoclopramide-induced movement disorders. Three other cases of respiratory failure from anaphylaxis or asthma exacerbation following metoclopramide administration have been reported. Respiratory failure in our patient may be due to anaphylaxis or bronchoconstriction from metoclopramide-induced cholinergic activity of the vagus nerve, possibly through inhibition of acetylcholinesterase. CONCLUSIONS: The use of metoclopramide in patients with pulmonary dysfunction may warrant caution.  相似文献

To confirm the existence and characterize the pathologic features of humoral (antibody-mediated) lung rejection, we prospectively studied 55 lung transplant recipients (24 male [44%] and 31 female [56%], age range 14 to 69 years [mean 45]). The time between transplantation and biopsy ranged from 2 to 1546 days (mean 274). We performed direct immunofluorescence with C3, immunoglobulin M, and immunoglobulin G antibodies on frozen sections of 106 transbronchial biopsies and one wedge biopsy and compared the results with 13 explanted lungs, one donor lung, and two controls. The histologic diagnoses of these 107 biopsies included acute cellular rejection (62, 58% [minimal 23, mild 33, moderate 5, and severe 1]), chronic rejection (eight, 7%), chronic vascular rejection (two, 2%), acute vasculitis (five, 5%), cytomegalovirus pneumonitis (two, 2%), acute pneumonia (two, 2%), acute organizing pneumonia (two, 2%), diffuse alveolar damage (one, 1%), no evidence of rejection or infection (30, 28%), lipoid pneumonia (one, 1%), and inadequate for histologic diagnosis (one, 1%). Eighty-nine of 106 (84%) transbronchial biopsies, the wedge biopsy, and control lungs were satisfactory for direct immunofluorescence, because each contained alveolate lung parenchyma and arterioles or venules. There was no demonstrable immunofluorescence in the wall of the blood vessels or in the lung parenchyma in any case. We conclude that (1) transbronchial biopsies and wedge biopsies provide adequate material to evaluate humoral rejection, and (2) in spite of the large population studied, the satisfactory material obtained, and the wide range of histologic diagnoses, we could not demonstrate the occurrence of humoral rejection in the lung.  相似文献
