Gene therapy research has advanced to clinical trials, but it is hampered by unstable nucleic acids packaged inside carriers and there is a lack of specificity towards targeted sites in the body. This study aims to address gene therapy limitations by encapsidating a plasmid synthesizing a short hairpin RNA (shRNA) that targets the anti-apoptotic Bcl-2 gene using truncated hepatitis B core antigen (tHBcAg) virus-like particle (VLP). A shRNA sequence targeting anti-apoptotic Bcl-2 was synthesized and cloned into the pSilencer 2.0-U6 vector. The recombinant plasmid, namely PshRNA, was encapsidated inside tHBcAg VLP and conjugated with folic acid (FA) to produce FA-tHBcAg-PshRNA VLP. Electron microscopy revealed that the FA-tHBcAg-PshRNA VLP has an icosahedral structure that is similar to the unmodified tHBcAg VLP. Delivery of FA-tHBcAg-PshRNA VLP into HeLa cells overexpressing the folate receptor significantly downregulated the expression of anti-apoptotic Bcl-2 at 48 and 72 h post-transfection. The 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay demonstrated that the cells’ viability was significantly reduced from 89.46% at 24 h to 64.52% and 60.63%, respectively, at 48 and 72 h post-transfection. As a conclusion, tHBcAg VLP can be used as a carrier for a receptor-mediated targeted delivery of a therapeutic plasmid encoding shRNA for gene silencing in cancer cells.  相似文献

Cellulose nanofibers (CNF) isolated from plant biomass have attracted considerable interests in polymer engineering. The limitations associated with CNF-based nanocomposites are often linked to the time-consuming preparation methods and lack of desired surface functionalities. Herein, we demonstrate the feasibility of preparing a multifunctional CNF-zinc oxide (CNF-ZnO) nanocomposite with dual antibacterial and reinforcing properties via a facile and efficient ultrasound route. We characterized and examined the antibacterial and mechanical reinforcement performances of our ultrasonically induced nanocomposite. Based on our electron microscopy analyses, the ZnO deposited onto the nanofibrous network had a flake-like morphology with particle sizes ranging between 21 to 34 nm. pH levels between 8–10 led to the formation of ultrafine ZnO particles with a uniform size distribution. The resultant CNF-ZnO composite showed improved thermal stability compared to pure CNF. The composite showed potent inhibitory activities against Gram-positive (methicillin-resistant Staphylococcus aureus (MRSA)) and Gram-negative Salmonella typhi (S. typhi) bacteria. A CNF-ZnO-reinforced natural rubber (NR/CNF-ZnO) composite film, which was produced via latex mixing and casting methods, exhibited up to 42% improvement in tensile strength compared with the neat NR. The findings of this study suggest that ultrasonically-synthesized palm CNF-ZnO nanocomposites could find potential applications in the biomedical field and in the development of high strength rubber composites.  相似文献

Roselle seed oil (RSO) is an underutilised seed oil, which is having beneficial properties to humans. Microencapsulation of vegetables and seed oil is an alternative to preserve these properties and deliver them to the human diet. In this study, microencapsulation of RSO was performed using co-extrusion technology and 1.5% w/w sodium alginate solution with 1.5% w/w high methoxyl pectin as the wall materials. Results showed that the microencapsulation efficiency was high (95.68 ± 1.95%) and the microencapsulation indeed preserved the content of unsaturated fatty acids, phytosterols, and tocopherols. During an accelerated storage condition at 65 °C for 24 days, most of these properties remained high for the first six storage days, including tocopherol which preserved high γ-tocopherol. Worth noting that the microencapsulated RSO (MRSO) was particularly effective in preserving the total unsaturated fatty acids (especially C18:1 and C18:2), even during the 24th day of storage. The total unsaturated fatty acids retained by MRSO were significantly (P < 0.05) higher (70 ± 0.4%) than the RSO (65.6 ± 1.6%). In short, microencapsulation was effective in preserving the RSO properties.  相似文献

OBJECTIVE: To test the usefulness of a commercial DNA hybridization assay for the detection of high-risk (HR) human papillomavirus (HPV) types in archival cervical smears and to compare the sensitivity with that of polymerase chain reaction (PCR) using consensus primers. STUDY DESIGN: Stained material was scraped from archival slides and the pellet volume noted. DNA was extracted using silica/guanidinium isothiocyanate and the quality checked by amplification of the beta-globin gene. HR-HPV DNA was detected using a commercial hybrid capture assay (HCA) and the results compared with an in-house amplification system with consensus primers. RESULTS: Of 156 archival smears stored for 12-13 years, 20 were positive by HCA using an HR probe cocktail. Ninety-eight were also tested by PCR, and 35 were positive. The percentage of HPV-positive samples increased with the increasing size of the pellet. HR-HCA detected more positives in samples with high grade squamous intraepithelial lesion (moderate/severe dyskaryosis). CONCLUSION: Both hybridization by HCA and amplification by PCR could be used to detect genital HPV in archival smears. The general primers PCR detected more positives than HR-HCA but included HPV 6/11. While variation in sample size and prolonged storage may reduce the quality of DNA, the use of archival material for longitudinal studies of HPV presence is potentially worthwhile.  相似文献

The problem of finding the shape of a perfectly conducting cylinder when illuminated by an E-polarized incident electromagnetic wave is considered. The given problem is cast in the form of a penalized optimization problem in which we seek a surface in some suitably constrained class which minimizes a functional involving two terms. The first term measures the mean square deviation of the far field of the solution to the direct scattering problem for a surface in the admissible class from given, measured, far field data whilst the second term, penalty, measures the error in satisfying the imposed boundary conditions. An optimisation algorithm is used which does not require that a direct problem be solved in the updating process. Explicit reconstructions are presented which illustrate the effectiveness of the method. Some results indicating the sensitivity of the algorithm to the choice of penalty parameter and the number of incident waves are also given.  相似文献

The authors demonstrate the use of radiography in the investigation of an historic painting and describe the potential benefits of computed radiography compared with conventional screen-film radiography. The subject for the comparison was a 16 x 19-foot oil-on-canvas painting, Scipio Africanus Freeing Massiva, by Giovanni Battista Tiepolo. Radiographs of the painting were obtained by using a portable, industrial radiographic unit and both conventional screen-film and photostimulable phosphor plate cassettes. For this investigation, computed radiography had a number of advantages over screen-film radiography, largely due to its wider dynamic range and its capabilities for enhancing the digital images with image processing tools such as magnification, edge enhancement, colorization, and airbrushing. The ability to electronically combine images from the large painting into a single composite image file was extremely valuable, as this technique was much less cumbersome and resulted in much higher quality composite images than could be achieved with conventional radiography. An additional advantage of computed radiography includes the capability to easily archive and transmit these images in a digital format for subsequent review.  相似文献

Existing IP routing algorithms have been developed mainly for non‐VLAN networks. For these routing algorithms to work, an end station must be attached to its associated network. The introduction of VLAN has, however, allowed the physical location of the end station to be changed from time to time. Therefore, some extra mechanism is required to support routing in VLAN environment. Existing approaches have some drawbacks in terms of inefficiency in traffic forwarding and switch software complexity. In this paper, a simple and efficient method for routing over IP subnet VLAN in Ethernet is proposed. This method has been implemented in hardware and found to work correctly.
