Interventions which make serum lipoproteins less susceptible to oxidation may be antiatherogenic. The antioxidant properties of garlic which have been demonstrated in vitro led us to investigate the effects of garlic supplements on lipoprotein oxidation susceptibility in humans. Ten healthy volunteers were given 600 mg/d of garlic powder (6 tablets of Kwai) for two weeks in a placebo-controlled, randomized, double-blind crossover trial. We found that although serum lipid and lipoprotein levels were not lowered in this short time period, the ex vivo susceptibility of apolipoprotein B-containing lipoproteins to oxidation was significantly decreased (-34%). Because garlic has been reported to beneficially affect serum lipid levels, platelet function, fibrinolysis and blood pressure, this additional effect of retarding lipoprotein oxidation may contribute to the potential antiatherosclerotic effect of garlic.  相似文献

We studied the effect of acute sustained hypoxia on ventilation (VE) and oxygen consumption (VO2) over one hour during quiet wakefulness in young (6 days) and older (6 weeks) piglets in thermoneutral conditions during baseline, moderate hypoxia (PaO2 approximately 45 mmHg), and severe hypoxia (PaO2 approximately 30 mmHg). During severe hypoxia, ventilation and pH increased while PaCO2 decreased in both age groups. Blood gas changes (decreases PACO2, increases pH), but not ventilatory changes, were greater in the older piglets (P < 0.05). VO2 decreased similarly (-30%) while VE/VO2 rose over 160% in both age groups. During moderate hypoxia, changes in blood gas, VE, and VO2 were in a similar direction, but smaller in magnitude. We conclude that: (1) changes in blood gases and VO2 are amplified by maturation and severity of hypoxia and (2) blood gas changes are greater in older vs young piglets despite similar ventilatory responses suggesting maturational differences in CO2 production or dead space ventilation.  相似文献

Vascular tones is an essential component in maintaining steady regional blood flow and dynamic responsiveness of a vascular bed. Sympathetic innervation can contribute to vascular tone. Although certain studies have reported evoked changes in cochlear blood flow (CBF) with activation of the sympathetic fibers to the cochlear vasculature, other studies have failed to show evidence of sympathetic contribution to CBF regulation when the cervical sympathetic fibers were unilaterally sectioned. We hypothesized that the bilateral 'sympathectomy of the stellate ganglia' would remove sufficient sympathetic input to the cochlea to yield a change in CBF resting level. To test this hypothesis a new technique was used to expose the stellate ganglia (SG) bilaterally and induce a chemical sympathectomy. We observed that unilateral SG blockade with 2 microliters of 4 mM lidocaine hydrochloride on either side produced a 5-10% increase in CBF, which recovered to baseline during the following 2 min. A subsequent blockade of the contralateral SG produced a rapid 25-35% increase, which then recovered partially during the following 3-4 min, remaining 5-15% above the baseline over a 20 min measurement period. Superior cervical ganglion transection did not affect CBF. Our results provide evidence for the existence of a tonic sympathetic component in the control of vascular tone in guinea pig cochlea. This neural effect is derived bilaterally from SG. This result is consistent with previous anatomical studies showing the bilateral innervation of the cochlea by the SG sympathetic fibers and with previous physiological studies on the bilaterality of evoked changes in CBF due to electric stimulation of SG.  相似文献

A new generation of wavelength stabilised, uncooled 980 nm pump modules in MiniDIL housings is presented, enabling 400 mW ex-fibre power over a temperature range of 10/spl deg/C to 70/spl deg/C. At 100/spl deg/C 200 mW power is still obtained with a robust fibre coupling scheme.  相似文献

Structured diagnostic interviews were used to determine DSM-III-R axis I and II diagnoses among 136 female psychiatric inpatients. To distinguish comorbidity of eating disorders with axis I and II disorders from simple diagnostic overlap, the frequency and distribution of diagnoses among the 31 patients with an eating disorder and the 105 without an eating disorder were compared. Social phobia, substance use disorders, borderline personality disorder, and avoidant personality disorder were diagnosed in a significantly larger proportion of the group with eating disorders. Future studies should focus on interpreting the meaning of the co-occurrence of these disorders in patients with eating disorders.  相似文献

INTRODUCTION: A factory producing lead ingots, located in Ca?apava, caused lead and cadmium contamination of the environment, in the Paraiba Valley region of Southeastern, Brazil, through the discharge of industrial waste and the recycling of batteries. The factory, set in a rural, dairy cattle breeding area, worried sanitary authorities who envisaged the possibility of these metals' having entered the food chain. For the purpose of assessing the levels of contamination of the milk produced in the region, due to the cattle's possible consumption of contaminated grass and water, the amounts of cadmium and lead present in the milk were verified. MATERIAL AND METHOD: Major producers, covering an area of up to 20 km from the contaminated source, authorized collection of 218 samples of both pasteurized and non-pasteurized milk, which were analysed. Lead and cadmium levels were determined by flame atomic absorption spectrophotometry, the lead being pre-concentrated by complexation with APDC (ammonium 1-pyrrolidinecarbodithioate) and further extraction with isobutyl methylketone. RESULTS AND CONCLUSIONS: Of the total number of samples, 43 presented lead levels over the maximum limit of 0.05 mg/kg established by Brazilian legislation. The median value found for lead was 0.04 mg/L. The variance analysis, with 95% confidence level, found no significant difference among the types of milk studied with regard to lead levels. As for cadmium, all samples showed levels below the 0.02 mg/L quantification limit of the method. In spite of the environmental contamination, the levels of cadmium found in the milk were below the 1.0 mg/kg limit established by Brazilian legislation.  相似文献

To identify new loci that are involved in the assembly and targeting of dynein complexes, we have screened a collection of motility mutants that were generated by insertional mutagenesis. One such mutant, 5B10, lacks the inner arm isoform known as the I1 complex. This isoform is located proximal to the first radial spoke in each 96-nm axoneme repeat and is an important target for the regulation of flagellar motility. Complementation tests reveal that 5B10 represents a new I1 locus, IDA7. Biochemical analyses confirm that ida7 axonemes lack at least five I1 complex subunits. Southern blots probed with a clone containing the gene encoding the 140-kDa intermediate chain (IC) indicate that the ida7 mutation is the result of plasmid insertion into the IC140 gene. Transformation with a wild-type copy of the IC140 gene completely rescues the mutant defects. Surprisingly, transformation with a construct of the IC140 gene lacking the first four exons of the coding sequence also rescues the mutant phenotype. These studies indicate that IC140 is essential for assembly of the I1 complex, but unlike other dynein ICs, the N-terminal region is not critical for its activity.  相似文献

Previous studies have suggested that human follicular fluid contains factors that reduce the zona-binding capacity of spermatozoa. The present study provides further evidence of the existence of such factors. Using the hemizona binding assay (HZA), we have shown that the inhibitory effect of human follicular fluid on the zona-binding capacity of spermatozoa is concentration-dependent, an inhibitory effect being detected when the concentration of human follicular fluid was > or = 10%. A 1% concentration of human follicular fluid did not possess this inhibitory activity. Heating human follicular fluid at 56 degrees C for 30 min did not affect its inhibitory properties; treatment with proteinase-K abolished such inhibition. Human follicular fluid was fractionated sequentially by concanavalin-A affinity chromatography, Mono Q ion-exchange chromatography and Superose-12 gel filtration. The zona binding inhibitory activity resided in the fraction which bound to the lectin and Mono Q column and contained molecules with native molecular weights of 32 and 192 kDa. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis analysis suggested that the 192 kDa glycoprotein was a tetramer, while the 32 kDa glycoprotein remained as a single molecular species under denaturing conditions. We conclude that two glycoproteins were responsible for the zona binding inhibitory activity of human follicular fluid. The physiological role of these factors remains unclear.  相似文献

Five monoclonal antibodies (mAbs) that recognize human glutamate dehydrogenase (GDH) have been selected and designated as monoclonal antibodies hGDH60-6, hGDH60-8, hGDH63-10, hGDH63-11, and hGDH91-14. A total of five mAbs recognizing different epitopes of the enzyme were obtained, two of which inhibited human GDH activity. When total proteins of human homogenate separated by SDS- PAGE, were probed with mAbs, a single reactive protein band of 55 kDa, which co-migrated with purified recombinant human GDH was detected. When the purified GDH was incubated with each of the mAbs, its enzyme activity was inhibited by up to 58%. Epitope mapping analysis identified, two subgroups of mAbs recognizing different peptide fragments. Using the individual anti-GDH antibodies as probes, the cross reactivities of brain GDH obtained from human and other animal brain tissues were investigated. For the human and animal tissues tested, immunoreactive bands on Western blots appeared to have the same molecular mass of 55 kDa when hGHD60-6, hGHD60-8, or hGHD91-14 mAbs were used as probes. However, the anti-human GDH mAbs immunoreactive to bands on Western blots reacted differently on the immunoblots of the other animal brains tested, i.e., the two monoclonal antibodies hGDH63-10 and hGDH63-11 only produced positive results for human. These results suggest that human brain GDH is immunologically distinct from those of other mammalian brains. Thorough characterization of these anti-human GDH mAbs could provide potentially valuable tool as immunodiagnostic reagents for the detection, identification and characterization of the various neurological diseases related to the GDH enzyme.  相似文献
