Neurosensory cutaneous discomfort in response to topical products is common, yet the relationship between symptoms such as stinging and visible irritation is currently unclear. The presence of a polymorphism at position ‐308 on the TNF‐α gene has been associated with skin irritation, i.e., erythema, dryness. Individuals with a G to A transition (AA/GA genotypes) have a lower threshold to experimentally induced irritation than those with the wild type (G allele, GG genotype). We investigated the effect of this polymorphism on neurosensory irritation (NSI). DNA genotyping was used to determine the allele type amongst a population of health care workers. The neurosensory response to lactic acid and water on the nasolabial folds and hands was assessed using a quantitative lactic acid sting test. Both genotypes had a more intense response to lactic acid compared with water on the face. The AA/GA genotypes had directionally higher scores from lactic acid (P = 0.1) and significantly higher stinging intensities from water (P = 0.001) on the face. For the hands, stinging intensities were higher for lactic acid and water amongst the AA/GA genotypes (P = 0.03 and 0.006 respectively). NSI to lactic acid was significantly higher on the face than on the hands (P < 0.05). Our findings indicate that subjects with the A transition at position ‐308 on the TNF‐α gene experience more intense NSI with common ingredients, i.e., lactic acid and water, than those with the wild type. TNF‐α polymorphism ‐308 may account for some of the inter‐individual variability in response to skin care practices.  相似文献

The elastohydrodynamic lubrication (EHL) problem of an axi-symmetric, elastomeric, reciprocating seal may be considered as one of a heavily loaded line contact with only small piezo-viscous effects. A method is presented to calculate the overall film thickness for the stationary, isothermal situation with sufficient lubricant supply and negligible surface roughness effects. It applies the inverse theory to the frictionless dry contact pressure distribution, considering the influence of the boosting action in the entry zone of the film. Its validity is shown for the isoviscous EHL of the Hertzian contact. The finite element method (FEM) may be used to calculate the contact situation of mounted and pressurized seals. Important aspects of such calculations are discussed. To calculate film thicknesses using such FEM results, a computer program PROGRES was written. Good correspondence between the predictions of PROGRES and experimental outstroke film thicknesses are obtained for a rectangular rod seal with rounded edges.  相似文献

Coagulase-negative staphylococci (CNS) are the most commonly isolated bacteria from goat milk, but they have often been identified with phenotypic methods, which may have resulted in misclassification. The aims of this paper were to assess the amount of misclassification of a phenotypic test for identifying CNS species from goat milk compared with transfer RNA intergenic spacer PCR (tDNA-PCR) followed by capillary electrophoresis, and to apply the tDNA-PCR technique on different capillary electrophoresis equipment. Milk samples were collected from 416 does in 5 Californian dairy goat herds on 3 occasions during lactation. In total, 219 CNS isolates were identified at the species level with tDNA-PCR and subjected to the API 20 Staph identification test kit (API Staph; bioMérieux, Durham, NC). If the same species was isolated multiple times from the same udder gland, only the first isolate was used for further analyses, resulting in 115 unique CNS isolates. According to the tDNA-PCR test, the most prevalent CNS species were Staphylococcus epidermidis, Staphylococcus caprae, and Staphylococcus simulans. Typeability with API staph was low (72%). Although the API Staph test was capable of identifying the majority of Staph. epidermidis and Staph. caprae isolates, sensitivity for identification of Staph. simulans was low. The true positive fraction was high for the 3 most prevalent species. It was concluded that the overall performance of API Staph in differentiating CNS species from goat milk was moderate to low, mainly because of the low typeability, and that genotypic methods such as tDNA-PCR are preferred.  相似文献

The initial stages of the electrocrystallization of nickel are studied by simultaneous ellipsometric and amperometric investigation during potentiostatic deposition. Theoretical equations for Δ and Ψ-time transients are derived for both conical and hemispherical growth forms. It is shown that these equations can adequately describe the experimental optical transient data. Moreover, the ellipsometric monitoring of the deposition process gives information regarding the topography of the deposit.  相似文献

The reaction mechanism of zinc and amalgamated zinc was investigated with the galvanostatic transient technique in the concentration range 1.5–10 M KOH. The Tafel slopes of the zinc electrode were 40 mV anodically and 120 mV cathodically. The cathodic reaction order in zincate was found to be +1. From the Tafel slopes and the dependence of the exchange current density on the activity of the KOH, the reaction orders in OH? were calculated, yielding values of 2.3± 0.8 in the anodic and ?0.8 ± 0.2 in the cathodic direction. These results are consistent with the suggested mechanism of Bockris et al.[16] for the zinc electrode. The Tafel slope in cathodic direction of the amalgamated zinc electrode was a function of the KOH concentration (120 mV at KOH concentrations up to 3 M; about 60 mV in 10 M KOH); the anodic Tafel slope was 30 mV over the whole concentration range. These results and measurements at constant ionic strength suggest a mechanism which involves the participation of water. The difference in behaviour of the zinc electrode and the amalgamated zinc electrode is probably caused by changes in the adsorption characteristics due to amalgamation.  相似文献

Using a short-term recognition memory task, the authors evaluated the carryover across trials of 2 types of auditory information: the characteristics of individual study sounds (item information) and the relationships between the study sounds (study set homogeneity). On each trial, subjects heard 2 successive broadband study sounds and then decided whether a subsequently presented probe sound had been in the study set. On some trials, the similarity of the probe item to stimuli presented on the preceding trial was manipulated. This item information interfered with recognition, and false alarms increased from 0.4% to 4.4%. Moreover, the interference was tuned so that only stimuli that were very similar to each other interfered. On other trials, the relationship among stimuli was manipulated to alter the criterion subjects used in making recognition judgments. The effect of this manipulation was confined to the trial on which the criterion change was generated and did not affect the subsequent trial. These results demonstrate the existence of a sharply tuned carryover of auditory item information but no carryover of the effects of study set homogeneity. The objectives of this study were (1) to determine the test characteristics and predictive values of cow-level milk somatic cell count (SCC) information from (multiple) test-day recordings before drying off to identify major-pathogen-infected cows at drying off; and (2) to explore to what extent (an estimate of) the herd prevalence of subclinical mastitis, milk yield level, and parity of the cows affects the estimates. In total, 550 cows from 15 commercial dairy herds with overall good udder health management were dried-off during a study period of 6 mo. Test-day SCC were available through milk recording and within 5 d before drying off cows were sampled for quarter-level bacteriological culturing serving as the gold standard. Sensitivity (Se), specificity (Sp), positive predictive value (PPV), and negative predictive value (NPV) were calculated at different threshold values of SCC, ranging between 50,000 and 500,000 cells/mL, to detect major-pathogen-infected cows. At a commonly used threshold of 200,000 cells/mL, the Se and Sp of (a combination of) test-day SCC before drying off ranged between 37.6 and 57.6% and between 66.7 and 79.3%, respectively. Still, estimates were modified by the herd level prevalence of subclinical mastitis and the cow's milk yield and parity. For instance, at the 200,000 cells/mL threshold using the geometric mean SCC of the 3 last test-days, the overall Se, Sp, PPV, and NPV were 37.6, 79.3, 30.8, and 83.9%, respectively, whereas these were 27.8, 87.5, 21.7, and 90.6%, respectively, for heifers and 40.3, 73.5, 33.3, and 78.9%, respectively, for multiparous cows. In conclusion, test-day SCC records obtained via milk recording are reliable to detect dairy cows at drying off that are not infected with major pathogens as determined by bacteriological culture and could eventually facilitate implementation of selective dry cow therapy in commercial dairy herds. Because estimates of the herd-level prevalence of subclinical mastitis, milk yield level, and parity of the cows affect the estimates of the test characteristics and predictive values to some extent, one should consider taking these parameters into account when differentiating infected from uninfected cows based on SCC data.  相似文献

An experimental trial was conducted to explore the effect of vaccination with a polyvalent vaccine against mastitis (Startvac) on the early immune response after experimental intramammary challenge with a heterologous killed Staphylococcus aureus strain. The effect of vaccination on milk production, clinical signs, quarter milk somatic cell count, milk polymorphonuclear neutrophilic leukocyte (PMN) concentration and viability, the concentration of antigen-specific antibodies [slime associated antigenic complex (SAAC) and J5] and their IgG1 and IgG2 subtypes in both serum and whey, and the antigen-specific IFN-γ, IL-4, and IL-17 production by blood lymphocytes after in vitro stimulation with S. aureus and Escherichia coli extracts were determined. A cohort of 8 clinically healthy end-term cows and heifers were conveniently selected, of which half was vaccinated with Startvac at 45 and 10 d before the expected calving date and half served as nonvaccinated control animals. At 15 d in milk, 2 contralateral quarters of each of the 8 animals were challenged with 2 × 109 cfu/mL of the formaldehyde-killed S. aureusC195strain. The 2 other quarters were infused with phosphate-buffered saline and served as control quarters. The increase in both quarter milk somatic cell count and PMN concentration and the drop in milk production after S. aureus inoculation was less pronounced in the vaccinates than in the nonvaccinates, reflecting a less severe inflammatory response. No significant differences in PMN viability between vaccinates and nonvaccinates could be demonstrated. The serum SAAC- and J5-specific antibody concentration significantly increased across the dry period in the vaccinated animals only. The whey concentration of SAAC-specific antibodies was significantly higher in vaccinates than in nonvaccinates at both 15 and 17 d in milk, independent from the challenge status of the quarters. No significant differences in the whey J5-specific antibody concentration were observed. Vaccination with Startvac seems to primarily evoke a Th2 response for S. aureus characterized by a shift toward the IgG1 antibody subtype and accompanied by a less pronounced Th1 response. The type of response against E. coli was less clear, though a weak but significant shift toward the IgG2 antibody subtype after vaccination and high IFN-γ levels after in vitro stimulation suggest a Th1 response. The increased SAAC-specific antibody concentration in whey in vaccinates compared with nonvaccinates most probably triggers the opsonization of the inoculated S. aureus bacteria, resulting in a more efficient elimination of the bacteria from the mammary gland.  相似文献

The proportion of cows in the UK dairy herd whose sires were misidentified was estimated using DNA markers. Genetic marker genotypes were determined on 568 cows (from 168 milk samples and 400 hair samples) and 96 putative sires (from semen samples). The estimated pedigree error rate from the hair samples was 8.8%, and from the milk samples, 13.1%, giving an overall estimate of the error rate of 10%. This level of pedigree errors will have a relatively large impact on the efficiency of progeny testing and the accuracy of cow predicted breeding values. We predict a loss of response to selection of approximately 2 to 3% given this error rate.  相似文献

The purposes of this study were 1) to investigate the heritability, reliability, and selection response for survival traits following a Weibull frailty proportional hazard model; and 2) to examine the relationship between genetic parameters from a Weibull model, a discrete proportional hazard model, and a binary data analysis using a linear model. Both analytical methods and Monte Carlo simulations were used to achieve these aims. Data were simulated using the Weibull frailty model with two different shapes of the Weibull distribution. Breeding values of 100 unrelated sires with 50 to 100 progeny (with different levels of censoring) were generated from a normal distribution and two different sire variances. For analysis of longevity data on the discrete scale, simulated data were transformed to a discrete scale using arbitrary ends of discrete intervals of 400, 800, or 1200 d. For binary data analysis, an individual's longevity was either 0 (when longevity was less than the end of interval) or 1 (when longevity was equal or greater than the end of interval). Three different statistical models were investigated in this study: a Weibull model, a discrete-time model (a proportional hazard model assuming that the survival data are measured on a discrete scale with few classes), and a linear model based upon binary data. An alternative derivation using basic expressions of reliabilities in sire models suggests a simple equation for the heritability on the original scale (effective heritability) that is not dependent on the Weibull parameters. The predictions of reliabilities using the proposed formulae in this study are in very good agreement with reliabilities observed from simulations. In general, the estimates of reliability from either the discrete model or the binary data analysis were close to estimates from the Weibull model for a given number of uncensored records in this simplified case of a balanced design. Although selection response from the binary data analysis depends on the end of interval point, there is a relatively good agreement between selection responses in the Weibull model and the binary data analysis. In general, when the underlying survival data is from a Weibull distribution, it appears that the method of analyzing data does not greatly affect the results in terms of sire ranking or response to selection, at least for the simplified context considered in this study.  相似文献
