Chronically implanted ultrasonic Doppler flowmeters were used to obtain simultaneously recorded flow velocity signals from internal carotid and vertebral arteries, and the sagittal sinus, in rabbits. All three signals increased to 144 +/- 7-215 +/- 35% of baseline during hypercapnia (arterial Pco2 55 mmHg) in both anesthetized and conscious animals. During the period of change in inspired CO2, the relationship between simultaneously recorded mean internal carotid and mean sagittal sinus signals was linear, with the correlation ranging from 0.83 to 0.96. Since forebrain arterial inflow must approximate forebrain venous outflow, the high correlation between internal carotid and sagittal sinus signals indicates that these measures provide reliable and valid indices of cerebral blood flow (CBF). Vertebral and internal carotid angiography confirmed the location of Doppler probe. Chronically implanted ultrasonic Doppler flowmeters can, thus, provide continuous noninvasive measurements of cerebral arterial flow in both anesthetized and conscious rabbits. 相似文献

Fluorescence detection of fluorogen-labeled neurotransmitters is demonstrated using 100 fs pulses from a titanium-sapphire mode-locked laser to achieve molecular excitation by simultaneous absorption of two and three photons of near-IR radiation. Two-photon excitation spectra are determined for the naphthalene-2,3 dicarboxaldehyde derivative of glycine and the fluorescamine derivative of leucine enkephalin, with the peak excitation cross section (o2) approximately equal to 1 x 10(-50) cm4 s/photon for both species. Three-photon-excitation fluorescence is demonstrated for o-phthaldialdehyde-labeled glutamate using excitation wavelengths between 965 and 1012 nm. The three-photon excitation cross section (o3) remains nearly constant in this wavelength range, with an absolute value of approximately 10(-84)-10(-85) cm6 s2/photon 2. Rapid cycling of analytes through the fluorescent excited state and detection that is free from background caused by Rayleigh and Raman scatter combine to make multiphoton-excited fluorescence a highly sensitive approach for detecting trace amounts of neurotransmitters. Measurements of two-photon-excited fluorescence of fluorescamine-labeled bradykinin and analysis of multiphoton-excited background reveal the potential of this method to detect fewer than 1000 neurotransmitter molecules. 相似文献

The fabrication of esthetic custom provisional restorations that provide optimum esthetics and function accomplished with minimal chairside time is described. The effects of mamelons, translucency, and incisal haloes are reproduced to mimic natural dentition. The technique is based on a provisional shell that is made in the laboratory and relined intraorally for a more precise fit. In addition, it can be characterized chairside to better satisfy the esthetic demands of the patient. The recombinant oncotoxin OLX-209 [e23(Fv)PE38KDEL] has been developed to target cancers with erbB-2 expression and is nearing a clinical trial. Important in clinical planning is the selection of patients on the basis of tumor expression of erbB-2. ErbB-2 gene amplification occurs in cancers of the breast, stomach, and ovary. Patients with these diseases and evident overexpression are candidates for OLX-209 therapy. In lung cancer, overexpression of erbB-2 is also frequent, but in most cases, it is not caused by gene amplification. This study demonstrates that OLX-209 has activity on lung cancer cells with varying levels of erbB-2 expression in the presence and absence of gene amplification. In vitro sensitivity of cell lines to OLX-209 is related to erbB-2 expression level. Normal bronchial epithelial cells were not sensitive. Effective treatment of lung cancer cell lines growing as xenografts in nude mice was shown with Calu-3 (a lung adenocarcinoma line with high levels of p185(erbB-2) caused by gene amplification) and three other lung adenocarcinomas (A549, NCI-H1466, and 201T) with lower levels of p185(erbB-2) and no gene amplification. The 201T cell line was isolated recently from a lung tumor with erbB-2 expression in the original tumor. The results of this study indicate that patients with erbB-2-positive, non-small cell lung cancer should be included in clinical trials of OLX-209. 相似文献
