Productive infection with HIV-1, the virus responsible for AIDS, requires the involvement of host cell factors for completion of the replicative cycle, but the identification of these factors and elucidation of their specific functions has been difficult. A human cDNA, TRBP, was recently cloned and characterized as a positive regulator of gene expression that binds to the TAR region of the HIV-1 genome. Here we demonstrate that this factor is encoded by a gene, TARBP2, that maps to human chromosome 12 and mouse chromosome 15, and we also identify and map one human pseudogene (TARBP2P) and two mouse TRBP-related sequences (Tarbp2-rs1, Tarbp2-rs2). The map location of the expressed gene identifies it as a candidate for the previously identified factor encoded on human chromosome 12 that has been shown to be important for expression of HIV-1 genes. Western blotting indicates that despite high sequence conservation in human and mouse, the TARBP2 protein differs in apparent size in primate and rodent cells. 相似文献

Tamoxifen has been used as an adjuvant therapy for breast cancer for nearly two decades. The benefits of adjuvant tamoxifen therapy in prolonging disease-free and overall survival have been shown in randomized clinical trials. Despite this, some developing evidence suggests that tamoxifen causes a 2- to 3-fold increase in endometrial cancer. This paper reviews the reports of endometrial carcinoma in tamoxifen-treated patients. Two hundred fifty cases of endometrial carcinoma are reported, but only one case is identified in a premenopausal woman. When documented, 77% (n=127) of the cases are good-grade (grade 1 or 2) and 80% (n=125) are stage-I disease. Since the distribution of good grade (79%) and stage I (74%) from the Surveillance, Epidemiology and End Results (SEER) data are comparable, concerns about more aggressive or late-stage disease appear to be unwarranted. The modest increase in the incidence of early-stage, good-grade endometrial carcinoma described during tamoxifen therapy suggests that it would be unreasonable to institute an aggressive detection strategy of endometrial biopsies. This approach would only lead to further detection bias and would not be cost-effective. Physicians should ensure that patients do not have pre-existing endometrial cancer prior to adjuvant tamoxifen therapy for breast cancer and, furthermore, they should educate patients about signs and symptoms of early endometrial carcinoma and when reported these should be followed up with a gynecologic examination. 相似文献

Evaluating the efficacy of neuroprotective drugs in rat models of focal cerebral ischemia has involved histological and behavioral batteries to examine treatment outcome. However, the behavioral tests used to date provide little insight into the nature of the neurological impairments. To provide an analysis of a possible "neglect" syndrome after occlusion of the middle cerebral artery, M. I. Posner's (1980) visual attentional paradigm was adapted for use in the rat. A paw-reaching task and a test of somatosensory "neglect" also were used to assess forelimb sensorimotor function. The lesion group displayed unilateral deficits; however, there was no evidence of attentional dysfunction. Results are consistent with the conclusion that the behavioral deficits identified arise from a somatosensory deficit rather than hemineglect due to dysfunctional spatial attention. 相似文献

Residual orientation in injection‐moulded plaques of atactic‐polystyrene I: The effect of processing conditions 下载免费PDF全文
Injection moulded polymer articles often have residual macromolecular or crystalline orientation which can have a significant impact on the optical and mechanical properties of the moulded article. Small angle neutron scattering (SANS) was used to measure the molecular shape and orientation of deuterated blends of injection moulded polystyrene. For ~1‐mm‐thick mouldings of uniform rectangular cross‐section, the eccentricity in the SANS pattern gave a direct measure of the residual molecular orientation over the length scale ~100–1,500 Å. The residual orientation was found to vary significantly with injection moulding conditions with comparative residual orientation decreasing with decreasing mould fill‐time, and increasing with mould thickness and moulding temperatures. The orientation was found to be a minimum in the centre of the mould and highest near the surface and the average orientation at a particular position in the mould was found to be strongly correlated with the volume of material deposited as a solid skin layer during injection moulding. We found previously that restriction of tyrosine (Tyr) and phenylalanine (Phe) inhibited growth and metastasis of B16BL6 murine melanoma and arrested these cells in the G0-G1 phase of the cell cycle. Here, we report that deprivation of these two amino acids in vitro induces apoptosis in B16BL6 and in human A375 melanoma cells but not in nontransformed, neonatal murine epidermal cells or human infant foreskin fibroblasts. Four days after deprivation of Tyr and Phe in vitro, 37% of B16BL6 and 51% of A375 melanoma cells were undergoing apoptosis. Apoptosis was not associated with elevation in intracellular calcium or alteration in p53 or c-myc protein expression. Expression and Tyr phosphorylation of focal adhesion kinase (FAK) were inhibited in both melanoma cell lines by deprivation of Tyr and Phe but not by deprivation of glutamine or serum. Tyr phosphorylation of FAK in Tyr- and Phe-deprived melanoma cells was enhanced within 30 min of refeeding with complete DMEM. FAK protein expression recovered within 60 min, and cell viability recovered within 24 h. Genistein, a tyrosine kinase inhibitor that specifically inhibits Tyr phosphorylation of FAK, did not induce apoptosis in A375 melanoma cells at a concentration of 50 microM. Genistein prevented the recovery of cell viability upon refeeding with Tyr and Phe to previously deprived A375 melanoma cells. These data collectively indicate that apoptosis induced by Tyr and Phe deprivation is FAK-dependent. 相似文献

Fatigue performance has been studied in a cast Al-7Si-Mg alloy with regard to various casting defects, particularly surface or subsurface defects. Fatigue tests were carried out under four-point bending to investigate the stress-life relationship and fatigue fracture characteristics in order to understand crack initiation and growth behaviour in conjunction with the examination of surface roughness and porosity. Surface hollows were found to control crack initiation of as-cast specimens. The fracture surfaces of polished specimens revealed that surface or subsurface shrinkage pores replaced the hollows to act as crack initiators when the rough surface was removed. The importance of oxide films in crack initiation was also demonstrated. The effects of all these casting defects on fatigue life are discussed. 相似文献

The goals of energy independence and sustainability have motivated many countries to consider biomass-based energy sources. The United States has substantial and increasing forest resources that could be used to produce both electricity and liquid fuel. However, these forest resources are highly heterogeneous in terms of the wood’s properties, the logging cost, the spatial distribution, and the value to other industries. These factors make predicting costs and selecting plant locations particularly challenging. When dealing with forest biomass, feedstock cost and location have frequently been highly simplified in previous studies. This paper presents a methodology for combining highly resolved forest inventory and price data with records of competing industries to develop detailed maps of feedstock availability. The feedstock sourcing strategy of the proposed bioenergy plants is modeled by a cost-minimizing linear program, as is the feedstock selection of the competing mills. Critical age, weight and body composition have been suggested as necessary correlates of sexual maturity. A genome scan to identify quantitative trait loci (QTL) for age and body weight at first egg (AFE and WFE) was conducted on 912 birds from an F(2) broiler-layer cross using 106 microsatellite markers. Without a covariate, QTL for body WFE were detected on chromosomes 2, 4, 8, 27 and Z and a single QTL for AFE was detected on chromosome 2. With AFE as a covariate, additional QTL for body WFE were found on chromosomes 1 and 13, with abdominal fat pad as covariate a QTL for body WFE was found on chromosome 1. With body WFE as covariate, additional QTL for AFE were found on chromosomes 1, 3, 4, 13 and 27. The QTL generally acted additively and there was no evidence for epistasis. Consistent with the original line differences, broiler alleles had positive effects on body WFE and negative effects on AFE, whereas the phenotypic correlation between the two traits was positive. The mapped QTL for body WFE cumulatively accounted for almost half the body weight difference between the chicken lines at puberty. Overlapping QTL for body WFE and body weight to 9 weeks of age indicate that most QTL affecting growth rate also affect body WFE. The co-localisation of QTL for body weight, growth and sexual maturity suggests that body weight and growth rate are closely related to the attainment of sexual maturity and that the genetic determination of growth rate has correlated effects on puberty. 相似文献

Simple protocols to convert molecular mechanics (MMX/PCMODEL), semiempirical PM3, and HF ab initio energies to accurate heats of formation for hydrocarbons with benzene rings are described. The data set consists of every hydrocarbon benzene derivative with an experimentally determined ΔHfo (g), and the ΔHfo (g)'s cover a range of -140 to +410 kJ/mol. The molecular structures are comprised of numerous structural types. Hierarchical sets of molecular structure parameters are defined to describe these molecules. The independent variables include atom types (level 1), group and ring terms (level 2), nonbonded atom interactions (level 3), and the calculated MMX, PM3 or ab initio HF energies, which contribute a final level 4 parameter for rectification of the ΔHfo (g) data. The additivity, level 1-3 parameters give an excellent correlation of the experimental ΔHfo (g)'s, average error = 3.4 kJ, and maximum error = 12.1 kJ. However, the correlations are further enhanced by addition of any level 4 parameter, with maximum improvement coming at the 6-31G*//STO-3G HF level of calculation. 相似文献
